Sugar transport in Coprinus cinereus.

Abstract

Two transport systems for glucose were detected: a high affinity system with a Km of 27 muM, and a low affinity system with a Km of 3.3 mM. The high affinity system transported glucose, 2-deoxy-D-glucose (Km = 26 muM), 3-O-methylglucose (Km = 19 muM), D-glucosamine (Km = 652 muM), D-fructose (Km = 2.3 mM) and L-sorbose (Km = 2.2 mM). All sugars were accumulated against concentration gradients. The high affinity system was strongly or completely inhibited by N-ethylmaleimide, quercetin, 2,4-dinitrophenol and sodium azide. The system had a distinct pH optimum (7.4) and optimum temperature (45 degrees C). The low affinity system transported glucose, 2-deoxy-D-glucose (Km = 7.5 mM), and 3-O-methylglucose (Km = 1.5 mM). Accumulation again occurred against a concentration gradient. The low affinity system was inhibited by N-ethylmaleimide, quercetin and 2,4-dinitrophenol, but not by sodium azide. The rate of uptake by the low affinity system was constant over a wide temperature range (30--50 degrees C) and was not much affected by pH; but as the pH of the medium was altered from 4.5 to 8.9 a co-ordinated increase in affinity for 2-deoxy-D-glucose (from 52.1 mM to 0.3 mM) and decrease in maximum velocity (by a factor of five) occurred. Both uptake systems were present insporelings germinated in media containing sodium acetate as sole carbon source. Only the low affinity system could initially be demonstrated in glucose-grown tissue, although the high affinity system was restored by starvation inglucose-free medium. The half-ti me for restoration of high affinity activity was 3.5 min and the process was unaffected by cycloheximide. Addition of glucose to an acetate-grown culture inactivated the high affinity system with a half-life of 5--7.5 s. Addition of cycloheximide to an acetate-grown culture caused decay of the high affinity system with a half-life of 80 min. Regulation is thus thought to depend on modulation of protein activity rather than synthesis, and the kinetics of glucose, 2-deoxy-D-glucose and 3-O-methylglucose uptake would be consistent with there being a single carrier showing negative co-operativity. Analysis of transport defective mutants revealed defects in both transport systems although the mutants used were alleles of a single gene. It is concluded that this gene (the ftr cistron) is the structural gene for an allosteric molecule which serves both transport systems.

Comparison of two continuous fungal bioreactors for posttreatment of anaerobically pretreated weak black liquor from kraft pulp mills.

The purpose of this work was to evaluate and compare two continuous systems of posttreatment of anaerobically pretreated weak black liquor (WBL). The first system consisted of a packed bed reactor (PBR) with Trametes versicolor (Tv) immobilized on wood cubes of holm oak (biocubes). The second system was a fluidized bed reactor (FBR) with Lentinus edodes (Le) immobilized on wood cubes of holm oak. The reactors operated for 65 days at a hydraulic retention time (HRT) of 5 days, at 28 degrees C, with continuous aeration. Response variables monitored were conventional and specific, unit, net removal efficiency (eta and eta(sun), respectively) of chemical oxygen demand (COD), color, and ligninoids, and enzymatic activities of manganese peroxidase (MnP), lignin peroxidase (LiP), laccase (Lac) and proteases. The PBR showed an average color eta superior to that of the FBR (52.42 +/- 21.78% and 25.34 +/- 14.38% for PBR and FBR, respectively); removals of COD and ligninoids presented a similar pattern to that of color. Lac activity was significantly larger in PBR than in FBR. Activity of MnP in PBR was higher than that of the FBR (0.004 and 0.002 U MnP/mL, respectively). This difference could be ascribed to the different fungi present in each bioreactor. LiP activity was very low in both reactors. Average value of proteases was almost double in the FBR as compared with PBR (0.472 and 0.209 U Proteases/mL, respectively). During the last 2 weeks of operation, biocubes in the FBR experienced a significant loss of the attached Le biomass, probably by attrition. This and higher protease activity in the FBR could explain the lower pollutant removals achieved in the FBR. Overall, PBR with immobilized Tv showed a better performance than the FBR with Le for the posttreatment of the recalcitrant anaerobic effluent. Extended and sustained pollutant removal (65 days) was achieved in the PBR, although more research is needed to evaluate bioreactor performance at shorter hydraulic retention times.

Post TLC developing technique for tyrosinase inhibitor detection.

Post TLC developing technique was developed to detect substances which can inhibit tyrosinase activity. The method involved spraying the TLC plate or chromatographic paper containing sample spot(s) with tyrosinase and l-tyrosine solutions successively. A positive result could be visualized directly as white spot(s) against a brownish-purple background. The method can either be used as a quick screening method for tyrosinase inhibitor detection or a guiding procedure for an isolation of tyrosinase inhibitors from mixtures or natural product extracts. The technique is sensitive enough to give a clear result in the presence of only 6 ng glabridin.

Enzymatic synthesis of arbutin undecylenic acid ester and its inhibitory effect on mushroom tyrosinase.

A novel tyrosinase inhibitor, an arbutin derivative having undecylenic acid at the 6-position of its glucose moiety, was enzymatically synthesized. Its inhibitory activity was studied in vitro by using catechol and phenol as substrates. The IC(50) value of the arbutin ester on tyrosinase using catechol (4 x 10(-4) M) was 1% of that when arbutin (4 x 10(-2) M) was used. Using phenol, IC(50) of the arbutin ester (3 x 10(-4) M) as substrate was 10% of that of arbutin (3 x 10(-3) M). These results suggest that the arbutin ester inhibits the latter part of the tyrosinase reaction, which consists of hydroxylation and oxidation.
