The Sakihara-type deep vascular ligation appratus was first devised in 1935 and has been used for many years by local Japanese surgeons. This apparatus is simple, easy to use, and can be applied in various surgical subspecialities. The structure and use of this apparatus is described in detail.
Several phosphorylation, annealing and ligation reaction conditions have been investigated for the enzymatic assembly of genes of interest. The use of longer oligodeoxyribonucleotides (40-60 bases long) also improved the enzymatic reaction. Furthermore, the use of longer oligonucleotides and the elimination of long stretches of G's or C's lowered the mutation rate.
A new instrument is described which holds, carries and ties ligatures in areas where access is limited or vision obscured during traditional hand tying. It has been used during deep pelvic surgery, for cholecystectomy, in highly selective vagotomy and may be useful in peraxillary transthoracic cervical sympathectomy.
