Tyrosinase-catalyzed binding of 3,4-dihydroxyphenylalanine with proteins through the sulfhydryl group.

The cytotoxicity of catechols has been ascribed to covalent binding of the omicron-quinone oxidation products to proteins through sulfhydryl groups. The nature of the covalent binding was studied with dopaquinone formed on tyrosinase oxidation of 3,4-dihydroxyphenylalanine (DOPA). After acid hydrolysis of the reaction products, cysteinyldopas liberated (protein-bound cysteinyldopas) were determined by HPLC with electrochemical detection. When 0.1 mM DOPA was oxidized in the presence of 0.2 mM bovine serum albumin, alcohol dehydrogenase or isocitrate dehydrogenase, protein-bound cysteinyldopas were formed in yields of 5.4, 44, or 33%, respectively. The covalent binding was almost completely inhibited by 1 mM cysteine or 1 mM ascorbic acid, but 10 mM lysine had no effect. These results unambiguously demonstrate that dopaquinone can bind with proteins mostly through sulfhydryl groups.

1,4-benzothiazines as key intermediates in the biosynthesis of red hair pigment pheomelanins.

Following the discovery of cysteinyldopas as the early intermediates in the biogenesis of pheomelanins, the typical red hair pigments, the reactivity of the biosynthetic precursors under biomimetic conditions was extensively investigated. As a result, the early stages of pheomelanogenesis were envisaged as involving oxidative cyclization of cysteinyldopas, mainly the 5-S-isomer, to 1,4-benzothiazine (BTZ) intermediates which undergo oxidative polymerization leading eventually to the pigments. In the last decade, several aspects of the chemistry and biosynthesis of pheomelanins were re-examined. In particular, (i) transient BTZ intermediates were identified by pulse radiolytic techniques and NMR analysis; (ii) the effect of reaction conditions and additives on the rearrangement vs. redox exchange reaction paths of such intermediates were investigated in detail; (iii) the mechanism of the oxidative polymerization of BTZs was characterized by the first isolation of oligomer species, and (iv) the pigment eventually resulting from oxidation of 5-S-cysteinyldopa (CD) was directly analyzed by spectroscopic and chemical methodologies in comparison with pheomelanins isolated from human hair. These advances led eventually to an integrated picture of the biogenetic route highlighting the intervention of various chemical and enzymatic factors which affect the kinetics of the different steps and the nature of the key benzothiazine precursors. A likely biogenetic route was also postulated for the delta2,2'-bi(2H-1,4-benzothiazine) pigments, termed trichochromes, whose origin had remained an open issue since their first isolation from red human hair and avian feathers. Finally, a more detailed description of the structure of pheomelanin pigments in terms of the monomer units, their mode of linking, and postsynthetic modifications was gained.

Identification of glypican-3 as a novel tumor marker for melanoma.

PURPOSE: We reported recently the novel tumor marker glypican-3 (GPC3) for hepatocellular carcinoma. In the present study, we investigated the expression of GPC3 in human melanoma cell lines and tissues and asked whether GPC3 could be a novel tumor marker for melanoma. EXPERIMENTAL DESIGN: Expression of GPC3 mRNA and protein was investigated in human melanoma cell lines and tissues using reverse transcription-PCR and immunohistochemical analysis. Secreted GPC3 protein was quantified using ELISA in culture supernatants of melanoma cell lines and in sera from 91 patients with melanoma and 28 disease-free patients after surgical removal of primary melanoma. All of the subjects were Japanese nationals. RESULTS: In >80% of melanoma and melanocytic nevus, there was evident expression of GPC3 mRNA and protein. Furthermore, GPC3 protein was evidenced in sera of 39.6% (36 of 91) of melanoma patients but not in sera from subjects with large congenital melanocytic nevus (0 of 5) and from healthy donors (0 of 60). Twenty-seven of 36 serum GPC3-positive patients were negative for both serum 5-S-cysteinyldopa and melanoma-inhibitory activity, well-known tumor markers for melanoma. The positive rate of serum GPC3 (39.6%) was significantly higher than that of 5-S-cysteinyldopa (26.7%) and of melanoma-inhibitory activity (20.9%). Surprisingly, we detected serum GPC3 even in patients with stage 0 in situ melanoma. The positive rate of serum GPC3 at stage 0, I, and II (44.4%, 40.0%, and 47.6%) was significantly higher than that of 5-S-cysteinyldopa (0.0%, 8.0%, and 10.0%). Also observed was the disappearance of GPC3 protein in sera from 11 patients after surgical removal of the melanoma. CONCLUSIONS: GPC3 is apparently a novel tumor marker useful for the diagnosis of melanoma, especially in early stages of the disorder.

