Gravitational effects in the passive osmotic flows across polymeric membrane of electrolytic solutions.

Results of experimental study of volume flux in one-membrane system were presented. This system contains horizontal, microporous and symmetrical flat polymeric membrane (Nephrophan), which separate water and electrolyte solution. As binary solutions, aqueous ammonia solutions, which density is lower than water density, were used. As ternary solutions the ammonia with KC1 (0.1 or 0.2 mole.l-1) in aqueous solution were used. The density of ternary solutions was lower, higher or the same as water density. Two configurations of membrane system (A and B) in gravitational field were studied. In configuration A, water was in compartment over the membrane and the solution was under the membrane. In configuration B the succession was reverse. The thermodynamic model of flux graviosmotic effect was elaborated, and the calculations of this effect were performed for A and B configurations of one membrane system. The experimental results are interpreted in terms of gravitational instability that reduces concentration boundary layer dimensions and increases the diffusion permeability coefficient value of the complex: boundary layer/membrane/boundary layer.

The Second Annual Symposium of the NASA Specialized Center of Research and Training (NSCORT) in Gravitational Biology.

The second annual meeting of the NSCORT in Gravitational Biology was held at Kansas State University on September 29-October 1, 1992. Symposium presentations at the meeting included ones on basic gravitational cellular and developmental biology, spaceflight hardware for biological studies, studies on Space Shuttle, and special talks on Space Station Freedom and on life support systems.

Illustrative disorders of ectopic skeletal morphogenesis: a childhood parallax for studies in gravitational and space biology.

Heterotopic ossification is a key feature of at least three distinct genetic disorders of osteogenesis in humans: fibrodysplasia ossificans progressiva, progressive osseous heteroplasia, and Albright's hereditary osteodystrophy. All three conditions are genetic disorders of childhood, but the pathobiology of osteogenic induction, the histopathology of osteogenesis, the anatomic distribution of heterotopic lesions, and the developmental patterns of disease progression differ among the three conditions. The phenotypic distinction of these disorders is critically important in counselling patients and families as well as in advancing research to define the molecular pathophysiology of heterotopic osteogenesis in these disabling genetic disorders. Genetic disorders of tissue modeling and morphogenesis provide an important parallax to disturbances of tissue re-modeling that are of paramount importance to gravitational and space biologists as humans begin to explore and live in environments beyond the planet on which they evolved. Disorders of osteogenesis are of particular concern to space biologists due to the dramatic changes in skeletal biology in altered gravitational fields.

Gravitational effects on biological systems.

The possible effects of the earth's gravitational field on biological systems have been studied from a quantitative point of view, focusing the attention to a very simple system, a solution containing proteins, which biochemists might use in experiments. Gravity has been compared with other forces which are known to influence protein activity, including thermic agitation, weak electrostatic interactions, Van der Waals forces and viscous dissipation. Comparisons have been described in terms of the energy of the interaction per mole, referring to some physically simple cases and substances of biological interest. From this study it is evident that the earth's gravitational energy should be taken into account when considering the chemical behaviour of solutions containing substances that have high molecular weight, such as a typical protein, since its value is comparable to other weak interactions. Moreover, since solutions represent the basis of much more complex biological processes taking place inside cells, the influence of gravity should extend also to cellular biochemical behaviour, especially in presence of altered gravity, both in microgravity (such as on satellites orbiting around the earth), and in macrogravity (such as in a centrifugating biological system).

[Periodical changes of various hematological parameters of the human body adaptation and gravitation fields variations].

Monitoring the content of lymphocytes and nucleated neutrophils (observation period 10.5 months) and the determination of the values of leucocytes coefficient and erythrocyte sedimentation rate in chronic patients revealed rhythms of oscillations of these parameters (from 3-5 to 33 days). The coincidence of these rhythms with the rhythms of variations of gravitational field indicates that gravitational field affects the quantitative blood cell composition and the rheological properties of blood.

[Changes in the spinal cord of rats following the effects of single and multiple gravitational overloads of the "head-pelvis" direction].

The state of the neurons of the spinal cord and intervertebral ganglia in rats was studied morphologically after effects of single (12 units) and repeated for ten days with increase (from 3 to 12 units) gravitation stresses of the head-pelvis direction. Single and repeated gravitation stresses caused both hemodiscirculatory and considerable dystrophic alterations in the spinal cord. These processes were of undulatory character and were better pronounced during a month after single stresses. After repeated stresses they were less pronounced.

Effect of hypokinesia and the combined action of gravitational load and hypokinesia on the structure of the hepatic portal system.

