Metabolism of L-phenylalanine and L-tyrosine by the phototrophic bacterium Rhodobacter capsulatus.

The phototrophic bacterium Rhodobacter capsulatus utilizes the aromatic amino acids L-phenylalanine and L-tyrosine as nitrogen source. L-Phenylalanine is hydroxylated to L-tyrosine, which is further converted into p-hydroxyphenyl pyruvate (pHPP) by a transamination reaction. The bacterium is unable to grow at the expense of these amino acids as the sole carbon source, although it is able to degrade them to homogentisate, probably by unspecific hydroxylation reactions. Metabolization of L-phenylalanine or L-tyrosine as nitrogen source requires phototrophic growth conditions and does not produce free ammonium inside the cells. A low aminotransferase activity with 2-oxoglutarate and L-tyrosine as substrates can be detected in crude extracts of R. capsulatus. Uptake of both amino acids by R. capsulatus was completely inhibited by ammonium addition, which also prevents aminotransferase induction.

The inactivation mechanism of low molecular weight phosphotyrosine-protein phosphatase by H2O2.

Low molecular weight phosphotyrosine-protein phosphatase (LMW-PTP) shares no general sequence homology with other PTPs, although it has an active site sequence motif CXXXXXR and a reaction mechanism identical to those of all PTPs. The main function of this enzyme is the down-regulation of platelet-derived growth factor and insulin receptors. Both human LMW-PTP isoenzymes are inactivated by H2O2. The enzymes are protected from inactivation by Pi, a competitive inhibitor, suggesting that the H2O2 reaction is directed to active site. Analysis of free thiols performed on the inactivated enzymes demonstrates that only two out of the eight LMW-PTP cysteines are modified. Time-course high performance liquid chromatography-electrospray mass spectrometry, together with specific radiolabeling and tryptic fingerprint analyses, enables us to demonstrate that H2O2 causes the oxidation of Cys-12 and Cys-17 to form a disulfide bond. Because both residues are localized into the active site region, this modification inactivates the enzyme. Fluorescence spectroscopy experiments suggest that the fold of the enzyme is modified during oxidation by H2O2. Because a physiological concentration of H2O2 produces enzyme inactivation and considering that the activity is restored by reduction with low molecular weight thiols, we suggest that oxidative stress conditions and other processes producing hydrogen peroxide regulate the LMW-PTP in the cell.

Reductive assays for S-nitrosothiols: implications for measurements in biological systems.

Bioactive SNOs are found in many tissues. We speculated SNOs might be misidentified in conventional assays which reduce NO-3 to NO. S-Nitrosothiols were exposed to saturated VCl3 in HCl, 1% KI in acetic acid, photolysis, or CuCl and CSH in He; NO was measured by chemiluminescence. S-Nitrosothiols were readily detected in VCl3 but not in KI. Reduction in CuCl/cysteine was linear (r2 = 1.0, n = 6), sensitive to 10 pmol, and eliminated by HgCl2; it did not detect NO-2, NO-3, or 3-nitrotyrosine. S-Nitrosothiols represented approximately 2.9% of NOx assayed by VCl3 in human serum, of which <5% were low-mass species. In summary, (i) conventional assays may misidentify NO-3, but not NO-2, as SNOs; and (ii) chemiluminescence/reduction systems may be sensitive and specific as SNO assays. We suggest that assay of the SNO fraction in biological NOx may be more relevant and feasible than is now appreciated.
