Lymphography.

Abstract

Lymphography is a valuable diagnostic procedure that may be declining in use due to the amount of time required for a radiologist to perform the procedure. Technologist participation in lymphography may be a means to allow better utilization of the radiologist's time, while improving diagnostic accuracy and reducing health care costs. This paper presents an overview of lymphography and details the technologist's role in the procedure.

Direct dye binding--a quantitative assay for solid-phase immobilized protein.

A direct dye-binding procedure was established for the quantification of protein after its immobilization on a solid phase, using IgG and BSA as model proteins. The assay, which in the range 0-5 mg protein/ml gel correlates well with indirect protein determination by A280 as well as determination of protein hydrolyzed from the gel, is based on a modified Bradford dye-binding assay. As the protein coupled to the gel binds the dye, a decrease in A465 of the supernatant is measured. Three solid supports commonly used for protein immobilization (Sepharose, Sephadex, Sephacryl) were found to be compatible with the dye-binding assay while nonspecific dye binding was found to HEMA gels. Protein was coupled to Sephacryl S-1000 using three different activation methods (aldehyde, hydrazine, and adipic acid dihydrazide). Artifactual dye-binding was not observed using any of the three different "linkers." The assay is easily carried out and represents a useful tool, e.g., when optimizing procedures for protein immobilization.

Photoelectrochemistry of dyes in phospholipid liposome.

The systems consisting of phenosafranin or thionine dye in the presence of phospholipid liposome in aqueous solution generate photovoltage when studied in a photoelectrochemical cell. A possible mechanism of photovoltage generation suggests the photoinduced electron transfer from phospholipid to dye in liposome through charge-transfer (CT) interaction. In these dye-phospholipid systems, a good correlation between the photovoltage (Voc) determined by photoelectrochemical studies and the equilibrium constant (Kc) determined by spectrophotometric studies, confirms the mechanism.

Transverse agarose pore gradient gel electrophoresis of DNA.

Transverse agarose pore gradient gels were prepared on GelBond in the concentration range of nominally 0.2-1.5% SeaKem GTG agarose, using density stabilization by glycerol and incorporation of a dye to define the gel concentration at each point on the pore gradient gel. The distribution of the dye was evaluated by photography, video-acquisition and digitization of the gradient mixture and by densitometry of the gel. The gel was applied to the electrophoresis of a 1-kb standard ladder of DNA fragments, using standard submarine apparatus. The method extends to agarose gel electrophoresis the benefits of semi-automated analysis of 'Ferguson curves' described in application to polyacrylamide gel by Wheeler et al. (J. Biochem. Biophys. Methods 24, 171-180).
