Physiological stress in laying hens.

Stress responses in laying hens were mediated by continuous infusion of adrenocorticotropin (ACTH) via osmotic pumps. The ACTH was dissolved in saline solution (0.85%), and each pump delivered 8 IU of ACTH per kilogram of BW per day at the rate of 1 microL/h for 7 d. Control hens received pumps loaded with saline. Measurements were made at 6 d postpump implantation, unless otherwise indicated. The ACTH-treatment increased BW and total carcass, rear half of carcass, intestinal, and liver weights. Proximate analyses of liver showed increases in dry weight, moisture, protein, fat, carbohydrate, and ash content. Weights of the front half of the carcass, as well as weights of the abdominal fat pad, heart, head, feet, and skin were unaffected by ACTH-treatment. Plasma corticosterone, glucose, cholesterol, and high-density lipoproteins were increased by ACTH, whereas triglycerides were decreased. Feed and water intake, total excreta, and excretory DM were all increased in ACTH-treated hens. The ACTH decreased carbohydrate in excreta, whereas ash, protein, fiber, and gross energy of excreta were unaffected. The ACTH did not affect digestibility of dry matter, proteins, carbohydrates, fats, or gross energy; however, absorption of ash, protein, carbohydrates, and gross energy were increased by ACTH. Antibody levels to sheep red blood cells, cell-mediated immunity (wattle index to phytohemagglutinin-phosphate), and relative spleen weight were reduced by ACTH, whereas heterophil:lymphocyte ratio was increased. Reproduction in hens was negatively affected by ACTH treatment, as measured by cessation of laying on the third day of treatment, atretic follicles, and decreased oviduct weight.

Control of induced infestations of adult Amblyomma hebraeum with sustained release ivermectin.

The efficacy of ivermectin, administered in a sustained release formulation by intraruminal pumps at approximate daily dose rates of 20, 40 and 60 micrograms/kg, was evaluated in 16 cattle against induced infestations of 3 strains of adult Amblyomma hebraeum. Engorged female ticks were mass-measured and incubated, and reproductive data recorded. There was an increase in mortality of male and female ticks compared to that of controls with increasing daily dose of ivermectin, and a decrease in the number of ticks engorging. Ticks fed on ivermectin-treated cattle had a smaller mass when engorged and laid smaller egg masses, both absolutely and as a proportion of engorged mass. Index of reproduction was reduced 100% at 60 micrograms/kg/day, greater than 99% at 40 micrograms/kg/day and 96% at 20 micrograms/kg/day. Differences occurred between the 3 strains of A. hebraeum used in the study, especially with regard to engorged mass and reproductive variables. Practical implications of the application of sustained release ivermectin for the control of A. hebraeum, specifically with reference to heartwater (Cowdria ruminantium), are discussed.

Changes in physiological, EEG and psychological parameters in women during the spontaneous menstrual cycle and following oral contraceptives.

In a controlled cross-over design study performed with 14 female subjects, serum hormone levels, the EEG and a number of performance tests were recorded during spontaneous and oral contraceptive-controlled menstrual cycles. The mean alpha-frequency showed cyclic changes, i.e. slower alpha-waves during the follicular phase and faster alpha-waves during the luteal phase. Smaller cycle stage-dependent differences in the power of the theta- and beta-bands also were noted. An increase in several performance task scores was noted during the periovulatory period, whereas lowest performance scores were recorded during the late luteal and early menstrual phases. No such effects were observed in the same subjects while they were taking oral contraceptives. These results demonstrate that the gross electrical activity of the brain changes in a parallel with changed hormone levels. The changes in performance tests coincide with increasing or decreasing alpha activities in the EEG. The common underlying mechanism may be an activation of central nervous system monoaminergic pathways which are known to be involved in steroid feedback.

Timing of postpartum tubal sterilization using the Filshie clips: an analysis of data from two developing-country centers.

Two-hundred-ninety-seven women at a Panamanian center and 148 women at a Philippine center had their tubal sterilization performed during their postpartum hospital stay after an uncomplicated vaginal delivery of a live birth. The sterilizations were all performed with the use of the Filshie clip via minilaparotomy. The timing of the sterilization varied from two hours to six days after delivery. At each of the two centers, women sterilized within 48 hours after delivery were compared with those sterilized at 49 or more hours with respect to surgical difficulties, tubal injuries, complications/complaints, technical failures and lengths of hospitalization after sterilization and before discharge. The one-year gross cumulative pregnancy rates were also compared. No significant differences were detected in any of the above outcome variables between the two timing groups at each center. While the data suggest that tubal sterilizations by the Filshie clip performed two to six days following childbirth are as safe and effective as those performed within 48 hours of delivery, further studies are urged.

