Identification and quantitation of eleven sesquiterpenes in three species of Curcuma rhizomes by pressurized liquid extraction and gas chromatography-mass spectrometry.

In this paper, GC-MS and pressurized liquid extraction (PLE) was developed for identification and quantitative determination/estimation 11 sesquiterpenes including germacrene D, curzerene, gamma-elemene, furanodienone, curcumol, isocurcumenol, furanodiene, germacrone, curdione, curcumenol and neocurdione in Ezhu which are derived from three species of Curcuma, i.e., Curcuma phaeocaulis, Curcuma wenyujin and Curcuma kwangsiensis by using an analogue as standard. The results showed the methodology could quantitatively compare the quality of three species of Curcuma. The contents of investigated sesquiterpenes in three species of Curcuma were high variant. Hierarchical clustering analysis based on characteristics of 11 identified peaks in GC profiles showed that 18 samples were divided into two main clusters, C. phaeocaulis and C. wenyujin, respectively. C. kwangsiensis showed the characters closed to C. phaeocaulis or C. wenyujin based on its location. Five components such as furanodienone, germacrone, curdione, curcumenol and neocurdione were optimized as markers for quality control of Ezhu.

Spectrophotometric method for polyphenols analysis: prevalidation and application on Plantago L. species.

The prevalidation strategy was applied to evaluate UV-vis spectrophotometric procedure with Folin-Ciocalteu's reagent for polyphenols determination. Favourable prevalidation characteristics verified this procedure as a valuable tool in polyphenols analysis and it was successfully applied for determination of total polyphenols and tannins in leaves, stems and flowers of Plantago L. species growing in Croatia. The results showed the variety of total polyphenols content between different plant parts (leaves: up to 10.15%; stems: up to 4.34% and flowers: up to 5.56%). The content of tannins in stems was from 0.28% to 1.00%, while leaves and flowers contained tannins in concentrations of 2.26% and 2.21%, respectively. The results of polyphenols determination were evaluated by using multivariate analysis (UPGMA and PCA) as a contribution to elucidation of relations between different taxa of genus Plantago L.

Application of the equivalence test according to a concept for analytical method transfers from the International Society for Pharmaceutical Engineering (ISPE).

The performance of the equivalence test in the context of analytical method transfers was investigated by means of a simulation study. An ISPE design proposal and typical error contributions for pharmaceutical routine control have been used for the testing of accuracy. Acceptable results (probability of a correct decision) have been obtained here. For total variations above 0.4% R.S.D. the basic design was not sufficient. An overview for the number of additional series needed corresponding to higher variations has been developed based on further simulations. An alternative approach may be the choice of wider acceptance criteria, which was also evaluated.
