Assay of trace D-amino acids in neural tissue samples by capillary liquid chromatography/tandem mass spectrometry.

A sensitive chiral capillary HPLC-MS/MS method well suited for the determination of amino acid enantiomers in biological samples was developed. The method involved precolumn derivatization of the sample with 7-fluoro-4-nitrobenzoxadiazole (NBD-F). After derivatization, NBD-amino acids were stacked on a C18 reversed-phase extraction microcolumn, thus enriching and cleaning up the analytes. Various chiral stationary phases (CSPs) including cyclodextrin-bonded silica, Pirkle-type, vancomycin, and teicoplanin-bonded silica particles were evaluated for resolving NBD-F tagged amino acid enantiomers with mobile phases compatible with MS detection. It was found that only teicoplanin aglycon CSP provided sufficient resolution of NBD-Asp and NBD-Ser enantiomers to quantify trace levels of D-Asp and D-Ser in tissue samples. MS/MS detection of NBD-amino acid derivatives was very sensitive and selective. The high selectivity allowed the use of a stable isotope-labeled analyte analogue (i.e., L-aspartic acid-2,3,3-d3) as internal standard for the quantitation to improve assay reproducibility and reliability. Neural tissue samples dissected from rat brain and the central nervous system (CNS) of Aplysia californica, a widely used neuronal model, were analyzed to determine the chirality of glutamic acid (Glu), aspartic acid (Asp), and serine (Ser). The former two are major excitatory amino acids in the brain, and the last one has been recently identified as a neuromodulator. Both D-Ser and D-Asp were detected in rat brain. While the D-Asp level decreased rapidly through the developmental stages of the rat, the D-Ser level increased steadily from 82.3 microg/g of wet tissue in 3-day prenatal rats to 241.3 microg/g of wet tissue in 90-day-old rats. Interestingly, no D-Ser was detected in the CNS of Aplysia, a "primitive" invertebrate. However, the D-Asp level in this animal was found to be high. In a particular connective nerve sample, D-Asp was at 323.2 microg/g of wet tissue and constituted 60.2% of total Asp. D-Glu was not detected either in rat brain or in Aplysia's CNS.

Altered cholesterol homeostasis in cultured and in vivo models of cystic fibrosis.

Determining how the regulation of cellular processes is impacted in cystic fibrosis (CF) is fundamental to understanding disease pathology and to identifying new therapeutic targets. In this study, unesterified cholesterol accumulation is observed in lung and trachea sections obtained from CF patients compared with non-CF tissues, suggesting an inherent flaw in cholesterol processing. An alternate staining method utilizing a fluorescent cholesterol probe also indicates improper lysosomal storage of cholesterol in CF cells. Excess cholesterol is also manifested by a significant increase in plasma membrane cholesterol content in both cultured CF cells and in nasal tissue excised from cftr(-/-) mice. Impaired intracellular cholesterol movement is predicted to stimulate cholesterol synthesis, a hypothesis supported by the observation of increased de novo cholesterol synthesis in lung and liver of cftr(-/-) mice compared with controls. Furthermore, pharmacological inhibition of cholesterol transport is sufficient to cause CF-like elevation in cytokine production in wild-type cells in response to bacterial challenge but has no effect in CF cells. These data demonstrate via multiple methods in both cultured and in vivo models that cellular cholesterol homeostasis is inherently altered in CF. This perturbation of cholesterol homeostasis represents a potentially important process in CF pathogenesis.

Utility of NBD-Cl for the spectrophotometric determination of some skeletal muscle relaxant and antihistaminic drugs.

A simple, accurate, precise and sensitive colorimetric method for the determination of some skeletal muscle relaxant drugs, namely orphenadrine citrate (I), baclofen (II), antihistaminic drugs as acrivastine (III) and fexofenadine hydrochloride (IV) is described. This method is based on the formation of charge transfer complex with 4-chloro-7-nitro-2,1,3-benzoxadiazole (NBD-Cl) in non-aqueous medium. The orange color products were measured at 472, 465, 475 and 469 nm for drugs I, II, III and IV, respectively. The optimization of various experimental conditions was described. Beer's Law was obeyed in the range (2.5-17.5), (5-70), (2.5-25) and (10-50)microg/ml for drugs I, II, III and IV, respectively. The molar absorptivity (epsilon), sandell sensitivity, detection((LOD)) and quantitation limits((LOQ)) are calculated. The procedure was favorably applied for determination of certain pharmaceutical dosage forms containing the studied drugs. The obtained results were compared with the official and reported methods. There were no significant differences between proposed, reported and the official methods.