Reactions of D-glucose with phenolic amino acids: further insights into the competition between Maillard and Pictet-Spengler condensation pathways.

The reactions of 5-S-cysteinyldopa, L-alpha-methyldopa and DL-m-tyrosine with D-glucose were investigated at 90 degrees C in phosphate buffer at pH ranging from 5.0 to 9.0. Whereas gave mainly the double Maillard condensation product N,N'-bis(1''-deoxy-D-fructos-1''-yl)-5-S-cysteinyldopa, as an inseparable mixture of beta-D-fructopyranosyl and alpha,beta-D-fructofuranosyl derivatives, 2 and 3 gave both Maillard and Pictet-Spengler products, although to different extents and with different regio- and stereochemistry. A peculiar pattern of reactivity was displayed by which gave, besides the Maillard product and the expected 6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline C-1 diastereoisomeric pairs, the unprecedented 7,8-dihydroxy-1,2,3,4-tetrahydroisoquinoline derivative via the ortho cyclization pathway. Pictet-Spengler cyclization of 2 and 3 proceeded with Felkin-Anh-type asymmetric induction, favouring the 1R isomer throughout the pH range 5.0-9.0. These results, which highlight the first example of carbohydrate-derived 7,8-dihydroxytetrahydroisoquinoline, provide new insights into the factors governing competition between Maillard and Pictet-Spengler condensation pathways.

Oxidation of tyrosine residues in proteins by tyrosinase. Formation of protein-bonded 3,4-dihydroxyphenylalanine and 5-S-cysteinyl-3,4-dihydroxyphenylalanine.

A simple and rapid method was developed for the determination of 3,4-dihydroxyphenylalanine (dopa) and 5-S-cysteinyl-3,4-dihydroxyphenylalanine (5-S-cysteinyldopa) in proteins with the use of high-pressure liquid chromatography. With this method, it is demonstrated that mushroom tyrosinase can catalyse hydroxylation of tyrosine residues in proteins to dopa and subsequent oxidation to dopaquinone residues. The dopaquinone residues in proteins combine with cysteine residues to form 5-S-cysteinyldopa in bovine serum albumin and yeast alcohol dehydrogenase, whereas dopa is the major product in bovine insulin, which lacks cysteine residues.

Melanin-related biochemistry of IGR 1 human melanoma cells.

Cultured melanoma cells have been of great value in the study of pigment metabolism. IGR 1 human melanoma cells, established by Dr Christian Aubert, produce melanin in large quantities. These cells have been used for isolation of human tyrosinase which enzyme has not previously been obtained in a pure form. IGR 1 cells contain large amounts of 5-S-cysteinyldopa which is the quantitatively most important catecholic amino acid. This review deals with the metabolism of dopa, cysteinyldopa, glutathionyldopa, cysteine and glutathione, compounds of central importance in pigment metabolism. The information available on tyrosinase, catecholic compounds and on thiols in IGR 1 melanoma cells makes these cells most suitable for further investigation of the metabolism of human melanoma cells.

Trichochromes in the urine of melanoma patients.