Growth and Specific Nodule Activity of Soybean during Application and Recovery of a Leaf Moisture Stress.

Soybean plants growing at day/night temperatures of 30/18, 26/18, and 22/18 C were subjected to a single drying and recovery cycle during an 18- to 19-day period in the early to midpod development stage. Leaf total electrochemical water potential was reduced to about -24 bars during the 4-day drying cycle at all temperatures, but recovered to control levels upon rewatering. The changes in dry matter accumulation in whole plants and plant parts, specific activity of nodules as measured by acetylene reduction, and levels of adenosine phosphates in nodules were measured periodically during stress and recovery.Vegetative and reproductive growth were about equally suppressed by the leaf moisture stress. Both rate of appearance and number of pods were reduced. However, a similar average weight per pod for both stressed and control plants at the conclusion of the recovery period suggests that individual pod development is not irreversibly affected by a single stress cycle and that yield potential is restricted by a decrease in number of pods or seed. Dry matter accumulation in plants and pods was unaffected by temperature.Specific nodule activity and energy charge of nodules declined concurrently with leaf moisture potential. Recovery of specific nodule activity following rewatering lagged behind recovery of leaf moisture potential, but energy charge of nodules recovered as rapidly as leaf moisture potential upon rewatering. Thus, the delayed recovery of specific nodule activity does not appear to be related to recovery of energy charge of the nodules.

A comparative study of 35 mcg and 50 mcg combined oral contraceptives: results from a multicenter clinical trial.

A comparative multicenter clinical trial of two combined oral contraceptives (OCs) differing only in the estrogen content (35 mcg ethinyl estradiol versus 50 mcg mestranol) was conducted at five clinics located in Yugoslavia, Egypt, Sri Lanka, Costa Rica and Mexico. The trial was designed to determine the differences between Norinyl 1+35 (Syntex) and Norinyl 1+50 (Syntex) in rates and reasons of discontinuation, and frequency of selected side effects which might contribute to method discontinuation. This report includes analysis of 1698 women, all of whom were interval patients (at least 42 days but within 26 weeks postpartum), randomly allocated to one of the above OCs between October 1982 and January 1984. Follow-up visits were scheduled at 1, 4, 8 and 12 months after admission. Significantly more women in the Norinyl 1+35 group (p less than .001) reported intermenstrual bleeding (primarily staining and spotting), as well as an increase in the occurrence of intermenstrual bleeding compared to women in the Norinyl 1+50 group. There were no significant differences between the groups for side effects with the exception of more women in the Norinyl 1+50 group (p less than .05) reporting breast discomfort. The lost to follow-up rate at 12 months was 19.3% for both the Norinyl 1+35 and the Norinyl 1+50 groups. The total discontinuation rate (including women lost to follow-up) at 12 months was 43.5% for the Norinyl 1+35 group and 41.0% for the Norinyl 1+50 group. There were no significant differences between the two groups for gross cumulative life table discontinuation rates (p greater than .05). There were six accidental pregnancies attributed to user failure reported during the study period; four in the Norinyl 1+35 group and two in the Norinyl 1+50 group.

Immediate postplacental insertion of GYNE-T 380 and GYNE-T 380 postpartum intrauterine contraceptive devices: randomized study.

OBJECTIVE: The purpose of the investigation was to compare the 1-year expulsion and efficacy rates of the GYNE-T 380 and the GYNE-T 380 Postpartum intrauterine contraceptive devices when inserted within 10 minutes after expulsion of the placenta in a term pregnancy. The two intrauterine contraceptive devices were identical, except that one was inserted by means of a temporary fundal suspension system and the other was placed directly into the uterine cavity. STUDY DESIGN: The study was a multicenter, randomized trial of intrauterine contraceptive devices in which 300 subjects accepted the GYNE-T 380 IUD and 292 subjects the GYNE-T 380 Postpartum IUD in clinics with adequate follow-up. RESULTS: At 1 year the gross cumulative expulsion rate was 13.2 per 100 cases (39 expulsions) with the GYNE-T 380 intrauterine contraceptive device and 16.2 per 100 cases (46 expulsions) with the GYNE-T 380 Postpartum device. There was no significant difference in the rate of expulsion between the two devices at any time during the year. There was one first-year intrauterine pregnancy, which occurred in a subject using the GYNE-T 380 device. The continuation rate for each device was above 80 per 100. CONCLUSION: The results indicate that both the GYNE-T 380 Postpartum and the standard GYNE-T 380 intrauterine contraceptive devices are safe and effective when inserted immediately after delivery of the placenta.

