A two-component tetrachlorohydroquinone reductive dehalogenase system from the lignin-degrading basidiomycete Phanerochaete chrysosporium.

Tetrachloro-1,4-hydroquinone (TClHQ) is an intermediate in the degradation of pentachlorophenol by the lignin-degrading basidiomycete Phanerochaete chrysosporium. Two enzymes required for the reductive dehalogenation of TClHQ to trichlorohydroquinone (TrClHQ) were identified in cell-free extracts of P. chrysosporium. In the presence of GSH, a membrane-bound enzyme converted TClHQ to the glutathionyl conjugate of TrClHQ (GS-TrClHQ). This membrane-bound glutathione transferase was specific for GSH as a cosubstrate. In the second step of the reductive dehalogenation reaction, a soluble enzyme fraction converted GS-TrClHQ to TrClHQ in the presence of GSH, cysteine, or dithiothreitol. Thus, this second enzyme appears to be a GS-conjugate reductase. These two enzyme fractions, working in tandem, also reductively dehalogenated TrClHQ and 2,6-dichlorohydroquinone, which are intermediates in the degradation of chlorophenols by this organism.

Location of translational initiation factor IF3 on the small ribosomal subunit.

The location of translational initiation factor IF3 bound to the 30S subunit of the Thermus thermophilus ribosome has been determined by cryoelectron microscopy. Both the 30S.IF3 complex and control 30S subunit structures were determined to 27-A resolution. The difference map calculated from the two reconstructions reveals three prominent lobes of positive density. The previously solved crystal structure of IF3 fits very well into two of these lobes, whereas the third lobe probably arises from conformational changes induced in the 30S subunit as a result of IF3 binding. Our placement of IF3 on the 30S subunit allows an understanding in structural terms of the biochemical functions of this initiation factor, namely its ability to dissociate 70S ribosomes into 30S and 50S subunits and the preferential selection of initiator tRNA by IF3 during initiation.

Synthesis of mycolic acids of mycobacteria: an assessment of the cell-free system in light of the whole genome.

Mycolic acids are 70-90 carbon, alpha-alkyl, beta-hydroxy fatty acids constituting a major component of the cell envelope of Mycobacterium tuberculosis. The fact that the mycolic acid biosynthetic pathway is both essential in mycobacteria and the target for many first-line anti-TB drugs necessitates a detailed understanding of its biochemistry. A whole cell-free, but cell particulate- and membrane-containing enzyme preparation for mycolic acid biosynthesis was developed a few years ago and studied extensively. This system was shown to catalyze the synthesis of mature mycolic acids from [14C]acetate, but allows only minimal deposition into the cell wall proper. In the meantime the sequence of the entire genome of M. tuberculosis has been elucidated and its analysis using numerous protein sequence-based algorithms predicted cytoplasmic localization and a soluble, not a particulate, nature for the enzymes involved in the mycolic acid synthetic pathway. Accordingly, we re-assessed the 'cell-free' system for mycolic acid synthesis and concluded that it is probably due to the presence of unbroken cells, since viable cells were recovered from the cell wall preparation. The amount of whole cells depended upon the efficiency of the cell disruption method and conditions, and the amount of mycolic acid synthesized by the putative cell-free system correlated with the content of whole cells. Thus, accumulated results from the use of this 'cell-free' cell wall-based system should be re-evaluated in the light of these new data.
