Constriction and Dnm1p recruitment are distinct processes in mitochondrial fission.

Mitochondria undergo cycles of fusion and fission crucial for organelle homeostasis. Fission is regulated partially by recruitment of the large GTPase Dnm1p to the outer mitochondrial membrane. Using three-dimensional time-lapse fluorescence imaging of Saccharomyces cerevisiae cells, we found that Dnm1p-EGFP appears and disappears at "hot spots" along mitochondrial tubes. It forms patches that convert rapidly into different shapes regardless of whether mitochondrial fission ensues or not. Moreover, the thickness of the mitochondrial matrix displays frequent temporal fluctuations apparently unrelated to fission or to recruitment of Dnm1p-EGFP. These results suggest that mitochondrial fission requires coordination of at least two distinct processes.

[Lymphocytes: what is "Vav"].

Guanine exchange factors (GEF) of the Vav family are critical activators of Rho GTPases, which control actin cytoskeletal reorganization and gene transcription. Among all GEFs identified, Vav proteins are the only GEFs regulated by tyrosine phosphorylation. Moreover, their structure contains several protein-protein or protein-lipid interaction domains. These domains are involved in the formation of multimolecular signalling complexes, highlighting the adaptor role of Vav proteins. The unique combination of these properties makes Vav proteins privileged integrators of multiple signalling pathways in a broad range of tissues and cells. Lymphocyte function during inflammatory and immune responses requires a dynamic remodeling of cellular architecture. Thus, it is not surprising that Vav proteins have been found to play a central role in the regulation of physiologic and pathologic lymphocyte responses.

Gem GTPase: between a ROCK and a hard place.

Gem GTPase is a member of a protein family that includes Rad, Rem and Rem2. Although until recently precious little was known about the function of Gem, recent studies have revealed that Gem may influence cell morphology by antagonising the actions of the Rho GTPase effector protein ROCK I.
