Method of x-ray anomalous diffraction for lipid structures.

Abstract

The structures of the unit cells of lipid phases that exhibit long-range crystalline order but short-range liquid-like disorder are of biological interests. In particular, the recently discovered rhombohedral phase has a unit cell containing either the structure of a membrane fusion intermediate state or that of a peptide-induced transmembrane pore, depending on the lipid composition and participating peptides. Diffraction from such systems generally presents a difficult phase problem. The existing methods of phase determination all have their limitations. Therefore it is of general interest to develop a new phasing method. The method of multi-wavelength anomalous dispersion is routinely used in protein crystallography, but the same method is difficult for lipid systems for the practical reason that the commonly used lipid samples for diffraction do not have a well-defined thickness. Here we describe a practical approach to use the multi-wavelength anomalous dispersion method for lipid structures. The procedure is demonstrated with the lamellar phase of a brominated lipid. The method is general to all phases as long as anomalous diffraction is applicable.

Scalable methodology for the catalytic, asymmetric alpha-bromination of acid chlorides.

The optimization of a practical, catalytic, asymmetric process for the alpha-bromination of acid chlorides to produce synthetically versatile, optically active alpha-bromoesters is reported. A range of products is produced in high enantioselectivity and moderate to good chemical yields with retention of both upon scale-up. The reactions herein are catalyzed by cinchona alkaloid derivatives, with the best performance achieved by the use of a proline cinchona alkaloid conjugate designed in a de novo fashion.

Determination of polybrominated diphenyl ethers at trace levels in environmental waters using hollow-fiber microporous membrane liquid-liquid extraction and gas chromatography-mass spectrometry.

In this study, we present a simple and easy-to-use extraction method that is based on a hollow-fiber microporous membrane liquid-liquid extraction (HF-MMLLE), as an extraction technique, followed by gas chromatography-mass spectrometry (GC-MS) to determine a group of brominated flame retardants (BFRs), polybrominated diphenyl ethers (PBDEs), at trace levels in aqueous samples. The hollow-fiber membrane (HF) filled with organic solvent was immersed into the aqueous sample, spiked with the analytes at ng l(-1) level, and stirred for 60 min. The proposed method could attain enrichment factors (E(e)) up to 5200 times, after optimising parameters, such as organic solvent, stirring speed and extraction time, that affect the extraction. The HF-MMLLE-GC-MS method was successfully applied to the extraction of PBDEs from tap, river and leachate water samples with spike recoveries ranging from 85% to 110%. The method validation with reagent and leachate water samples provided good linearity, detection limits of 1.1 ng l(-1) or lower, both in reagent and leachate water, as well as satisfactory precision in terms of repeatability and reproducibility with values of % relative standard deviation (%RSD) lower than 8.6 and 16.9, respectively.
