Analysis of the immune response to FMDV structural and non-structural proteins in cattle in Argentina by the combined use of liquid phase and 3ABC-ELISA tests.

The successful sanitary campaign implemented to control the 2000-2002 outbreaks of foot-and-mouth disease virus (FMDV) in Argentina was greatly assisted by the combination of an ELISA test (3ABC-ELISA) that detects antibodies directed against FMDV viral non-structural proteins (NSPs) and a liquid phase blocking competitive ELISA (lpELISA) for the detection of antibodies against the viral structural proteins (SPs). The combined use of these two assays in large-scale analysis of field samples allowed for a clear differentiation between infected and uninfected animals, with high specificity and sensitivity, regardless of the animal's vaccination status. In order to validate the application in indirect vaccine potency assays and assessment of vaccination efficiency, a preliminary correlation between serological response and protection from challenge with O1/Campos and A/Arg/01 FMD virus strains was established with data derived from commercial vaccine series challenge trials. Determination of antibodies to NSPs in vaccinated and revaccinated animals proved helpful in the analysis of vaccine purity. A review and discussion of the epidemiological status of cattle herds and real time monitoring of FMD in Argentina using these assays before, during and after the outbreaks is presented.

Pertussis in Argentina and France.

We have studied the epidemiology of pertussis in two countries, Argentina and France, which have similar histories of long-term mass vaccination with a whole-cell vaccine. Both countries display a comparable epidemiology, with an increase of the incidence of the disease in non-vaccinated newborns. We used pulsed-field gel electrophoresis (PFGE) analysis and genotyping to compare Bordetella pertussis clinical isolates recovered in Argentina and France in 2001-2004. The majority of the isolates harbors prn2 allele and belongs to PFGE IVbeta group. Isolates were found to be very similar genetically suggesting a common evolution of the disease in these two countries using the same vaccine.

Dirofilariasis in Argentina: historical review and first report of Dirofilaria immitis in a natural mosquito population.

Argentina is one of the four South American countries where the presence of Dirofilaria immitis is currently confirmed. The objective of this study was to review information on dirofilariasis in the country, and to report our recent findings on mosquito vectors. Since the first report of dogs with unidentified microfilariae in 1926, D. immitis was found in seven provinces and canine prevalence ranged 0-71% at local scale. National prevalence was 8% by the end of the 1980s and current information is available only for Buenos Aires Province. Four pulmonary human infections of D. immitis and one subcutaneous of Dirofilaria sp. were documented. The common coati was the only wild host found, and natural infection in mosquitoes was not previously reported in the country. In our recent mosquito survey in Greater Buenos Aires, we captured and dissected 2380 mosquitoes belonging to 20 species. According to a minimum temperature of 14 degrees C, the potential transmission period (PTP) for D. immitis in Buenos Aires covers 6 months, and the most favourable period (mean temperature above 20 degrees C) takes place from the middle of November to the beginning of April. To identify potential vectors of the parasite, we assessed weekly abundances of mosquito species during those PTP estimated previously. We found two specimens of Culex pipiens and one of Aedes aegypti carrying non-infective stages of D. immitis. These two highly anthropophilic mosquitoes may enhance the role of D. immitis as zoonotic agent in temperate Argentina.

Epidemiological studies on intestinal helminth parasites of the patagonian grey fox (Pseudalopex griseus) in Tierra del Fuego, Patagonia Argentina.

The present work was performed to study the intestinal helminths of the patagonian grey fox (Pseudalopex griseus) and to obtain information about its possible role in the sylvatic life cycle of Echinococcu granulosus in Tierra del Fuego, Patagonia Argentina. Eighty-one foxes were captured and subject to post-mortem analysis. Thirty-one foxes (38.3%) harboured helminths. A total of six helminth species were recovered. Only one adult of E. granulosus was found in the studied samples. The current study is the first report of the intestinal helminths of the patagonian grey fox in Tierra del Fuego, Argentina and showed that this specie is probably not an important reservoir host for E. granulosus.

