Doripenem.

Abstract

Doripenem is a new member of the carbapenem class of beta-lactam antibiotics with broad-spectrum coverage of Gram-positive, Gram-negative and anaerobic pathogens similar to imipenem and, especially, meropenem. This parenteral antibiotic may offer slightly more activity than meropenem against selected pathogens, including coverage of some strains of Pseudomonas aeruginosa strains not susceptible to the other antipseudomonal carbapenems. Empiric therapy with doripenem, as with other antipseudomonal carbapenems, may be useful in hospital and intensive care unit settings where multidrug resistance has emerged, especially in Gram-negative enteric pathogens. When P. aeruginosa or multidrug-resistant Gram-positive pathogens such as methicillin-resistant Staphylococcus aureus or vancomycin-resistant enterococci are involved, doripenem must be used in combination. Doripenem has a broad spectrum of activity against many common hospital pathogens, including P. aeruginosa and Burkholderia cepacia, which is similar to meropenem.

Performance characteristics of a two-stage dark fermentative system producing hydrogen and methane continuously.

The performance of a mesophilic two-stage system generating hydrogen and methane continuously from sucrose (10-30 g/L) was investigated. A hydrogen-generating CSTR followed by an upflow anaerobic filter were both inoculated with anaerobically digested sewage sludge, and ORP, pH, gas output, %H(2), %CH(4) and %CO(2) monitored. pH was controlled with NaOH, KOH or Ca(OH)(2). Using NaOH as alkali with 10 g/L sucrose, yields of 1.62 +/- 0.2 mol H(2)/mol hexose added and 323 mL CH(4)/gCOD added to the hydrogen and methane reactors respectively were achieved. The overall chemical oxygen demand (COD) reduction was 92.6% with 0.90 +/- 0.1 g/L sodium and 316 +/- 40 mg/L residual acetate in the methane reactor. Operation at 20 g/L sucrose and NaOH as alkali led to impaired volatile fatty acid (VFA) degradation in the methane reactor with 2.23 +/- 0.2 g/L sodium, 1,885 mg/L residual acetate, a hydrogen yield of 1.47 +/- 0.1 mol/mol hexose added, a methane yield of 294 mL/gCOD added and an overall COD reduction of 83%. Using Ca(OH)(2) as alkali with 20 g/L sucrose gave a hydrogen yield of 1.29 +/- 0.3 mol/mol hexose added, a methane yield of 337 mL/gCOD added and improved the overall COD reduction to 91% with residual acetate concentrations of 522 +/- 87 mg/L. Operation at 30 g/L sucrose with Ca(OH)(2) gave poorer overall COD reduction (68%), a hydrogen yield of 1.47 +/- 0.2 mol/mol hexose added, a methane yield of 138 mL/gCOD added and residual acetate 7,343 +/- 715 mg/L. It was shown that sodium toxicity and overloading are important issues for successful anaerobic digestion of effluent from biohydrogen reactors in high rate systems.

A logistic model for the prediction of the influence of water on the solid waste methanization in landfills.

This article deals with the impact of water content of solid waste on biogas production kinetics in landfills. This impact has been proved in the laboratory thanks to anaerobic biodegradation experiments on paper/cardboard waste samples. A strong dependence with the moisture level was observed for both kinetic rates and maximum methane production. In this article, a logistic model is proposed to simulate the biogas production rate. It is chosen as simple as possible in order to allow for a correct identification of the model parameters given the experimental data available. The moisture dependency is introduced through a linear weighing of the biomass specific growth rate and of the amount of accessible organic substrate. It is directly linked to physical properties of the waste: the holding capacity and the minimal moisture level allowing the presence of free water.

Evaluation of thermophilic anaerobic digestion processes for full-scale Class A biosolids disinfection at Hyperion Treatment Plant.

This paper describes 5 phases of full-scale testing at the City of Los Angeles Hyperion Treatment Plant (HTP) for producing Class A biosolids (U.S. EPA Part 503 Biosolids Rule) by thermophilic anaerobic digestion. Phases I and II were tests with a two-stage continuous-batch process in a thermophilic battery of six digesters and a designated post-digestion train that was isolated from mesophilic operations. These tests demonstrated that digester outflow biosolids met the Class A limits for fecal coliforms and Salmonella sp. However, fecal coliform densities sharply increased during post-digestion. The recurrence was possibly related to a combination of a large drop of the biosolids temperature after the dewatering centrifuges and contamination of thermophilically digested biosolids from mesophilic operations. Phase III was conducted after insulation and electrical heat-tracing of the post-digestion train to maintain a biosolids temperature throughout post-digestion at about the same level as in the digester outflow. Biosolids monitoring at the last points of plant control (silos at Truck Loading Facility and farm for land application) indicated that fecal coliform recurrence was prevented. After completing the conversion of HTP to thermophilic operation, certification tests of Phases IV and V demonstrated Class A compliance of a two-stage continuous-batch process under Alternatives 1 and 3 of the Part 503 Biosolids Rule, respectively. HTP received the permit for Class A (indeed exceptional quality) biosolids land application in Kern County, California, in December 2002 under Alternative 3. Since 2003, HTP has consistently complied with the federal and local standards for Class A biosolids, indicating that Class A limits can be met under conditions less stringent than defined by the Alternative 1 time-temperature requirement for batch treatment.
