Random sampling by glitter drop method.

Abstract

In the course of a study on glioblastoma tissue and cultured cell lines, a glitter drop technique for random sampling was introduced (1). Here, we report the initial description of the details of this procedure and its application to cell counts from explant slides and to determining bromodeoxyuridine (BUdR) labeling indices (LI) in tissues. The method is without bias, easy to perform and takes only a short time. It compares well with three other generally used random sampling procedures.

Sister-chromatid exchange in highly purified human CD4+ and CD8+ lymphocytes.

Sister-chromatid exchange (SCE) frequencies were determined in human peripheral blood CD4+ and CD8+ T lymphocyte subpopulations which were rapidly and highly purified from pooled T lymphocytes by immunological methods. The purified lymphocytes were stimulated with phytohemagglutinin (PHA) for 4 days. CD4+ lymphocytes showed significantly higher SCE frequencies than autologous CD8+ lymphocytes when measured simultaneously after identical bromodeoxyuridine (BrdU) incubation times. Differences in SCE frequencies between CD4+ and CD8+ lymphocytes were also detected when mitomycin C (MMC) was added to the cultures. Higher SCE frequencies in CD4+ lymphocytes were associated with lower proliferating rate indices (PRI) as compared to autologous CD8+ lymphocytes. Abnormalities in CD4+ T lymphocyte function and number in peripheral blood have been observed in several diseases characterized by immunological disorders. Thus, our data may suggest a link between some immunological disturbances and abnormal SCE frequencies in T lymphocyte subsets.

Bromodeoxyuridine

Identification of DNA-replicating lymphocyte subsets using a new method to label the bromo-deoxyuridine incorporated into the DNA.

A flow cytometric procedure measuring the 5-bromo-2-deoxyuridine incorporated into the DNA of cells in S phase was modified to make it compatible with the immunofluorescence labelling of cell surface antigens. The modifications were introduced at the stages of cell fixation and DNA denaturation. Ethanol was replaced by Tween 20-containing paraformaldehyde and hydrochloric acid was used for the denaturation of DNA by bovine pancreatic DNase-I. These two modifications permitted the preservation of immunofluorescence properties and the use of fluorochromes of the phycobiliprotein family such as phycoerythrin and allophycocyanin. This new procedure is suitable for evaluating leucocyte subsets proliferating in vitro following stimulation. As an illustration the immunosuppressive effect of cyclosporin A on PHA stimulated T4-lymphocytes was evaluated.