The levonorgestrel intrauterine system: more than a contraceptive.

Levonorgestrel can be released directly into the uterine cavity, where it causes pronounced endometrial suppression, although the dose of hormone is so low that the effect on ovarian function is negligible. The levonorgestrel-releasing intrauterine system (LNG-IUS) consists of a plain Nova-T device with a silastic reservoir attached to the vertical arm. The silastic reservoir is impregnated with levonorgestrel and is covered with a rate-limiting silastic membrane. The release rate oflevonorgestrel is approximately 20 microg/24 h for at least 5 years. The contraceptive efficacy of the LNG-IUS has been studied in a randomized, comparative trial over 5 years. A total of 1,821 women were fitted with the LNG-IUS and were compared with 937 women who were using the copper-releasing device, Nova-T. The continuation rates were 46.9 for the LNG-IUS and 44.5 for the Nova-T. The cumulative gross pregnancy rates were 0.5 for the LNG-IUS and 5.9 for the Nova-T. The Pearl index after 5 years was 0.09/100 woman-years for the LNG-IUS and the ectopic pregnancy rate was 0.02/100 woman-years. There were less withdrawals because of bleeding problems and pelvic inflammatory disease in the LNG-IUS group compared with the Nova-T group, but there were more withdrawals because of hormonal side-effects and absence of bleeding. There were no differences in the return of fertility after removal of the LNG-IUS and the Nova-T. All women will notice a change in their bleeding pattern after the LNG-IUS has been inserted and some will initially experience many days of spotting. It is extremely important to counsel women about the changes in bleeding pattern that will occur with the LNG-IUS before the system is fitted. One of the advantages of the LNG-IUS is that menstrual blood loss will decrease (and therefore hemoglobin levels will increase), which means that the LNG-IUS is one of the most effective reversible long-term treatments of idiopathic menorrhagia. The LNG-IUS can be used successfully throughout the reproductive period for effective contraception and treatment of menorrhagia. In addition, this system provides endometrial protection. Women who suffer from climacteric symptoms while they are using the LNG-IUS can be given estrogen to relieve their symptoms.

Chronic toxicity of thiabendazole (TBZ) in CD-1 mice.

Male and female CD-1 mice (50 mice per group) were administered thiabendazole (TBZ) in diet at levels of 0 (control), 0.031, 0.125 and 0.5% for 78 weeks. A life time study was terminated after 78 weeks due to enhanced strain specific mortality. There were no significant differences in mortality between the control and treated groups. Mean body weights of high-dose groups showed significant decreases compared with the controls. The bladder weights of male and female mice of the 0.5% group were significantly higher than those of the control mice. Gross findings in treated mice included the renal atrophy, hydronephrosis, calculi in renal pelvis and/or bladder and ovarian atrophy. Microscopic findings in the kidneys of treated mice included the nephrosis, hydronephrosis or hyperplasia of transitional epithelium of renal pelvis or papilla. In the bladder of treated mice, hyperplasia or squamous metaplasia of transitional epithelium and one transitional cell papilloma were observed. Dose-dependent decreases in the incidence of spontaneous lesion in the male or female reproductive system were recognized. It is concluded that TBZ is not carcinogenic to CD-1 mice of both sexes. However, caution should be exercised in the long-term application of high TBZ doses.

Swimming speeds and buoyancy compensation of migrating adult chum salmon Oncorhynchus keta revealed by speed/depth/acceleration data logger.

Although the homing migration of Pacific salmon is well documented, the swimming behaviour of the returning salmon has been poorly described, principally as a result of the difficulties encountered in monitoring salmon behaviour in the sea. The present study describes the use of a recently developed electronic data logger to obtain simultaneous recordings of the swimming speed, depth, fin-beating activity and body angle of free-ranging chum salmon Oncorhynchus keta during their homing migration in coastal waters. Chum salmon migrated horizontally at speeds of 1.5-3.0 km h(-1). The gross horizontal distance salmon moved during total recording periods were 1.24- to 19.0-fold greater than the net distance from the release site to the retrieval points. It is suggested that homing salmon did not drift passively but swam actively to the spawning grounds. Salmon preferred the surface water, but also made frequent vertical migrations. The travelled depth of each salmon ranged from 0.36 to 0.64 km per hour. Salmon descended at faster rates and steeper angles than they ascended. Both tailbeat frequency and tail thrust were higher during the ascent than the descent phase. These results suggest that chum salmon spent more energy during the ascent than the descent phase. Profiles of descent rate assumed an arched shape with respect to a change in hydrostatic pressure, while ascent rate increased with decreasing depth. High tailbeat frequencies were found during the course of ascent, which suggests that the salmon did not regulate the volume of air in the swim bladder during short-term vertical migrations.

