Lead poisoning.

Abstract

While lead is a very old poison whose toxicity was known to the ancient Greeks, new information about mechanisms and effects of lead toxicity is continually being discovered. Family practitioners should be alert to the possibility of lead poisoning and the situations in which it occurs. Symptoms may be non specific and careful questioning of patients or their parents may be required to identify possible exposure.

Immunological analyses of human papillomavirus capsids.

Recombinant human papillomavirus (HPV) virus-like particles (VLPs) are promising vaccine candidates for controlling anogenital HPV disease. Questions remain, however, concerning the extent of capsid antigenic similarity between closely related virus genotypes. To investigate this issue, we produced VLPs and corresponding polyclonal immune sera from several anogenital HPV types, and examined these reagents in enzyme-linked immunosorbent assays (ELISAs) and in cross-neutralization studies. Despite varying degrees of L1 genetic sequence relatedness, VLPs of each type examined induced high-titer serum polyclonal antibody responses that were entirely genotype-specific. In an in vitro infectivity assay, only cognate VLP antisera were able to neutralize pseudovirions of HPV-16, HPV-18 and HPV-33, with two exceptions: HPV-31 and HPV-45 VLP post-immune sera demonstrated low levels of neutralizing activity against pseudovirions of HPV-33 and HPV-18, respectively. In other experiments, epitopes shared between closely related types were found to be less immunogenic than, and antigenically distinct from, primary type-specific B-cell determinants of the viral capsid. In addition, results from epitope blocking experiments suggested a close correlation between primary type-specific capsid antigenic sites and virion neutralization. These findings support the view that papillomavirus genotypes denote unique viral serotypes, and suggest that a successful vaccine for these viruses will likely require the inclusion of VLPs of each serotype for which protection is desired.

In situ nasal absorption of midazolam in rats.

Intranasal (i.n.) midazolam (MDZ) administrations may be used successfully for preoperative sedation, especially in young patients. However, clinicians have to use the commercial parenteral formulation, the low pH of which (3.3), necessary to solubilize MDZ (pK(a) 6.1), is probably responsible for the signs of local irritation frequently reported. As a starting point to design a formulation suitable for the nasal route, MDZ nasal absorption was investigated in rats. The effects of the MDZ solution concentration (10--100 microg/ml), osmolality (from less than 10 mOsm/kg up to 450 mOsm/kg) and pH (3.3--7.4) were studied using an in situ perfusion technique. MDZ was determined by reversed-phase HPLC in the circulating solution and results were expressed in clearance terms. MDZ absorption was independent of its concentration. The pH of the solutions was the key-parameter and only a pH above 4 allowed significant absorption. These results were consistent with a passive diffusion absorption of MDZ and partly followed the pH partition theory. In conclusion, satisfactory MDZ absorption should be expected with a formulation at a pH suitable for the nasal route in human (5.5--6.5).

Sorption and desorption studies on chitin gels.

The aim of this work was to study various transport phenomena in chitin gels obtained by N-acetylation of chitosan in a water-alcohol mixture. Three kinds of transport were investigated: the sorption of solutes interacting with chitin, the desorption of solutes without significant interaction with the polymer, and osmosis phenomena. In the case of interactive sorption, dyes having different chemical structures such as C.I. Acid Blue 74, C.I. Reactive Violet 5 or C.I. Direct Red 28 were tested. Sorptions of C.I. Acid Blue 74 and C.I. Reactive Violet 5 depend on the charge density of the polymer network and, as a consequence, on DA, pH and the dielectric constant of the media. This result reveals the importance of electrostatic interactions. On the other hand, the sorption of C.I. Direct Red 28 is mainly due to hydrophobic interactions and H-bonding, it is limited to the extreme surface of the gel. Concerning the non-interactive desorption, solutes of different steric hindrance such as PP vitamin, B1 vitamin and caffeine exhibit similar diffusion coefficients located within 3.7-5.6x10(-6) cm(2) s(-1). Finally, the osmotic behaviour of the gel immersed in a concentrated solution of gelatin allows us to multiply by 25 the concentration of chitin in the gel without any penetration of gelatin.
