Microalgae-Mediated Synthesis of Gold Nanoparticles from Indonesian Chlorella vulgaris InaCC M205 with Potential Anticancer Properties for Biomedical Application.

Sustainable nanomaterial synthesis has emerged as a critical strategy to reduce the environmental burden associated with conventional chemical synthesis method. Microalgae-derived biomolecules offer a promising platform for the green production of metal nanoparticles due to their rich bioactive compounds capable of acting as natural reducing and stabilizing agents. Here, we report the eco-friendly synthesis of gold nanoparticles (AuNPs) using extract of Indonesian microalga Chlorella vulgaris extract. To optimize the synthesis process, the effects of precursor-to-extract ratio, temperature, and incubation time were evaluated. Optimal synthesis of C5-AuNPs was obtained at 37 °C for 20 h with precursor to extract ratio of 6:4, resulting in moderately stable C5-AuNPs characterized by a surface plasmon resonance (SPR) peak at 541 nm. Furthermore, Fourier-transmission infra-red (FT-IR) analysis revealed the involvement of functional groups of C. vulgaris extract in the interaction with Au+ during the production of C5-AuNPs. Transmission electron microscopy (TEM) demonstrated the formation of uniformly spherical nanoparticles with an average diameter of approximately 8.8 nm. Biological evaluation showed that the synthesized C5-AuNPs exerted pronounced dose-dependent cytotoxicity against MCF-7 breast cancer cells with an IC50 threshold of 21.17 ppm, while no toxicity appears in normal HEK293 cells. Mechanistically, the C5-AuNPs induced early apoptosis and inhibit cell-cycle progression at the stage of G0/G1. Collectively, these findings demonstrate that C. vulgaris-mediated AuNPs represent a promising preliminary in vitro findings for cancer therapy candidate.

Size-controlled assembly of gold nanoparticles induced by a tridentate thioether ligand.

The ability to control the size and shape of nanoparticle assemblies is essential for the ultimate applications in sensors, catalysis, medical diagnostics, information storage, and quantum computation. This report demonstrates a novel mediator-template strategy toward this ability by exploring molecular driving forces exerted by a tridentate thioether as a mediator and tetraoctylammonium bromide as a templating agent. A combination of the ligand mediation, the surfactant templating, and their relative concentrations served as the driving forces. This combination leads to unprecedented spherical assemblies of gold nanoparticles in controllable sizes via manipulation of the relative concentrations of mediating and templating components.

(Non)-migration of radiopaque markers used for on-line localization of the prostate with an electronic portal imaging device.

PURPOSE: Radiopaque gold markers can be implanted in the prostate to visualize its position on portal images during radiation therapy. This procedure assumes that the markers do not move within the prostate. In this work we test this assumptiom. METHODS AND MATERIALS: Three markers were implanted transrectally in the prostate of patients undergoing external radiation therapy. An orthogonal pair of portal images was acquired periodically throughout the course of radiation therapy with an a-Si electronic portal imaging device (EPID). The marker coordinates were determined, and the distances between the implanted markers were recorded. The distance time trend is used to evaluate the magnitude of marker migration. RESULTS: The average standard deviation (SD) of the distances between markers was 1.3 mm (range 0.44 to 3.04 mm). Three of the 11 patients show a SD larger than 2 mm. For these patients, all three distances show a simultaneous reduction with time, compatible with a shrinking of the prostate. All had been treated with neoadjuvant hormone therapy. For 1 of the 3 patients, this reduction in volume was confirmed with a repeat computed tomographic scan. CONCLUSION: None of the 33 markers studied migrated significantly. The implantation of three radiopaque gold markers enables accurate and precise on-line verification of the prostate position during external beam radiation therapy. The use of three markers provides a tool to monitor prostate position and volume changes that can occur over time due to hormone or radiation therapy.