General hypokinesia during 1--6 weeks resulted in dilatation of the interlobular veins. sinusoids and central veins. The sequence of alterations corresponded to terms of hypokinesia. After exposure to "gravitation stress--hypokinesia for 1--6 weeks" stagnation in the portal system of the liver was less than after exposure to hypokinesia alone, but unevenness of lumens in the interlobular veins and sinusoids was more pronounced. The foci of the vessel spasm were determined. The signs of stagnation in the system of the portal vein and unevenness of the width of all the links of the portal bed were most pronounced after combination "hypokinesia for 1--6 weeks-- gravitation stress".

Gravitational influences upon the maintenance requirements of rabbits.

The gravitational component of the feed requirement for the maintenance of body mass has been reported previously for chickens. Similar measurements are reported here for chronically accelerated Polish rabbits, which have mature body masses around 2 kg, approximately the same as for the single-comb white Leghorn chickens used in the earlier experiments. The gravitational influences upon the maintenance requirement and upon body composition were found to be essentially similar for rabbits and chickens, indicating a general phenomenon for small homoiotherms. The principal response in both groups is a greater feed maintenance requirement which increases linearly with the applied acceleration field. The nature of this response is similar to that which follows lesioning of the lateral hypothalamus in rats, though no causal connection between the two phenomena is necessarily implied by these present studies. In monkeys a qualitatively different response of increased body fat is observed.

[Morphologic changes in the nervous apparatus of the pharynx, esophagus and stomach of cats following a single gravitational stress].

Under investigation were the pharynx, oesophagus and stomach of 23 cats subjected to a single maximum endurable gravitation stress. The material was treated after Bielschowski-Gross. It has been established that as early as within 48--72 hours after gravitation effects there appear changes of different elements of the nervous apparatus of the organ under study such as increased argirophilia of nervous structures and reactive changes of different sensory nerve terminations. Motor animal nerve terminations do not change. Most of myelinated afferent fibres and non myelinated vegetative fibres undergo reactive changes. Within 7 days there appear certain myelinated fibres which are in the stage of granular desintegration,

Remodeling of the rat saphenous vein network in response to long-term gravitational load.

Our main objective was to test whether chronic orthostatic body position induces network changes in the saphenous vein superficial tributary system of the rat. Fourteen male Sprague-Dawley rats were kept in tilted tube cages (45-degree head-up position) for two weeks to induce chronic gravitational load to their leg veins. Ten animals housed in normal cages and four animals kept in horizontally positioned tube cages served as controls. The whole superficial network of the left saphenous vein was microprepared surgically under anesthesia, superfused with saline and observed under a videomicroscope, while normal flow and pressure were maintained in the lumen. Branching angles, lengths of venous segments and their diameters were measured offline from digitized images using special image-analyzing software. Several branching angles at the popliteal confluence were significantly reduced by 12.5-15.8 %. The in vivo diameter of the main branch (936+/-34 vs. 805+/-44 microm) and of one of the popliteal tributaries (776+/-38 vs. 635+/-36 microm) increased (p<0.05), comparing vessels from tilted animals with those from normal controls. Maintaining the animals in horizontal tube cages did not induce the above alterations. The increased diameters and reduced branching angles of the saphenous vein network observed are adaptive responses of the venous network to a long-term gravitational load.

[Effect of mild gravitational overload on the progression of lower limb arteriosclerosis obliterans].

The paper analyses the effect of mild gravitational overload (MGO) produced by patient's centrifugation in short-radius centrifuge in cranio-sacral orientation (+Gz) as a part of combined therapy for lower limb arteriosclerosis obliterans. Total 211 patients with I-II grade disease were divided into 3 groups: the first group (59 patients) was treated only by MGO, the second (100 patients) received both MGO and conventional conservative therapy, the third control group (52 patients) underwent conventional therapy only. The best clinical outcome was achieved in the 1st and 2nd groups. Pain-free walking distance increased 2-5-fold (vs 1.5 in control), mean volume blood flow velocity in different arterial beds increased 21-33% (vs 7.5% in control), index of regional perfusion - 25-29% (vs 10% in control), ankle-brachial index - 0.09-0.17 (vs 0.03 in control). Computer termography revealed recovery of leg and foot termographic picture in 86.67%, 92.59%, and 42.86% of cases in the 1st, 2nd and 3rd groups, respectively. Similarly, exercise tolerance measured by bicycle ergometry increased in the 1st and 2nd groups 1.5-1.9-fold (mean 60-85 s) and 1.3-fold (mean 37 s) in control. Thus mild gravitational overload has demonstrated clinical effectiveness in complex therapy of lower limb arteriosclerosis obliterans.