The urine of patients with melanoma metastases and increased urinary excretion of 5-S-cysteinyldopa was examined for trichochromes. Five of 16 patients showed urinary excretion of trichochromes B and C. None excreted trichochromes E or F. All patients showing trichochrome excretion had very large amounts of cysteinyldopa in their urine.

Melanins in IGR 1 melanoma cells.

Information on the composition of melanins is obtained by analysis both of 4-amino-3-hydroxyphenylalanine (AHP) after hydriodic acid degradation and of pyrrole-2,3,5-tricarboxylic acid (PTCA) after potassium permanganate oxidation. Analysis of thiazole-4,5-dicarboxylic acid (TDCA) and pyrrole-2,3-dicarboxylic acid (PDCA) after permanganate oxidation, provides additional information on the composition, TDCA on pheomelanin residues, and PDCA on indolic residues without carboxy groups. Using model melanins formed from dopa and cysteinyldopa in different proportions, we found the TDCA/(PTCA+PDCA) ratio to yield a reliable estimate of the relative proportions of pheomelanin and eumelanin. The PDCA/PTCA ratio reflects the relationship between indole residues with and without carboxy groups. We have analyzed degradation products from cultures of IGR 1, an extensively studied melanoma cell line. Cell cultures were harvested after 2, 4, and 7 days. Culture media were changed after 2 days in all series, and also after 4 days in one series harvested at 7 days. Cells without medium change had seven times the amount of melanin found in cultures with medium change. The PDCA/PTCA ratio decreased with increasing amounts of melanin. With increased melanization, eumelanin is increased relatively more than pheomelanin. The cell content of 5-S-cysteinyldopa (5-S-CD) was similar in all cultures, while 6-hydroxy-5-methoxyindole-2-carboxylic acid (6H5MICA), a eumelanin precursor metabolite, was found in increased amounts of media of heavily pigmented cultures.

Quantitative analysis of tyrosinase transcripts in blood.

BACKGROUND: Tyrosinase is an enzyme unique to pigment-forming cells. Methods using this transcript for detection of melanoma cells in blood have given divergent results. Quantitative analytical procedures are therefore needed to study the analytical performance of the methods. METHODS: Mononucleated cells were isolated by Percoll centrifugation. RNA was isolated by each of three methods: Ultraspec(TM)-II RNA isolation system, FastRNA(TM) GREEN Kit, and QIAamp RNA Blood Mini Kit. cDNA was synthesized using random hexamer primers. A tyrosinase-specific product of 207 bp was amplified by PCR. As an internal standard (and competitor) we used a 207-bp cDNA with a base sequence identical to the tyrosinase target except for a 20-bp probe-binding region. The PCR products were identified by 2, 4-dinitrophenol (DNP)-labeled probes specific for tyrosinase (5'DNP-GGGGAGCCTTGGGGTTCTGG-3') and internal standard (5'DNP-CGGAGCCCCGAAACCACATC-3') and quantified by ELISA. RESULTS: The calibration curves were linear and had a broad dynamic measuring range. A detection limit (2 SD above zero) of 48 transcripts/mL of blood was obtained from a low control. The analytical imprecision was 50% and 48% at concentrations of 1775 and 17 929 transcripts/mL (n = 12 and 14, respectively). With the cell line SK-Mel 28 added to blood and RNA extracted with the Ultraspec, Fast RNA, and QIAamp RNA methods, we found (mean +/- SD) 1716+/-1341, 2670+/-3174, and 24 320+/-5332 transcripts/mL of blood. Corresponding values were 527+/-497, 2497+/-1033, 14 930+/-1927 transcripts/mL of blood when the cell line JKM86-4 was added. One high-risk patient was followed by repeated analysis of tyrosinase transcripts in blood. The melanoma marker 5-S-cysteinyldopa in serum and urine was within reference values, but tyrosinase mRNA was slightly increased (120-168 transcripts/mL of blood). The tyrosinase mRNA increased to 1860 transcripts/mL concomitant with the increase in 5-S-cysteinyldopa; later a spleen metastasis was found. CONCLUSIONS: The results obtained with different RNA extraction methods illustrate the importance of quantitative methods for validation of methods. The use of QIAamp RNA improved the extraction efficiency considerably. Data from a case study suggest the assay is suitable in the follow-up of patients with high risk of developing metastases.

