Freshly generated stainless steel welding fume induces greater lung inflammation in rats as compared to aged fume.

It has been previously reported that both short- and long-lived reactive oxygen species (ROS) are present on the surface of freshly generated fumes. The objective of this study was to determine if freshly formed welding fume induces greater lung inflammation and injury in rats due to the presence of reactive oxygen species than aged welding fume. Fume was collected during gas metal arc welding using a stainless steel consumable electrode and found to be of respirable size with a mean diameter of 0.77 microm +/- 0.48. Male CD/VAF rats were dosed intratracheally with the welding fume 30 min (fresh) and 1 and 7 days (aged) after fume collection at a dose of 1.0 mg/100 g b wt. Bronchoalveolar lavage (BAL) was performed 24 h post-instillation. Lung injury and inflammation were assessed by measuring the concentration of neutrophils, albumin, lactate dehydrogenase (LDH), and glucosaminidase (GLU) in the recovered BAL fluid. More neutrophils and enhanced GLU activity were observed for the 'fresh' group as compared to both 'aged' groups (P < 0.05). Slight, but not significant, elevations were seen in albumin content and LDH activity for the 'fresh' group as compared to the 'aged' groups. No significant differences were observed for any of the parameters when fume aged for 1 and 7 days were compared. When the 'fresh' and 'aged' fumes (12.5, 25, and 50 microg/ml) were suspended in dichlorofluorescin (15 microM), a probe which becomes fluorescent when oxidized, the concentration-dependent increases in fluorescence were greater for the 'fresh' fume versus the 'aged' fumes. We have demonstrated that freshly generated stainless steel welding fume induces greater lung inflammation than 'aged' fume. This is likely due to a higher concentration of ROS on fresh fume surfaces.

Measurement of volatile organic compounds by the US Environmental Protection Agency Compendium Method TO-17. Evaluation of performance criteria.

An evaluation of performance criteria for US Environmental Protection Agency Compendium Method TO-17 for monitoring volatile organic compounds (VOCs) in air has been accomplished. The method is a solid adsorbent-based sampling and analytical procedure including performance criteria for four merit parameters. These are: (1) the method detection limit (MDL); (2) the method precision; (3) the agreement between two samples taken at different flow-rates over the same time period [a distributed volume pair (DVP)]; and (4) audit accuracy. A two-adsorbent tube packing of Carbotrap and Carboxen 1000 (Supelco) was tested. Synthetic mixtures containing 41 compounds at concentrations of 10 ppb (v/v) or 2 ppb (v/v) each in humidified zero air, and indoor air from a personal residence, were sampled for 1-h periods during which 1-l and 4-l samples were obtained simultaneously. For synthetic samples, the MDL was determined to be < or = 0.5 ppb (v/v) for 29 of the 41 compounds examined for samples at 47% relative humidity (RH), and for 27 of 41 compounds at 85% RH. The method precision at both 2 and 10 ppb (v/v) was < or = 20% for 90+ % of the samples using five sampling runs and the DVP samples were within 25% for 80% of the samples. For the indoor samples, 26 target compounds were tentatively identified and 12 unknowns were detected; all but 10% of these compounds met the DVP criteria.

Physico-chemical characterization of grain dust in storage air of Bangalore.

An Anderson personal cascade impactor was used to study the particle mass size distribution in the storage air of two major grain storage centers in Bangalore. Dust levels in storage air as well as the personal exposures of workers were determined along with a detailed study on the particle size distribution. Protein and carbohydrate content of the dust were also determined respectively in the phosphate buffer saline (PBS) and water extracts by using the standard analytical techniques. Personal exposures in both of the grain storage centers have been found to be much above the limit prescribed by ACGIH (1995-96). But the results of particle size analysis showed a higher particle mass distribution in the non-respirable size range. The mass median diameters (MMD) of the storage air particulate of both the centers were found to be beyond the respirable range. Presence of protein and carbohydrate in the storage air dust is indicative of the existence of glyco-proteins, mostly of membrane origin.