Epidemiological surveillance of ovine hydatidosis in Tierra del Fuego, Patagonia Argentina, 1997-1999.

Cystic echinococcosis is the most prevalent zoonosis in Tierra del Fuego province, Argentina, with important economic, productive and public health consequences. The present work was performed to determine the ovine prevalence in Tierra del Fuego, Argentina, as well as to evaluate the quality of diagnostic systems in slaughterhouses. Moreover, genetic analyses to characterize the strain of Echinococcus granulosus involved in the region were done. The first actions to perform a diagnosis of the epidemiological situation of hydatidosis in Tierra del Fuego were done between 1976 and 1977. A canine prevalence of 80% and an ovine prevalence of 55% results were obtained. Since 1979 the control program of Hydatidosis of Tierra del Fuego was implemented. It was based on semiannual canine anthelmintic treatment with praziquantel at dose of 5mg/kg, and complemented with sanitary education and canine and ovine epidemiological surveillance. During May 1997-January 1999: 5,916 sheep coming from 20 farms of the programmatic area were evaluated. In the lamb category, hydatid cysts were not found. In the adults category, 62 infected animals were found (3.2%). The ovine prevalence was 1.1% and there was 100% of coincidence between diagnosis in the slaughterhouse, re-inspection in the laboratory and histopathological study. The marked decrease in the prevalence observed for sheep infection evidenced a destabilization of the biological cycle of the parasite. This could be explained by the application of a control program with uninterrupted systematic actions. Polymerase chain reaction-ribosomal ITS-1 DNA (rDNA) restriction fragment length polymorphism (PCR-RFLP) analysis and partial sequencing of the mitochondrial cytochrome c oxidase subunit 1 (CO1) gene were used to characterize E. granulosus isolates collected from different regions of Tierra del Fuego to determine which genotypes occurred in this region. The results revealed the presence of the G1 genotype (sheep-dog strain). This is the first time that a molecular analysis was performed for the E. granulosus isolates from Tierra del Fuego.

Anthelmintic resistance in cattle nematode in the western Pampeana Region of Argentina.

During the past three years there have been several reports of cattle anthelmintic resistance (AR) in Argentina. Twenty-five cattle herds in the west fattening area of the Pampeana Region were surveyed to assess the prevalence of AR and the species of nematode involved. The survey involved fattening and cow-calf systems with a mean of 6760 bovines. Resistance was determined in vivo by a faecal egg count reduction test (FECRT). Efficacies from the World Association for the Advancement of Veterinary Parasitology (WAAVP) formula were compared with those obtained from the Abbott formula. Sixty 6-12-month-old calves were selected from each property and divided into four groups, namely, IVM (ivermectin, 0.2mg/kg), BZD (fenbendazole, 5mg/kg or albendazole 7.5mg/kg), LVM (levamisole, 7.5mg/kg) and Control (untreated group). Nematode larvae were identified by four pooled faecal cultures for each group. Cooperia and Ostertagia were the predominant nematode larvae in pre-treatment cultures. The mean FECRT efficacies for IVM, BZD and LVM were 81.0+/-21%, 93.3+/-8% and 97.6+/-4%, respectively. AR was detected in 16 (64%) herds. IVM resistance was present in 15 herds and BZD resistance in eight herds (32%). Resistance to both ivermectin and fenbendazole was present in seven herds. LVM resistance was not detected. Cooperia was the predominant post-treatment nematode in all the herds with IVM resistance. In one herd with IVM resistance, Ostertagia counts were slightly increased over pretreatment counts. Ostertagia was the prevalent post-treatment genera in herds with BZD resistance. A high level of resistance to ivermectin was recorded in this fattening region of Argentina. Also several herds with resistance to benzimidazole were observed. These results show the importance of AR in Argentinian cattle production. High correlation between WAAVP and Abbott formulae was observed. However, under some uncertain diagnosis, both formulae could be applied as complementary to help result interpretations.

