A rapid colorimetric method for the determination of Losartan potassium in bulk and in synthetic mixture for solid dosage form.

Two new rapid reproducible and economical spectrophotometric methods are described for the determination of Losartan potassium in bulk and in synthetic mixture for solid dosage forms. Both methods are based on the formation of an orange-red and orange ion-pair complex due to the action of Calmagite (CT) and Orange-II (O-II) on Losartan potassium in acidic medium (pH 1.2). Under optimised conditions, they show an absorption maxima at 491 nm (CT) and 486 nm (O-II), with molar absorptivities of 1.74 x 10(3) and 1.75 x 10(3) l mol(-1) cm(-1) and Sandell's sensitivities of 0.2649 and 0.2637 per 0.001 absorbance unit for CT and O-II, respectively. The colour is stable for 5 min after extraction. In both cases Beer's law is obeyed between 10 and 100 microg ml(-1). The proposed method was successfully extended to synthetic mixture for solid dosage forms.

Application of TLC-densitometry, first-derivative UV-spectrophotometry and ratio derivative spectrophotometry for the determination of dorzolamide hydrochloride and timolol maleate.

Three methods are described for the simultaneous determination of dorzolamide hydrochloride (DORZ) and timolol maleate (TIM) in ophthalmic solutions. The first method is based on application of thin layer chromatographic separation of both drugs followed by the densitometric measurements of their spot areas. After separation on silica gel GF(254) plates, using methanol-ammonia 25% (100:1.5 v/v) as the mobile phase, the chromatographic zones corresponding to the spots were scanned at 253 and 297 nm, respectively. The calibration function was established in the ranges of 2-18 microg for DORZ and 0.5-4.5 microg for TIM. The second method depends on first derivative ultraviolet spectrophotometry, with zero-crossing measurement method. The first derivative values D(1) at 250.2 and 312.5 nm were selected for the assay of DORZ and TIM, respectively. Calibration graphs follow Beer's law in the range 10-64 and 2.5-16 microg ml(-1), respectively. The third method is based on ratio first derivative spectrophotometry. The signals in the first derivative of the ratio spectra at 244 and 306.2 nm were selected to determine DORZ and TIM in the mixture and calibration graphs are linear in the range of 5-40 and 5.0-17.5 microg ml(-1), respectively. The proposed methods were successfully applied to the determination of these compounds in synthetic mixtures and in pharmaceutical preparations. The proposed methods are simple, rapid and suitable for quality control application.

Pulmonary artery pressure-flow relations after prostacyclin in primary pulmonary hypertension.

Exercise tolerance improves within a few weeks after prostacyclin initiation in patients with primary pulmonary hypertension even in the absence of significant changes in resting pulmonary vascular resistance and/or in patients who fail to respond to an acute vasodilator challenge. We tested the hypothesis that this early effect of prostacyclin may be ascribable to an improved pressure-flow response of the pulmonary circulation to exercise. Pulmonary hemodynamic variables at rest and during exercise and the 6-min walking distance were determined before and after 6 wk of continuous intravenous prostacyclin treatment (11 +/- 1.5 ng/kg/min) in seven patients unresponsive to an acute nitric oxide vasodilator test. Mean pulmonary arterial pressure/cardiac index coordinates obtained during exercise were pooled, and the slopes of these plots were compared using covariance analysis. All hemodynamic variables at rest were unchanged after prostacyclin. By contrast, the 6-min walking distance improved in all patients (mean increase, 81 m) and the slope of the mean pulmonary artery pressures/cardiac indexes plot decreased with prostacyclin, from 18.2 to 13.1 mm Hg/L/min/m(2) (p < 0.01). These results suggest that the improvement in exercise tolerance seen after 6 wk of prostacyclin therapy may be ascribable to a decrease in incremental pulmonary vascular resistance during exercise.
