Computer simulation for teaching membrane potential fundamentals.

Abstract

A computer simulation of membrane potential generation is presented. It allows the user to vary the intra and extracellular concentrations of sodium, potassium and chloride ions (Na+, K+ and Cl-), and determine the membrane potential and the equilibrium potential for each ion. The permeability coefficients for the ionic species considered, and the temperature may also be changed at will. Concentration-potential curves may be obtained at any time. The user may also select a voltage clamp option, which allows him to impose a certain membrane potential value, and study the resulting driving force for each ion. The use of the program in general physiology courses has shown it to be a useful aid for teaching the principles of membrane potential generation.

Binding geometry and photophysical properties of DNA-threading binuclear ruthenium complexes.

The DNA binding conformation and the photophysical properties of the semiflexible binuclear ruthenium complex [micro-bidppz(phen)4Ru2]4+ (2) were studied with optical spectroscopy and compared to the rigid, planar homologue in syn conformation [micro-dtpf(phen)4Ru2]4+ (3) and the parent "light-switch" complex [Ru(phen)2dppz]2+ (1). Comparison of calculated and observed absorption bands of the bridging ligand, bidppz, confirm earlier suggestions that 2 is significantly nonplanar, both free in solution and when intercalated into poly(dAdT)2, but the conclusion that the intercalated conformation is an anti rotamer is not substantiated by comparison of linear and circular dichroism spectra of 2 and 3. The behavior of the emission quantum yield as a function of temperature is similar for the two binuclear complexes 2 and 3 in different protic solvents, and a quantitative analysis suggests that, in solution, the solvent is more strongly hydrogen bonded to the excited state of 2 than to 1. However, the observation that for 2 the radiative rate constant increases to a value similar to 1 upon intercalation into DNA suggests that the difference between 1 and 2 in accepting hydrogen bonds is less pronounced when intercalated.

Current status of the role of Cl(-) ion in the oxygen-evolving complex.

This minireview summarizes the current state of knowledge concerning the role of Cl(-) in the oxygen-evolving complex (OEC) of photosystem II (PSII). The model that proposes that Cl(-) is a Mn ligand is discussed in light of more recent work. Studies of Cl(-) specificity, stoichiometry, kinetics, and retention by extrinsic polypeptides are discussed, as are the results that fail to detect Cl(-) ligation to Mn and results that show a lack of a requirement for Cl(-) in PSII-catalyzed H(2)O oxidation. Mutagenesis experiments in cyanobacteria and higher plants that produce evidence for a correlation between Cl(-) retention and stable interactions among intrinsic and extrinsic polypeptides are summarized, and spectroscopic data on the interaction between PSII and Cl(-) are discussed. Lastly, the question of the site of Cl(-) action in PSII is discussed in connection with the current crystal structures of the enzyme.

N1303K and IVS8-5T, clinical presentation within a family with atypical cystic fibrosis.

The CFTR genotype N1303K/IVS8-5T can cause very mild cystic fibrosis (CF) and congenital bilateral absence of the vas deferens (CBAVD). We report one family consisting of five affected patients in two generations, presenting minor symptoms of CF at different ages, segregating the CFTR mutations N1303K and IVS8-T5-TG13 in trans. Common features were chronic sinopulmonary symptoms and borderline or slightly elevated sweat chloride values. One patient had CBAVD.