Toxoplasmosis in captive Bennett's wallabies (Macropus rufogriseus) in Argentina.

Wallabies and other Australian marsupials are among the most susceptible species to Toxoplasma gondii. Fatal generalized toxoplasmosis was diagnosed in two captive 3 year-old female Bennett's wallabies (Macropus rufogriseus) from Argentina (w 1 and w 2) with a history of sudden death. Both animals had internal joeys which died 2 days after their mothers. Serologically, both females and one adult male without clinical signs from the same enclosure (w 3) had antibody titers for T. gondii>or=800 by the modified agglutination test (MAT); another adult male (w 4) was negative (MAT titer<25). Microscopically, tachyzoites were observed associated to non-suppurative meningoencephalitis, hepatitis, myositis, myocarditis and severe enteritis in hematoxylin and eosin stained sections from both w 1 and w 2. Immunohistochemically, parasites in heart, brain and liver sections of both female wallabies reacted with T. gondii antiserum. T. gondii was isolated from brain tissues of w 1 and w 2 by bioassay in mice and by culture in bovine monocytes and both isolates were cryopreserved. Genomic DNA was isolated from tachyzoites grown in cultures derived from both animals. The primer pair B22/B23 specific for T. gondii produced 115bp amplicons on poliacrylamide electrophoretic gels. Stray cats were suspected as the possible source of infection. Not all infected macropods were ill, showing that the infection may be asymptomatic and is not always fatal. A vertical infection could not be proved in the joey from w 2. As far as we know, this is the first confirmed report of toxoplasmosis in Bennet's wallabies in Argentina.

Neospora caninum infections in bovine foetuses and dairy cows with abortions in Argentina.

Antibodies to Neospora caninum were measured in bovine foetuses, dairy cows and beef cows in Argentina using the IFAT, the N. caninum agglutination test, and the recombinant NCDG1 and NCDG2 ELISA. Serum antibodies (IFAT titre 1:80) were found in 20 of 82 (24.4%) dairy cow foetuses and one of 22 (4.5%) beef cow foetuses. Microscopic lesions suggestive of neosporosis were seen in brains of seven of eight foetuses with IFAT titres of 1:80. Antibodies (IFAT) were found in 122 of 189 (64.5%) dairy cows that aborted. Serum antibody titres (IFAT) of 189 dairy cows that aborted were: < 1:25 (67 cows), 1:25 (four cows), 1:50 (16 cows), 1:200 (seven cows), 1:> or = 800 (95 cows). Of the 87 sera with IFAT titres of < or = 1:50, 57 had no antibodies in 1:40 dilution and 30 had titres of 1:40 in the N. caninum agglutination test. Thus, sera from at least 56 dairy cows which had aborted were seronegative both in the N. caninum agglutination test and the IFAT. The distribution of positive and negative sera was similar when measured by ELISA, except that, depending on cut-off titre, the ELISA indicated a greater number of seropositive cows that were negative by the IFAT and N. caninum agglutination test. These results suggest that transplacental transmission of N. caninum in dairy cows in Argentina is frequent.

Seroprevalence of human herpesvirus 6 in Andino Puneños (Argentina).

We carried out a seroepidemiological survey to define the prevalence of human herpesvirus 6 (HHV6) infection in an aboriginal population (Andino Puneños) from a remote region in north-west Argentina. Antibodies against HHV6 (total IgG and the 4 subclasses of IgG) were studied in 84 serum samples (collected in 1995 and stored at -70 degrees C), using core blood mononuclear cells infected with HHV6 in an immunofluorescence assay. Of the 84 samples, 70 (83%; 95% confidence interval, 75-91%) exhibited IgG antibodies against HHV6. No significant differences in the frequency of humoral immunity were found among the 4 age-groups studied (mean ages 13, 31, 47 and 70 years) namely, 75%, 89.7%, 79.2% and 100%, respectively. HHV6-specific IgG1 was found in all the positive serum samples tested but none of them contained specific IgG2, IgG3 and IgG4. These results confirmed a high rate of infection with HHV6 within this aboriginal group in Argentina and an IgG1 anti-HHV6 activity compatible with a maintenance of immunity.

