Basidiospore identification.

Abstract

Basidiospores are a significant component of the airspora and have been implicated in both allergic rhinitis and asthma. Identification of these spores is hindered by the lack of available reference material. The preparation of reference slides from local specimens is necessary. Collecting procedures and a simple method for making permanent reference slides are presented.

Invisible Threats, Relentless Hunters: Biosurveillance of Airborne Plant Pathogens.

Airborne dispersal enables plant pathogens to travel across fields, regions, and continents, fueling rapid epidemics and emerging disease threats. Biosurveillance, the systematic monitoring of airborne inoculum, offers the opportunity to detect pathogens before symptoms appear and informs timely, risk-based management. Recent advances in air sampling, molecular diagnostics, metagenomics, and imaging technologies have expanded the scale and resolution of pathogen monitoring, from single-species qPCR assays to community-level aerobiome surveys. Integration of biosurveillance data with decision-support systems, remote sensing, and artificial intelligence is transforming early-warning capabilities and providing novel insights into pathogen ecology, evolution, and fungicide resistance. Yet major challenges remain, including assay standardization, data interpretation, and translation into actionable tools for growers. This review synthesizes current approaches, highlights case studies in which biosurveillance has advanced disease management, and outlines future directions toward coordinated surveillance networks and precision agriculture applications.

Cytotoxic fungal spores in the indoor atmosphere of the damp domestic environment.

Eight-three fungal isolates collected and cultured from the air spora of damp public sector Scottish housing have been screened for cytotoxic properties. A bioassay procedure has been utilised for this purpose involving cytotoxic effects on human embryonic diploid fibroblast lung cells grown as monolayer cultures within the wells of microtitre plates. The MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] biochemical assay system employed has revealed that 39 (47%) of the moulds investigated possessed spore-associated cytotoxic properties; a value which emphasises the potential health hazards of the continual inhalation of toxigenic fungal propagules by occupants of mouldy houses. Penicillia, the most prevalent fungi found in the internal atmosphere of the domestic environment, accounted for the great majority of cytotoxic strains identified.

Thermal desorption cold trap-injection in high-resolution gas chromatography: multivariate optimization of experimental conditions.

In studies of low concentrations of volatile compounds in air, the method of adsorption on porous polymers and determination by thermal desorption cold trap-injection high-resolution gas chromatography is finding increasing application. Factors considered important for injection and chromatographic separation of volatile compounds by this method were investigated with the use of multivariate techniques. For the amount injected on to the chromatographic column, the factors of main importance were found to be the temperature of the injection block, the thickness of the internal coating of the cold trap and the flow-rate. Strong interaction effects were noted. For the sharpness of the chromatographic peaks, the flow-rate was the most important factor.