[The connection of medical indicators with oscillations in the natural gravitational field].

The hourly amount of 3140 hospital deaths, 2220 births by terminated labours, and 617 investigations of some elementary functions of haemodynamics and external respiration are correlated with tidal gravitation cycles calculated at the same time of day. The statistical analysis revealed significant dependence of the obtained data on local geometrical properties of 12-hour lunar tidal waves. Their distinctive signs were convexity-concavity and growing-sinking which the 12-hour lunar wave into 4 intervals. To each interval corresponded specific patterns of death and birth curves during 24-hours and divergent distribution of values of haemodynamics and external respiration. The investigation results and literature data point to the fact that periodic fluctuations of natural gravitation fields carrying the information on changes in space-time may serve as an outside time-marker (Zeitgeber) of biological rhythms.

[Morphofunctional properties of the blood cells in patients with ischemic heart disease following gravitational correction].

Red cell and platelet surface membranes, platelet adhesion to glass were examined in coronary patients treated among other treatment modalities with gravitation surgical correction of the blood. Scanning electron microscopy was used in the study. The studies revealed the minimal injurious effect of the extracorporeal contour on blood cells; a manifest improvement of cellular morphology and function was observed after a course of procedures. An attempt was made to assess the mechanism of gravitation surgery effect on blood cells.

[Effect of rotation and vibration on orientation in man in relation to the gravitational vertical].

The man's ability to get oriented in relation to the gravitational vector was investigated. The test subjects were exposed either to rotation in 1 m arm centrifuge or tilting in a chair. They were simultaneously exposed to total-body vertical vibration of 20 Hz. As the exposure continued, the ratio of the perceived vertical (in the absence of visual keys) and the apparent body position changed significantly. It is concluded that the tests used to evaluate the pilot ability for spatial orientation should include evaluations of the subjective vertical and body position relative to the gravitational vector. The tests should be performed during exposures to simulated dynamic flight factors.

[Effect of weightlessness and artificial gravitation on thyroid gland morphology].

The investigation of the thyroid gland was carried out in Wistar rats, SPF colony 4.5--13 h and 25 days after a 18.5 days flight on board the space biosatellite "Cosmos-936". In animals subjected to weightlessness, moderate symptoms of the thyroid hypofunction were observed, statistically significant decrease in number and volume of the nuclei in calcitonin-secreting cells (C-cells) was especially pronounced during 4.5--9 h after landing. Similar but less pronounced changes were observed in C-cells of the rats subjected to artificial conditions of space flight, besides weightlessness. The similarity of the changes in the animals of both groups made it possible to connect the increasing amount of C-cells and the morphological symptoms of their functional inhibition with the effect of weightlessness and hypokinesia. During the space flight, the animals were kept under the conditions of artificial gravitation on board the biosatellite and therefore morphological peculiarities specific for the earth conditions were preserved in C-cells and the thyroid gland. Thus, it was concluded that artificial gravitation prevented the development of the thyroid changes which appeared under the influence of weightlessness.

[Effect of artificial gravitation on the skeletal musculature of rats during space flights].

Comparative morphological and histochemical investigation of the rat skeletal muscles after a 18.5 days flight in the space ship "Cosmos-936" under conditions of weightlessness and artificial gravitation created by means of a centrifuge has demonstrated that weightlessness creates muscular atrophy, with fibres of different types affected in different muscles. The greatest changes take place in the soleus muscle. Metabolic disorders in the muscular tissue are manifested as an excessive deposition of glycogen and an increased content of phospholipids in it. The artificial gravitation favourably affects the state of the skeletal musculature preventing the development of metabolic disorders, though it does not completely avert the appearance of atrophic processes in it.

[Effect of combined exposure to hypokinesia and gravitational stress on the structure of the wall of the renal vein].

Renal veins from 25 rabbits were investigated by means of imbedding in paraffin with subsequent hematoxylin--eosin staining, after Van Gieson and with fuchselin after Hart. Prolonged hypokinesia (over 3 weeks) with subsequent single application of gravitational overloading was stated to produce some structural changes in the wall of the renal vein. They are manifested as loosening of the middle vascular sheath with vacuolization of myocytes and outgrowth of its elastic elements. Hypokinesia of longer duration (8 weeks) with subsequent gravitational overloading produces atrophy of smooth muscle tissue in the middle sheath of the venous wall that brings about decreasing in tonus of the wall of the renal vein.