Epidemiological and clinical aspects of scorpionism by Tityus trivittatus in Argentina.

This is a descriptive study of epidemiological and clinical aspects of stings caused by the scorpion Tityus trivittatus in Argentina. We analyzed 511 cases recorded from different health centers in 22 provinces. Most accidents took place during the period November-April (76%), in or nearby houses (86%). Over 50% of the accidents involved children and teenagers. Envenomation by T. trivittatus was mainly characterized by local symptoms: pain (85%), edema (26.6%), burning sensation (24.7%), erythema (20.7%), local hyperthermia (13.1%), paresthesia (9.8%) and general manifestations such as vomiting (25%), paleness (18.8%), headache (11.4%) and sweating (8.2%). Neurological, cardiovascular and respiratory disorders were uncommon. Almost 90% of the injured people got treated with specific antivenom within 2 h, 6% were treated 2-h after the accident and only 2 people were treated 12 or more hours after being stung. The global mortality recorded was 6 per 1000 cases. Scorpionism in Argentina is a public health problem under control due to the relatively low incidence and the accessibility of specific antivenom.

Polychlorinated biphenyls in different trophic levels from a shallow lake in Argentina.

Polychlorinated biphenyls (PCBs) were determined in a aquatic community from Los Padres Lake, Argentina. Twenty four PCB congeners from tri- to octa-chlorinated isomers were detected and quantified using conventional gas chromatography with electron capture detector (GC-ECD). The aim of this study was to investigate the concentrations of PCBs in freshwater organisms from a shallow lake of Argentina. Stems of bulrush (Schoenoplectus californicus), whole tissues of false loosestrife (Ludwigia sp.) and grass shrimp (Palaemonetes argentinus), and liver, gonads, muscle and mesenteric fat (if present) of fish species (Rhamdia sapo) and (Oligosarcus jenynsi) were analyzed. Two areas were selected to macrophytes sampling: the input area, main PCB source of the lake (Station 1), and the output area, a potential anoxic zone (Station 2). Macrophytes from Station I bioconcentrated higher total PCB levels than Station 2, showing that the former have received PCBs washed down from upstream areas. Penta- and hexa-congeners were enriched relative to other congeners in animal biota and macrophytes from Station 1, consistent with commercial mixture of Aroclor 1254 used in this region. In bulrush from Station 2 a predominance of tri- and tetra-chlorinated congeners was observed. Grass shrimp showed the lowest PCB values among animal biota. PCB concentrations in fish tissues varied with the species and the gonadal development. Mesenteric fat, only present in post-spawning organisms of R. sapo, had the highest values of PCBs relative to other tissues. A clearance of total PCBs in ovaries of post-spawning females of R. sapo was observed, but not in testes. O. jenynsil P. argentinus biomagnification factor (BMF) had a mean value of 18.7. Congeners 44, 52 and 151, showed the highest BMF values, being 64, 66 and 62, respectively. These values would be a consequence of the low depuration rate of 44 and 52 congeners with orthochlorine substitution conducted by O. jenynsi and the high depuration rate of congener 151, which lacks 4 4'- chlorine substitution, carried out by grass shrimp. Although the most of congeners have been biomagnified, they did not clearly displayed a concomitantly increasing with log K(ow).

Analysis of the psychometric properties of the Spanish version of the Beck Depression Inventory in Argentina.

Increased international collaboration in clinical trials has created a need for cross-culturally valid instruments to assess quality of life and behavioral disorders. Cross-cultural studies of depressive symptomatology, in particular, must be preceded by an exhaustive study of the psychometric properties of the instruments, to ensure the validity of the comparison. In this article, we examine the validity, reliability and factor structure of the Spanish version of the Beck Depression Inventory (BDI) in a random stratified sample of 608 community residents in Argentina. Our findings indicate that this version of the BDI is psychometrically strong and appropriate for use in Argentina, and we suggest that it is suitable for assessing depressive symptomatology in Spanish speakers.

