A versatile modification of on-column oligodeoxynucleotides using a copper-catalyzed oxidative acetylenic coupling reaction.

We report herein a versatile postsynthetic modification of on-column oligodeoxynucleotides (ODNs) using a copper-catalyzed oxidative acetylenic coupling reaction. Hexamers supported on resins via a methylamino-modified linker were prepared, and on-column modifications of ODNs were examined. ArgoPore resin proved to be the best choice for the modification, and introduction of functional molecules, such as anthraquinone, biotin, and fluorescein, resulted in good yields at not only the 5'-terminal but also the internal 3'-end of the ODNs. This method is applicable to the modification of 12mer ODN consisting of a random sequence. The resulting ODN9 possessing fluorescein at its 5'-terminal acts as a non-RI primer for primer extension assays using the Klenow fragment.

Synthesis of enantioenriched propargylic alcohols related to polyketide natural products. A comparison of methodologies.

[reaction: see text] Applications of methodology for the synthesis of propargylic alcohols related to polyketide natural products were examined. Noyori's asymmetric transfer hydrogenation of alpha-chiral alkynones was found to be highly selective and catalyst controlled. Additions of TMS acetylene to alpha-chiral aldehydes, catalyzed by a Ti(O-i-Pr)(4)-BINOL complex, were diastereoselective but substrate dependent.

Sonogashira coupling using bulky palladium-phenanthryl imidazolium carbene catalysis.

[structure: see text] Bulky phenanthracenyl imidazolium-derived carbene ligands were investigated for copper-free Sonogashira coupling with terminal acetylenes. Aryl bromides and iodides gave coupled products in excellent yields from the Pd(PPh(3))(2)Cl(2) complex with potassium t-butoxide and 18-crown-6 in THF. A remarkable dependence on the size of the ligand was found. The highest yields were obtained with the bulky 2,9-dicyclohexyl-10-phenanthryl ligand 5.