Compromised development of calves (Bos gaurus) derived from in vitro-generated embryos and transferred interspecifically into domestic cattle (Bos taurus).

Advanced reproductive technologies, incuding IVF and interspecies embryo transfer, are becoming increasingly important for the preservation of endangered species. Previous attempts at interspecies transfers between Bos gaurus and Bos taurus have yielded compromised offspring. The goal of this investigation was to characterize the effects of interspecies transfer of IVF-derived embryos on subsequent neonatal outcome. To achieve this goal, fresh Bos gaurus IVF-derived embryos were transferred into Holstein (Bos taurus) recipients. Four fetuses were carried to term. Calf weight, temperature, heart rate, and respiration rate were recorded after birth. Blood samples also were obtained for determination of blood glucose, pH, packed cell volume (PCV), total hemoglobin (tHB), PO2, and PCO2. After parturition, milk production and health status of the recipients were recorded. Two calves were alive at birth, and two calves were stillborn. One of the calves that was born alive died within minutes after birth, while the other lived until approximately 26 h of age. Blood samples obtained from the calf that lived for 26 h showed it to be extremely acidotic and hypoglycemic; this calf also had marked difficulty thermoregulating. At necropsy, all calves showed evidence of in utero gasping and hypoxia, suggestive of premature placental separation. None of the recipient cows showed typical signs of impending parturition. After parturition, lactogenesis in all recipient cows was markedly decreased. On gross examination, placentae resulting from the interspecies transfers had fewer cotyledons that were also much larger in size compared to cotyledons from normal gaur placentae. Calves in this study had abnormalities consistent with those noted from previous interspecies transfers and with IVF and nuclear transfer (cloned) calves. Due to the design of this study, it is not possible to differentiate between problems resulting from the IVF process and those resulting from potential interspecies incompatibilities. However, interspecies transfers of in vitro-produced gaur embryos into Bos taurus are strongly discouraged.

A comparative study of two techniques used in immediate postplacental insertion (IPPI) of the Copper T-380A IUD in Shanghai, People's Republic of China.

Chinese women in Shanghai who delivered vaginally and who chose to use an IUD for contraception received a Copper T-380A IUD inserted vaginally within 10 minutes after delivery of the placenta (i.e., immediate postplacental insertion, IPPI). Among them, 97.7% were primipara. The women were randomly divided into two groups: IUD inserted by hand and IUD via ring forceps. The follow-up rate of six months was 95.2%. Using Tietze's life table method and log rank test, the expulsion and other discontinuation rates were compared at three and six months postinsertion between these two different insertion techniques. Expulsions were the main reason for discontinuation. The six-month gross cumulative expulsion rates were 13.3 and 12.7 per 100 women in the hand-insertion group and ring forceps-insertion group, respectively. Discontinuation rates for medical removals (bleeding/pain) were 2.1 and 1.0 in these two groups, respectively. Neither of the differences was statistically significant (p > 0.05). No uterine perforation, infection or pregnancy occurred. The results suggest that these two different insertion techniques do not significantly affect discontinuation rates in vaginal IPPI using the TCu 380A, and the TCu 380A appears to be suitable for postpartum insertion in Chinese women. Other relevant issues, such as breastfeeding and IUD placement in uterine cavity, are also analyzed and discussed in this report.

Somatic tissue responses of male rhesus monkeys treated with a contraceptive steroid formulation.

Rhesus monkeys (Macaca mulatta) were treated with testosterone (100 micrograms/kg/day) plus estradiol (0.5 micrograms/kg/day) via subcutaneous polydimethylsiloxane (PDS; Silastic) implants for thirteen months. This steroid regimen inhibited dramatically spermatogenesis. Gross and histopathological examination of the musculoskeletal, circulatory, endocrine (excluding the testis), central nervous, gastrointestinal and respiratory systems failed to uncover any untoward effects of the long-term exposure due to the contraceptive formulation. Similarly, no remarkable effects were observed in the ionic, chemical and formed elements of blood or secondary sex structures. Failure to detect secondary complications attributed to the steroid treatment offers further justification for evaluating a contraceptive strategy based on administering naturally occurring steroids at sustained rates approximating those at which they are produced endogenously in the human male.

