Competitive interaction between two different spherical sinks.

Abstract

Competitive interactions within diverse mixed populations of chemically active sites are prevalent throughout nature, science, and engineering. Their effects are readily seen in the distribution of dead and surviving aerobic cells within a thick biofilm and particle shape changes during the growth and coarsening of crystals. Even in the most dilute case, competition for a reactant requires at least two spheres/cells, and the solution of the two-spherical sink problem is of interest for several reasons. The solution accurately describes lower cell concentration behavior (10(8) cells/l), and like the Smoluchowski diffusion-reaction treatment for a single sphere, the analysis is extremely helpful in understanding the fundamental phenomena of the effect on the first spherical sink of the presence of a second different spherical sink. In addition these exact solutions are required for the systematic extension to higher density behavior by rigorous expansions in the spherical sink densities. The method of the twin spherical expansion is used with a formal matrix elimination scheme to generate an exact solution for two distinct spherical sinks of differing sizes and kinetics. The two sinks exist in a medium, which supplies a reactant to the sinks via Fickian diffusion. The two sinks compete for the same reactant with different first-order reactions occurring at the surface of each sink. Earlier work focused on two spherical sinks of the same size with identical surface reaction kinetics. This work has been advanced to allow for diversity in the theory of cellular or reactive sink competition. A number of interesting higher order interactive phenomena are observed in this paper when the different reactive sinks are in close proximity. (c) 2004 American Institute of Physics.

Dielectric characterization of neutralized and nonneutralized chitosan upon drying.

Isothermal dielectric loss spectra of neutralized and nonneutralized chitosan were acquired in successive runs from -130 degrees C up to increasing final temperatures, in a frequency range between 20 Hz and 1 MHz. Essentially, three relaxation processes were detected in the temperature range covered: (i) a beta-wet process, detected when the sample has a higher water content that vanishes after heating to 150 degrees C; (ii) a beta process, which is located at temperatures below 0 degrees C, becoming better defined and maintaining its location after annealing at 150 degrees C independently of the protonation state of the amino side group; and (iii) a sigma process that deviates to higher temperatures with drying, being more mobile in the nonneutralized form. Moreover, in dried neutralized chitosan, a fourth process was detected in the low frequency side of the secondary beta process that diminishes after annealing. Whether this process is a distinct relaxation of the dried polymer or a deviated beta-wet process due to the loss of water residues achieved by annealing is not straightforward. Only beta and sigma processes persist after annealing at 150 degrees C. The changes in molecular mobility upon drying of these two relaxation processes were evaluated.

Transcription of plasmid DNA: influence of plasmid DNA/polyethylenimine complex formation.

Polyethylenimine (PEI) is one of the most potent non-viral vectors. We have developed a lactosylated PEI (Lac-PEI) to enhance cell-specific transfection and have shown that Lac-PEI is more efficient than unsubstituted PEI for gene transfer into immortalized cystic fibrosis airway epithelial SigmaCFTE29o-cells. As both intact PEI/plasmid and Lac-PEI/plasmid complexes are found in the cell nucleus, we have investigated the transcription efficiency of the plasmid complexed with PEI or Lac-PEI, according to the polymer nitrogen/DNA phosphate (N/P) ratio (from 0 to 20). The initiation of transgene transcription was analyzed in an acellular nuclease S1 transcription assay. For both PEI and Lac-PEI complexes, transcription efficiency varied with the N/P ratio of the complexes. Transcription inhibition was observed when plasmid DNA was either loosely (N/P<5) or tightly condensed (N/P>15). For an N/P ratio of 5 and up to 15, transcription of the complexed plasmid was as efficient as that of the free plasmid. Similar results were observed when gene expression was studied after nuclear microinjection of the complexes into SigmaCFTE29o-cells. Our study shows that condensation of DNA influences the accessibility of the plasmid to the transcription machinery. Interestingly, the charge ratios that allow the most efficient transcription are those usually known to be the most efficient for gene transfer in vitro and in vivo.

Utilization and value addition of copper tailing as an extender for development of paints.

The present study deals with characterization of copper tailing waste, test for possibilities of hazards and its potential reuse as an extender in paints. The waste is a siliceous material containing aluminum oxide, iron oxide and sulphate in significant concentrations. In the primary stage waste is acidic in nature, which makes it unsuitable for paint. This acidity is removed from the waste by simple sieving and grinding. The prepared mass was characterized for basic properties of an extender like oil absorption, specific gravity, pH, etc. Toxicity studies were also conducted in term of leaching of heavy metals by standard techniques (USEPA using TCLP). Properties of the prepared paint's film in terms of hardness, adhesion, resistance to abrasion, resistance to impact, resistance to corrosion (under humidity and salt fog), etc. were evaluated and compared with a similar formulation of conventional extender and found satisfactory. Results from the experiments indicated that developed extender is environmentally clean and cost-effective.
