Indirect bonding.

Abstract

The advantages of indirect bonding have been recognised for more than twenty years. However the technique has not gained widespread acceptance as an alternative to conventional direct bonding. Reports of difficulty in achieving adequate isolation of posterior teeth and problems with resin flash and low bond strength may explain the reluctance of the profession to make use of the technique. This paper describes an improved indirect technique which uses a heat cured bonding resin in a way that appears to overcome the reported problems.

A pH-dependent colon targeted oral drug delivery system using methacrylic acid copolymers. I. Manipulation Of drug release using Eudragit L100-55 and Eudragit S100 combinations.

Lactose-based placebo tablets were coated using various combinations of two methacrylic acid copolymers, Eudragit L100-55 and Eudragit S100, by spraying from aqueous systems. The Eudragit L100-55-Eudragit S100 combinations (w/w) studied were 1:0, 4:1, 3:2, 1:1, 2:3, 1:4, 1:5 and 0:1. The coated tablets were tested in vitro for their suitability for pH dependent colon targeted oral drug delivery. The same coating formulations were then applied on tablets containing mesalazine as a model drug and evaluated for in vitro dissolution rates under various conditions. The disintegration data obtained from the placebo tablets demonstrate that disintegration rate of the studied tablets is dependent on: (i) the polymer combination used to coat the tablets, (ii) pH of the disintegration media, and (iii) the coating level of the tablets. Dissolution studies performed on the mesalazine tablets further confirmed that the release profiles of the drug could be manipulated by changing the Eudragit L100-55 and Eudragit S100 ratios within the pH range of 5.5 to 7.0 in which the individual polymers are soluble respectively, and a coating formulation consisting of a combination of the two copolymers can overcome the issue of high gastrointestinal (GI) pH variability among individuals. The results also demonstrated that a combination of Eudragit L100-55 and Eudragit S100 can be successfully used from aqueous system to coat tablets for colon targeted delivery of drugs and the formulation can be adjusted to deliver drug at any other desirable site of the intestinal region of the GI tract on the basis of pH-variability. For colon targeted delivery of drugs the proposed combination system is superior to tablets coated with either Eudragit L100-55 or Eudragit S100 alone.

A novel phosphatidylglycerol-selective phospholipase A2 from macrophages.

In our recent studies on the synthesis of bis(monoacylglycero)phosphate (BMP), we postulated that the first step involved a PLA2 that cleaved the 2-acyl group from phosphatidylglycerol (PG). In the present study, a novel lysosomal PLA2 was partially purified and characterized from RAW 264.7, macrophage like cells. Cells were homogenized and delipidated, and the PLA2 activity in the soluble fraction was purified by Sephacryl S100 and DEAE Sephacel. Further purification was performed using Con-A Sepharose, Phenyl Sepharose, DEAE Sephacel, and Superdex 75 FPLC. The enzyme at this stage of purification showed a dominant band around 45 kDa plus several minor bands on SDS-PAGE. The molecular mass determined by Superdex 75 column FPLC was about 45 kDa. The highly purified fraction hydrolyzed at the sn-1 position, implying that this PLA2 also has some intrinsic PLA1 activity. This enzyme preferentially hydrolyzed PG, has an acidic pH optima, and does not require divalent metal ions. Comparison using PG with various acyl chains on the sn-2 position showed that oleate and linoleate were preferred relative to arachidonate. MAFP, a known cytosolic PLA2 inhibitor, strongly inhibited this PLA2 activity. MJ33, AACOCF3, DENP, and Amiodarone also gave moderate inhibition. The characteristics of this enzyme showed this to be a new type of PLA, and the overwhelming preference for PG as substrate suggests its physiological role is in the biosynthesis of BMP.

Enhancement of enzyme reaction of magnetically anisotropic polyacrylamide gel rods immobilized with ferromagnetic powder and beta-D-galactosidase in an alternating magnetic field.

An immobilized polyacrylamide gel containing beta-D-galactosidase and Sr-Ba-ferrite was magnetized in a static magnetic field. The gel rods (10 mm long, O 2 mm) exhibiting magnetic anisotropy could move at lower than 100 Hz but not at higher than 250 Hz in an alternating magnetic field of 200 Oe. In case of immovability of gel rods, the apparent enzymic activity increased 3 times higher under exposure of an alternating magnetic field of 500 Oe (570 Hz). It could be explained that the ferromagnetic powder inside the gel might vibrate under the influence of elasticity of gel in the alternating magnetic field of 100 or 500 Oe and 0.2-12 kHz. This might facilitate faster diffusion of the substance inside the gel and transportation of the substrate and the product through the surface of gel. Consequently, the enzyme reaction was apparently activated.
