Protein lipidation.

Abstract

This chapter describes the hydrophobic modification of peripheral membrane-anchored proteins by isoprenylation and S-acylation. The coupling of bacterially expressed protein moieties with chemically synthesized lipopeptides is described as an in vitro alternative for the generation of lipoproteins.

Distorted hexagonal phase studied by neutron diffraction: lipid components demixed in a bent monolayer.

The recent discovery of a distorted hexagonal phase in 1,2-dioleoyl-sn-glycero-3-phosphatidylethanolamine/1,2-dioleoyl-sn-glycero-3-phosphatidylcholine (DOPE/DOPC) mixtures raised the intriguing question as to whether lipid mixtures demix in a bent monolayer. We performed neutron diffraction on a mixture of headgroup deuterated DOPC-d(13) and nondeuterated DOPE to study the lipid distribution in the distorted hexagonal phase. The 1:1 lipid mixture in full hydration and 25 degrees C was in a homogeneous lamellar phase. Upon dehydration the mixture transformed to a rhombohedral phase, then to a distorted inverted hexagonal phase, and finally to a regular inverted hexagonal phase. In the distorted hexagonal phase, the diffraction pattern showed a two-dimensional monoclinic lattice with two reciprocal vectors of equal length (1.5 nm(-)(1)) forming an angle 53 degrees between them. Diffraction intensities measured while varying the D(2)O/H(2)O ratio in the humidity was used to solve the phase problem. The neutron scattering length density distribution of the distorted hexagonal phase was constructed. The constant density contours are approximately elliptical. The difference in the eccentricities of the contours between the water and lipid distributions indicates that the DOPE/DOPC ratio is not uniform around the elliptical lipid tube in the unit cell. DOPE is preferentially distributed at the vertex regions where the curvature is the highest. Thus for the first time it is shown that when a monolayer of a homogeneous lipid mixture is bent, the lipid components are partially demixed in reaching the free energy minimum.

Imaging orientational order and lipid density in multilamellar vesicles with multiplex CARS microscopy.

Multiplex coherent anti-Stokes Raman scattering (CARS) microscopy is used to measure the width of the orientational order distribution of lipid acyl chains within a three-dimensionally confined microscopic probing volume. A theoretical model is developed to describe and simulate the polarization-dependent measurements. We observe that the orientational order in phosphatidylcholine multilammellar vesicles increases significantly upon addition of small amounts (<or=15 mol%) of cholesterol and is significantly reduced for unsaturated lipids. Based on these measurements and using the quantitative nature of multiplex CARS microscopy the exact local concentration of lipid molecules within the vesicles can be measured in terms of the number of lipid bilayers present in the microscopic probing volume.

White-core endosperm floury endosperm-4 in rice is generated by knockout mutations in the C-type pyruvate orthophosphate dikinase gene (OsPPDKB).

We have isolated a floury endosperm-4 (flo4) rice mutant with a floury-white endosperm but a normal outer portion. Scanning electron microscopic analysis revealed that this abnormal endosperm consisted of loosely packed starch granules. The mutant phenotype was generated by T-DNA insertion into the fifth intron of the OsPPDKB gene encoding pyruvate orthophosphate dikinase (PPDK). Plants containing flo4-1 produced no OsPPDKB transcript or the OsPPDKB protein in their developing kernels and leaves. We obtained two additional alleles, flo4-2 and flo4-3, that also showed the same white-core endosperm phenotype. The flo4 kernels weighed about 6% less than wild-type ones. Starch contents in both kernel types were similar, but the total protein content was slightly higher in the mutant kernels. Moreover, lipid contents were significantly increased in the flo4 kernels. Expression analyses demonstrated that the cytosolic mRNA of OsPPDKB was induced in the reproductive organs after pollination, and greatly increased until about 10 days after fertilization. This mRNA was localized mainly in the endosperm, aleurone, and scutellum of the developing kernel. Our results suggest that cytosolic PPDK functions in rice to modulate carbon metabolism during grain filling.