[Correlation between differentiation and malignancy in human malignant melanocytes "in vivo" and "in vitro" (author's transl)].

The relationships between differentiation and malignant transformation were studied in human malignant melanomas in vivo and in vitro. Melanocyte differentiation was assessed by ultrastructural morphological characteristis (the appearance of the melanosomes and related structures) localization of dopa-oxidase and assay of 5-S-cysteinyldopa, a specific metabolite. The transformed characteristic of the cells in vitro was evaluated by their ability to give rise to established cell lines, karyological modifications and heterotransplantation in Nude mice and Syrian hamsters. Morphological variability of the cells in malignant melanomas is accompanied by variability in the localization of dopa-oxidase, the level of 5-S-cysteinyldopa, chromosome pattern and their heterotransplantibility. The lack of pigmentation in some malignant melanoma lines can result from either an irreversible loss of some functions which give rise in melanization and the malignancy in maintained, or by phenomenon of regulation determined by intra or extra-cellular factors with the loss of heterotransplantability. Modulation phenomena affecting tumorigenicity and pigmentation although sometimes concomitant are not identical.

Melanocyte metabolites in the urine of people of different skin colour.

The urinary excretion of 2 melanocyte metabolites was studied in normal people of different skin type. The sulphur-free indole derivative 6-hydroxy-5-methoxyindole-2-carboxylic acid was excreted in larger quantities by people with genetically dark skin, whereas the excretion of 5-S-cysteinyldopa was not related to pigment type. No correlation between 5-S-cysteinyldopa and 6-hydroxy-5-methoxyindole-2-carboxylic acid excretion emerged.

Metabolism of 5-S-cyteinyldopa by O-methylation.

5-S-cysteinyldopa is metabolized by O-methylation on incubation with a liver extract and S-adenosyl methionine. O-methylated 5-S-cysteinyldopa isolated from melanoma urines by ion exchange and paper chromatography was identified by gas chromatography-mass spectrometry and NMR.

Sunlight, melanogenesis and radicals in the skin.

Melanocytes are cells of neural crest origin residing at the dermal-epidermal juncture. They produce specialized organelles called melanosomes within which the biochemical processes of melanization occurs. UV radiation is capable of inducing melanogenesis and, during the biosynthesis of melanins, several of the putative precursors "leak out" of the melanosome and can be detected in the skin, serum and urine of individuals undergoing active melanogenesis. Most notable are the cysteinyldopas (formed by nucleophilic addition of cysteine to dopaquinone) and several dihydroxyindoles (formed by intramolecular cyclization of dopaquinone). These catechols often are methylated in the melanocyte to afford a mixture of the monomethoxy derivatives and, in some cases, the dimethoxy species. Recent investigations in our laboratories have demonstrated that the cysteinyldopas, dihydroxyindoles, and their various methylated derivatives are photochemically unstable. Irradiation with biologically relevant ultraviolet radiation (i.e. wavelengths greater than 300 nm) results in the rapid destruction of the precursors/metabolites and the production of a variety of free radical species. The photochemistry and potential photobiological significance of melanogenic intermediates is discussed.

Covalent binding of catechols to proteins through the sulphydryl group.