Regional blood flow occlusion during extensive pelvic procedures for ovarian cancer: a randomized trial.

The objective of this study was to determine the effects of intraoperative aortic occlusion on blood loss and operative time when used during en bloc resection of internal reproductive organs, pelvic peritoneum, and rectosigmoid colon [modified posterior exenteration (MPE)] for primary cytoreduction of ovarian cancer. Patients undergoing MPE, without palpable distal aortic plaque or calcification, were randomized to: (a) complete distal aortic occlusion (</=60 min, with heparin and protamine reversal) with a vascular clamp immediately before MPE, (b) bilateral hypogastric artery occlusion, or (c) no regional blood flow occlusion. Outcomes were compared with respect to blood loss, operative time, and the transfusion rate (anova analysis of variance). Fifty-six patients were accrued. Groups were equivalent with respect to age, disease severity, extent of upper abdominal surgery done, and cytoreductive outcomes. Aortic occlusion significantly reduced the total operative time (P = 0.02), estimated blood loss (P = 0.01), transfusion rate (P = 0.02), hospital stay (P = 0.05), and both operative time (P </= 0.001) and blood loss (P </= 0.001) specifically associated with MPE. There were no immediate or delayed complications due to aortic clamping. Aortic occlusion significantly reduces the blood loss and operative time for patients requiring MPE in the context of primary cytoreductive operations.

Energy requirements for growth in relation to sexual size dimorphism in marsh harrier Circus aeruginosus nestlings.

Food consumption was measured in six female and seven male hand-raised marsh harrier (Circus aeruginosus) nestlings. Females consumed on average 4,321 g and males consumed 3,571 g of food during the nestling stage from 0 to 36 d. Total consumption until 56 d was 6,960 g and 5,822 g for females and males, respectively. On the basis of Fisher's sex ratio theory, this food intake ratio of 0.46 (intake male/[intake male + female]) would explain the observed male-biased fledging sex ratio of 55% males in marsh harrier broods. Growth, gross energy intake, and metabolizable energy intake were measured, along with metabolism of the nestlings, enabling us to determine energy allocation. The assimilation quotient (Q = 0.72) did not differ systematically between the sexes. Differences in metabolic rates between males and females at 15 and 30 d of age were fully attributable to the difference in body mass. Sexual size dimorphism in marsh harriers (female body mass around 60 d of age is 1.28 times greater than male mass) did not fully explain the difference in food intake between male and female nestlings: an analysis of energy requirements for growth and body mass in 16 avian species shows that energy intake was less than proportional to the average body mass at release. The data presented in this study are in agreement with Fisher's theory of inverse proportionality between the sex-specific ratios of energy requirements for growth and of offspring numbers in the marsh harrier population.

A high-quality draft genome assembly of Johnsongrass illuminates relationships between polyploidization, crop-wild hybridization, and reproductive biology.

Johnsongrass [Sorghum halepense (L.) Pers.] is an allopolyploid, rhizomatous, perennial grass species and one of the most troublesome weeds in global agriculture. We assembled the first Johnsongrass genome to clarify poorly understood genetic factors influencing variable rates of crop-wild hybridization with cultivated sorghum [S. bicolor (L.) Moench]. The draft genome assembly has a total size of 3.26 Gb and BUSCO completeness of 95.3%. We also report the first evolutionary analysis of INHIBITION OF ALIEN POLLEN (IAP), the only known cross-(in)compatibility locus in the genus. Our results reveal an evolutionary history of genome instability, including the loss of distinct parental subgenomes, and suggest that Nebraska accession 'J-37,' the genome donor, is a segmental allotetraploid that may function as a diploid or aneuploid during meiosis. Genome instability could explain observations of variable ploidies in Johnsongrass and facilitate ongoing hybridization with sorghum where gamete ploidies and IAP alleles match. Given this information, we provide a suggested research framework for studying evolution and gene expression in the Sorghum genus where crop-wild hybridization occurs and for predicting the potential for hybridization between specific crossing partners. Collectively, this work will bolster efforts to study and manage reproductive biology in other crop-wild polyploid complexes.