A highly prevalent polymorphism at codon 72 of HIV-1 reverse transcriptase in Argentina prevents hybridization reaction at codon 74 in the LiPA genotyping test.

The aim of this study was to compare the performance of two commercial drug-resistance HIV-1genotyping kits: LiPA (Innogenetics, Belgium) and TruGene (Visible Genetics, Canada). Samples from 103 HIV-1 infected individuals from Argentina were studied. The average correlation between the two methods was 92.81%. More codons could be analysed by TruGene than by LiPA (610/618 and 541/618 codons, respectively). The sequences of the samples and the LiPA probes were aligned showing that a silent mutation at codon 72 (AGA-->AGG) of the HIV-1 reverse transcriptase, which was not recognised by the LiPA probes, was present in 35/36 non-reactive samples for position 74. The overall prevalence of this polymorphism in the population studied was 39.81%. When sequences from different parts of the world were analysed 189/3395 (5.63%) carried the mutation at codon 72, while 23/133 (17.29%) of the sequences from Latin America (excluding Argentina) had this mutation. In both cases, the prevalence of this polymorphism in the Argentinean population was significantly higher. This highlights the importance of carrying out studies on the performance of genotyping kits outside the United States and Europe, especially in areas where non-B HIV-1 subtypes are prevalent.

Variability of aflatoxin and cyclopiazonic acid production by Aspergillus section flavi from different substrates in Argentina.

Aspergillus section flavi strains isolated from peanuts, wheat and soybean grown in Argentina were screened for aflatoxins (type B and G) and cyclopiazonic acid (CPA) production. Aspergillus flavus was the predominant species in all substrates, although there was almost the same proportion of A. flavus and Aspergillus parasiticus in peanuts. Aspergillus nomius was not found. Incidence of aflatoxigenic A. flavus strains was higher in peanuts (69%) than in wheat (13%) or soybeans (5%) while the ratio of CPA producers A. flavus isolated from all substrates was very high (94% in peanuts, 93% in wheat and 73% in soybeans). Isolates of A. flavus able to produce simultaneously aflatoxins type B and CPA were detected in all substrates, suggesting the possibility of co-occurrence of these toxins. Almost all isolates of A. parasiticus resulted aflatoxins (type B and G) producers but did not produce CPA. Five of sixty-seven strains isolated from peanuts showed an unusual pattern of mycotoxin production (aflatoxins type B and G simultaneously with CPA). These strains also produced numerous small sclerotia like S strains of A. flavus detected in cottonseed in Arizona and in soils of Thailand and West Africa. The atypical strains are not widely distributed in Argentina and were found uniquely in peanuts.

The diversity of rhizobia nodulating beans in Northwest Argentina as a source of more efficient inoculant strains.

The common bean (Phaseolus vulgaris L.) is cultivated widely in Central and South America and particularly in the Northwest of Argentina. In order to describe the diversity of the common bean nodulating rhizobial population from the bean producing area in Northwest Argentina (NWA), a collection of about 400 isolates of common beans recovered from nodules and soil samples from NWA were characterized by using nifH-PCR, analysis of genes coding for 16S rRNA and nodC, and REP-fingerprinting, respectively. It was found that species Rhizobium etli is predominant in common bean nodules although a high degree of diversity was found within the species. Other bean nodulating genotypes recovered from soils by using Leucaena sp. as the trapping host was found to have the 16S rDNA alleles of species such as Sinorhizobium fredii, Sinorhizobium saheli, Sinorhizobium teranga, Mesorhizobium loti, and Rhizobium tropici. Some of the bean genotypes that were found to be more efficient in green house experiments were selected and assayed in two successive bean-cropping seasons in the field environment in NWA, and an increase in yields with inoculation was found. The performance of strains isolated from the region indicates potential for exploiting the diversity.

Genetic characterization and phylogeny of Andes virus and variants from Argentina and Chile.