The cytotoxicity of catechols has been ascribed to covalent binding of the o-quinone oxidation products to proteins through the sulphydryl group. We have previously shown that dopaquinone can bind covalently to proteins through cysteine residues to form protein-bound cysteinyldopas. In this study, we have compared the reactivities of o-quinones, derived from tyrosinase oxidation of various catechols, with the cysteine residue in bovine serum albumin. o-Quinone forms of dopamine, norepinephrine, N-acetyldopa, N-acetyldopamine, 3,4-dihydroxyphenylacetic acid, pyrocatechol and 4-methylcatechols were much more reactive than dopaquinone, while o-quinone forms of 5-S-cysteinyldopa and epinephrine were much less reactive. The yield of protein-bound cysteinylcatechols appears to depend on a competition between the intermolecular nucleophilic reaction of sulphydryl groups in protein and the intramolecular nucleophilic reaction of an amino group in the side chain.

Chemical characterization of pheomelanogenesis starting from dihydroxyphenylalanine or tyrosine and cysteine. Effects of tyrosinase and cysteine concentrations and reaction time.

Two types of melanin pigment are produced in mammals; the brown-to-black eumelanins and the yellow-to-reddish-brown pheomelanins. The switch from one type of melanin to the other appears to be regulated by the levels of tyrosinase and thiols, such as cysteine and glutathione. This study examines the process of pheomelanin formation starting from dihydroxyphenylalanine (dopa) or tyrosine and cysteine. We prepared pheomelanins by tyrosinase oxidation of dopa or tyrosine in the presence of cysteine. Experimental variables were reaction time, tyrosinase concentration, and dopa or tyrosine to cysteine ratio. Following the reactions, we measured concentrations of tyrosine, dopa, cysteine and cysteinyldopas, amounts of total melanin (TM) by Soluene-350 solubilization and aminohydroxyphenylalanine (AHP), a specific indicator of pheomelanin, formed by hydriodic acid hydrolysis, and absorbance ratio, A650/A500. It was found that (1) mixed melanogenesis is a heterogeneous process in which pheomelanogenesis proceeds first, followed by eumelanogenesis, as shown by changes in the tyrosine and cysteinyldopa concentrations, the AHP/TM ratio, and the A650/A500 ratio during the course of melanogenesis and (2) lower tyrosinase concentration favors pheomelanogenesis even when the availability of cysteine is limited, as shown by AHP/TM ratios that were higher than the corresponding tyrosine to cysteine ratios. These results indicate that the switch from eumelanogenesis to pheomelanogenesis can be achieved by lowering the tyrosinase activity, which conforms to our proposal that tyrosinase activity is the major factor controlling the course of melanogenesis.

Transient quinonimines and 1,4-benzothiazines of pheomelanogenesis: new pulse radiolytic and spectrophotometric evidence.

Biosynthetic and model in vitro studies have shown that pheomelanins, the distinctive pigments of red human hair, arise by oxidative cyclization of cysteinyldopas mainly 5-S-cysteinyldopa (1) via a critical o-quinonimine intermediate, which rearranges to unstable 1,4-benzothiazines. To get new evidence for these labile species, fast time resolution pulse radiolytic oxidation by dibromide radical anion of a suitable precursor, the dihydro-1,4-benzothiazine-3-carboxylic acid 7 was performed in comparison with that of 1. In the case of 7, dibromide radical anion oxidation leads over a few microseconds (k = 2.1 x 10(9) M(-1) s(-1)) to a phenoxyl radical (lambda(max) 330 nm, epsilon = 6300 M(-1) cm(-1)) which within tens of milliseconds gives rise with second-order kinetics (2k = 2.7 x 10(7) M(-1) s(-1)) to a species exhibiting an absorption maximum at 540 nm (epsilon = 2200 M(-1) cm(-1)). This was formulated as the o-quinonimine 3 arising from disproportionation of the initial radical. The quinonimine chromophore is converted over hundreds of milliseconds (k = 6.0 s(-1)) to a broad maximum at around 330 nm interpreted as due to a 1,4-benzothiazine or a mixture of 1,4-benzothiazines, which as expected are unstable and subsequently decay over a few seconds (k = 0.5 s(-1)). Interestingly, the quinonimine is observed as a labile intermediate also in the alternative reaction route examined, involving cyclization of the o-quinone (lambda(max) 390 nm, epsilon = 6900 M(-1) cm(-1)) arising by disproportionation (2k = 1.7 x 10(8) M(-1) s(-1)) of an o-semiquinone (lambda(max) 320 nm, epsilon = 4700 M(-1) cm(-1)) directly generated by dibromide radical anion oxidation of 1. Structural formulation of the 540 nm species as an o-quinonimine was further supported by rapid scanning diode array spectrophotometric monitoring of the ferricyanide oxidation of a series of model dihydrobenzothiazines.

