Diagnostic significance of in vitro T-cell proliferative responses to house-dust mite Der p 1 in children with dust-mite allergy.

The study aimed to investigate the possible diagnostic significance of T-cell proliferative responses to purified Der p 1 antigen in allergic children with and without allergic sensitization to house-dust-mite (HDM) allergens. T-cell reactivity to purified Der p 1 antigen was analyzed in 24 children with allergic sensitization to HDM demonstrated by RAST and/or positive skin prick tests. Twelve healthy young adults and 11 children with allergic sensitizations other than HDM served as controls. Of 25 HDM-allergic patients, 16 displayed strong T-cell reactivity to Der p 1 (stimulation indices > 3): nine patients showed no T-cell proliferation despite the presence of specific IgE, and five showed no responses despite positive skin prick test. In two patients, a weak T-cell response to Der p 1 could be demonstrated in the absence of specific IgE and negative skin test result. The two affected subjects did not show evidence of mite allergy. No T-cell responses were observed in adult controls (stimulation indices < 3). We conclude that the assessment of T-cell reactivity to Der p 1 is of little value for the diagnosis of HDM allergy. The importance of T-cell proliferative responses for the study of the pathogenesis of HDM allergy remains unchallenged.

Role of bioavailable iron in coal dust-induced activation of activator protein-1 and nuclear factor of activated T cells: difference between Pennsylvania and Utah coal dusts.

Activator protein-1 (AP-1) and nuclear factor of activated T cells (NFAT) are two important transcription factors responsible for the regulation of cytokines, which are involved in cell proliferation and inflammation. Coal workers' pneumoconiosis (CWP) is an occupational lung disease that may be related to chronic inflammation caused by coal dust exposure. In the present study, we demonstrate that coal from the Pennsylvania (PA) coalmine region, which has a high prevalence of CWP, can activate both AP-1 and NFAT in JB6 mouse epidermal cells. In contrast, coal from the Utah (UT) coalmine region, which has a low prevalence of CWP, has no such effects. The PA coal stimulates mitogen-activated protein kinase (MAPK) family members of extracellular signal-regulated kinases (ERKs) and p38 MAPK but not c-Jun-NH(2)-terminal kinases, as determined by the phosphorylation assay. The increase in AP-1 by the PA coal was completely eliminated by the pretreatment of cells with PD98059, a specific MAPK kinase inhibitor, and SB202190, a p38 kinase inhibitor, further confirming that the PA coal-induced AP-1 activation is mediated through ERKs and p38 MAPK pathways. Deferoxamine (DFO), an iron chelator, synergistically enhanced the PA coal-induced AP-1 activity, but inhibited NFAT activity. For comparison, cells were treated with ferrous sulfate and/or DFO. We have found that iron transactivated both AP-1 and NFAT, and DFO further enhanced iron-induced AP-1 activation but inhibited NFAT. These results indicate that activation of AP-1 and NFAT by the PA coal is through bioavailable iron present in the coal. These data are in agreement with our previous findings that the prevalence of CWP correlates well with levels of bioavailable iron in coals from various mining regions.

Follow-up study on pulmonary function of dust workers: an epidemiological evaluation of discontinued workers of dust operations.

An epidemiologic study on 176 dust workers from various facilities was initiated in 1978. Their mean age was 48.3 years and mean duration of employment was 21.6 years. No workers complained of chronic cough and phlegm. Their chest X-rays were found to belong to Category I of the diagnostic criteria for pneumoconiosis in Japan. Pulmonary function test was performed on these workers, however, 54 workers discontinued taking part in the study by the end of 1979, and 49 workers had discontinued by 1981. A total of 73 workers had a five-year follow-up survey of pulmonary function. Forced expiratory volume in one second divided by forced vital capacity (FEV1.0/FVC) and forced expiratory flow rate at 25% of FVC divided by height (V25/H) were mainly compared by smoking habit, work years, type of job, size of company, and the time when they discontinued their jobs. Predicted percentage values of lung function were used for the adjustment of the influence of workers' ages. Decrement of FEV1.0/FVC and V25/H in the five-year follow-up group were larger than the screening level. The mean values of those functional parameters in the early discontinued group were lower than late discontinued and active working groups. The active working group was considered to be a physically healthy workers' population.

[House dust mold in Istanbul. I. Detection of mold flora in bed dust].

In our study, mycological analyses were carried out on 110 randomly chosen houses for bed dusts. 748 mould strains belonging to 117 genus and species were isolated and identified. Air-borne pathogenic fungi, fungi that are causative agents of pulmonary allergy such as Cladosporium spp., Aspergillus glaucus group, Alternaria, Ulocladium, Aureobasidium pullulans, Aspergillus niger group were isolated.

[Correlation between changes in lungs and the composition of amino acids in blood plasma caused by industrial dust. II. Studies on workers occupationally exposed to industrial dust].

Results of determination of free aminoacids in blood plasma of workers exposed to ceramic and grinding dusts as well as welding fumes are presented. In the blood plasma of those employed in ceramic factory or welders a significant decrease of free aminoacids in comparison with the control group was found except for hydroxyproline, whose content increased during the 25 year-employment. In experiments on animals a significant increase of total aminoacids content in lungs with simultaneous decrease of free aminoacids amount in blood plasma was observed. Similar changes in workers' blood plasma make us conclude that aminoacids content in human lungs may also increase, which is indicative of fibrinogenic processes.