Andes virus, one of five hantaviruses known to cause hantavirus pulmonary syndrome (HPS), emerged in 1995 in southwestern Argentina (López et al. (1996) Virology 220, 223-226). The complete nucleotide sequence of Andes virus S genome segment was determined and compared with sequences of viral RNAs in autopsy tissues of more recently reported HPS cases from southwestern Argentina and south of Chile (cases ESQ H-1/96 and CH H-1/96). Andes virus S segment was found to be 1876 nucleotides in length and to encode the nucleocapsid protein (N), 428 amino acids in length. S segment analysis also revealed a long 5' non-coding region (547 nucleotides) which displays three copies of an octanucleotide sequence repeat. Comparisons of S segment sequences of ESQ H-1/96 and CH H-1/96 (82% of the entire genome sequence) with the corresponding sequences of Andes virus revealed identities of 97.2% and 98.5%, respectively. Sequence motifs identical and in the same positions as exhibited in Andes virus 5' non-coding region were found in both, ESQ H-1/96 and CH H-1/96 sequences. Three genome fragments of the M segment sequence of the viruses (representing approximately 34% of the entire sequence) were also analyzed. Comparisons of S and M segment sequences of Andes virus with the corresponding sequences of ESQ H-1/96 showed S and M segment identities which differ by less than 1.4%. Andes virus and CH H-1/96 have S segments that differ by 1.5% from one another while their M segment fragments differ by 5.5-8.2%. Phylogenetic analysis showed that Andes virus along with ESQ H-1/96 and CH H-1/96 form a distinct lineage within the clade containing Bayou and Black Creek Canal viruses. It also showed that Andes virus branch of trees derived from comparisons of S or M sequences differed. It is concluded that Andes virus variants causing HPS circulate east and west of the Andes mountains.

Genetic diversity of GBV-C/HGV strains among HIV infected-IVDU and blood donors from Buenos Aires, Argentina.

GBV-C/HGV RNA was investigated in serum samples from 70 HIV(+) intravenous drug users (IVDU), as well as from 200 blood donors from Buenos Aires, Argentina. Viral RNA was demonstrated in 21 IVDU by reverse transcription-nested PCR of the 5' UTR. c-DNA amplified products were analyzed and their sequences compared with those downloaded from GenBank. A phylogenetic tree based on 171 sequences demonstrated the presence of three major genogroups, including two subgroups, within local samples, i.e. group 1 (n=1), 2a (n=11), 2b (n=4) and 3 (n=5). These results agreed entirely with those obtained by a novel RFLP (J. Clin. Microbiol. 37, 1340-1347, 1999) of the same 5' UTR amplicons. As expected, GBV-C/HGV RNA prevalence was significantly higher among IVDU than among blood donors (P<0.0001), although within the latter group an unexpectedly high rate was also detected, since 11 of 200 sera (5.5%) proved positive. These viral isolates were ascribed either to subgroup 2a (n=5), subgroup 2b (n=5) or genogroup 3 (n=1). Briefly, this partial view of GBV-C/HGV molecular epidemiology in Argentina shows: (i) different rates of GBV-C/HGV infection within both IVDU and blood donors; (ii) a high prevalence of viral RNA among blood donors; and (iii) a predominant circulation of genogroup 2, with minor contribution of groups 3 and 1.

Are market-oriented health insurance reforms possible in Latin America? The cases of Argentina, Chile and Colombia.

The process of health care reform benefits tremendously from comparing characteristics and performance across nations. This paper studies market-oriented health insurance reforms in three Latin American countries: Argentina, Chile and Colombia. Chile allowed private health insurers to compete for workers payroll contributions in the 1980s, permitting the modernization of the private health sector but relatively impoverishing the public health sector as a consequence of selection practices by private carriers. In the 1990s, Argentina and Colombia started liberalizing the health insurance sector but using policies to avoid the adverse effects encountered in the Chilean experience. These policies are scrutinized while challenges for these and future health insurance reform processes are discussed.