Environmental toxicants cause sperm DNA fragmentation as detected by the Sperm Chromatin Structure Assay (SCSA).

Studies over the past two decades have clearly shown that reproductive toxicants cause sperm DNA fragmentation. This DNA fragmentation can usually be detected prior to observing alterations of metaphase chromosomes in embryos. Thus, Sperm Chromatin Structure Assay (SCSA)-detected DNA damage is viewed as the molecular precursor to later gross chromosome damage observed under the light microscope. SCSA measurements of animal or human sperm consist of first obtaining a fresh or flash frozen neat semen sample in LN2 or dry ice. Samples are then sent to a SCSA diagnostic laboratory where the samples are thawed, diluted to approximately 1-2 x 106 sperm/ml, treated for 30 s with a pH 1.2 detergent buffer and then stained with acridine orange (AO). The low pH partially denatures DNA at the sites of DNA strand breaks and the AO-ssDNA fluoresces red while the AO-dsDNA fluoresces green. Flow cytometry measurements of 5000 sperm/sample provide statistically robust data on the ratio of red to green sperm, the extent of the DNA fragmentation and the standard deviations of measures. Numerous experiments on rodents treated with reproductive toxicants clearly showed that SCSA measures are highly dose responsive and have a very low CV. Different agents that act on germ cells at various stages of development usually showed sperm DNA fragmentation when that germ cell fraction arrived in the epididymis or ejaculate. Some of these treated samples were capable of successful in vitro fertilization but with frequent embryo failure. A 2-year longitudinal study of men living a valley town with a reported abnormal level of infertility and spontaneous miscarriages and also a seasonal atmospheric smog pollution, showed, for the first time, that SCSA measurements of human sperm DNA fragmentation were detectable and correlated with dosage of air pollution while the classical semen measures were not correlated. Also, young men spraying pesticides without protective gear are at an increased risk for elevated sperm DNA fragmentation. Extensive DNA fragmentation probably cannot be repaired by the egg and the spontaneous abortion rate is approximately 2x higher if a man has more than 30% of sperm showing DNA fragmentation. DNA fragmentation is an excellent marker for exposure to potential reproductive toxicants and a diagnostic/prognostic tool for potential male infertility.

Decreased glutathione S-transferase expression and activity and altered sex steroids in Lake Apopka brown bullheads (Ameiurus nebulosus).

A number of freshwater lakes and reclaimed agricultural sites in Central Florida have been the receiving waters for agrochemical and municipal runoff. One of these sites, Lake Apopka, is also a eutrophic system that has been the focus of several case studies reporting altered reproductive activity linked to bioaccumulation of persistent organochlorine chemicals in aquatic species. The present study was initiated to determine if brown bullheads (Ameiurus nebulosus) from the north marsh of Lake Apopka (Lake Apopka Marsh) exhibit an altered capacity to detoxify environmental chemicals through hepatic glutathione S-transferase (GST)-mediated conjugation as compared with bullheads from a nearby reference site (Lake Woodruff). We also compared plasma sex hormone concentrations (testosterone, 17-beta estradiol, and 11 keto-testosterone) in bullheads from the two sites. Female bullheads from Lake Apopka had 40% lower initial rate GST conjugative activity toward 1-chloro-2,4-dinitrobenzene (CDNB), 50% lower activity towards p-nitrobutyl chloride (NBC), 33% lower activity toward ethacrynic acid (ECA), and 43% lower activity toward Delta5-androstene-3,17-dione (Delta(5)-ADI), as compared with female bullheads from Lake Woodruff. Enzyme kinetic analyses demonstrated that female bullheads from Lake Apopka had lower GST-catalyzed CDNB clearance than did female Lake Woodruff bullheads. Western blotting studies of bullhead liver cytosolic proteins demonstrated that the reduced GST catalytic activities in female Lake Apopka bullheads were accompanied by lower expression of hepatic GST protein. No site differences were observed with respect to GST activities or GST protein expression in male bullheads. Female Lake Apopka bullheads also had elevated concentrations of plasma androgens (testosterone and 11-ketotestosterone) as compared with females from Lake Woodruff. In contrast, male Lake Apopka bullheads had elevated levels of plasma estrogen but similar levels of androgens as compared with male bullheads from Lake Woodruff. Collectively, our studies indicate the presence of reduced GST protein expression, reduced GST conjugative capacity and altered sex steroid homeostasis in female bullheads from a contaminated field site in Central Florida. The implications of these physiological alterations in terms of pollutant biotransformation and reproduction are discussed.