Oxidative Polymerization of the Pheomelanin Precursor 5-Hydroxy-1,4-benzothiazinylalanine: A New Hint to the Pigment Structure.

Biosynthetic studies have shown that pheomelanins, the distinctive pigments of red human hair, arise from oxidative polymerization of cysteinyldopas via 1,4-benzothiazinylalanine intermediates. However, the mode of formation of the pigment polymer remains controversial. To address this point, we have investigated the conversion of the major biosynthetic precursor 5-S-cysteinyldopa (2a) to pheomelanin under biomimetic conditions. Peroxidase/H(2)O(2) oxidation of 2a was shown to lead in the early stages to the 1,4-benzothiazinylalanine 8a, which rapidly declines with concomitant formation of a distinct pattern of oligomeric products. Reduction of the reaction mixture at this stage allowed the isolation of dimer 17 in 10% yield, along with trimers 18 and 19 in smaller amounts. A restricted rotation about the ethereal C-O bond of 17 was apparent by the presence of two NMR-detectable conformational isomers, separated by an activation energy barrier of 17.83 +/- 0.03 kcal mol(-)(1). Under similar oxidation conditions, the model catechol 2b gave the related dimers 15 and 16. The structure of oligomers 17-19, all characterized by C-C and C-O bonds between the benzothiazine units, would suggest that the peroxidase-promoted polymerization proceeds by phenol-type coupling of an aryloxy radical generated by initial one-electron oxidation of 8a. Overall, these results point to a structural model for the pheomelanin polymer which is basically different from that proposed on the basis of degradative studies.

The usefulness of 4-amino-3-hydroxyphenylalanine as a specific marker of pheomelanin.

Reductive hydrolysis of pheomelanin with hydriodic acid (HI) gives two aminohydroxyphenylalanine isomers, 4-amino-3-hydroxyphenylalanine ('specific AHP') and 3-amino-4-hydroxyphenylalanine (3-aminotyrosine, AT), which derive from the oxidative polymerization of 5-S-cysteinyldopa, and 2-S-cysteinyldopa, respectively. Since we first introduced this analytical method, the combined amount of AHP and AT ('total AHP') has been extensively used as a marker of pheomelanin. However, one problem with using total AHP as a marker is that background levels originate from precursors other than pheomelanin. Considerable and variable amounts of background AT are produced from other sources, most likely nitrotyrosine residues in proteins. In order to overcome this problem, we developed HPLC conditions which enable the direct injection of the HI reduction products into the HPLC system allowing good separation of AHP and AT. In this way we could study the importance of both degradation products separately and their specificity as markers for pheomelanin. The usefulness of the present method is validated using human hair samples of various colours which were divided into dark, fair or red colours. The combined amount of specific AHP and AT shows an excellent correlation with total AHP, and the amount of specific AHP also correlates with the amount of total AHP. We also examined total AHP and specific AHP values against pyrrole-2,3,5-tricarboxylic acid (PTCA) values in the human hair samples. These results show that specific AHP measurement gives a more prominent segregation for the ratio of specific AHP to PTCA among hairs of various colours than the ratio of total AHP to PTCA. Thus, we conclude that 'specific AHP' is a more specific marker of pheomelanin than is 'total AHP'.
