[Yeasts utilizing methyl alcohol].

Abstract

The ability to utilize methanol was found in the yeast strains isolated from forest substrates (Zygowillia pini IBFM y-655, Torulopsis molicshiana IBFM y-657) and in the collection yeast cultures (Candida boidinii IBFM y-588 and IBFM )-587). These yeasts assimilated not only methanol but other alcohols (ethanol, glycerol, erythrol, mannitol, and sorbitol), nitrates or nitrites. Their fermentation ability was low.

Metabolic engineering of Candida yeasts for biotechnological applications.

Candida yeasts represent a versatile yet underexploited platform for industrial biotechnology. These yeasts utilize a remarkably broad range of carbon sources, particularly for hydrophobic carbon sources, coupled with robust growth and diverse biosynthetic capacities, making them promising hosts for sustainable production of chemicals, fuels, and proteins. Despite these advantages, industrial deployment of Candida species has been hindered by concerns regarding opportunistic pathogenicity and the historical lack of efficient genetic manipulation tools, leading to a substantial gap between metabolic potential and practical utilization. Recent advances in functional genomics, genome editing, and systems metabolic engineering are rapidly overcoming these barriers, enabling more precise and efficient strain development. In this review, we systematically summarize recent progress in the metabolic engineering of Candida species as microbial cell factories, with particular emphasis on expanding genetic toolkits, utilizting renewable and non-conventional carbon sources, and biosynthesizing high-value compounds. In addition, we propose a biosafety-oriented classification framework to support their safe industrial deployment. Finally, we discuss current challenges and emerging opportunities, emphasizing that the synergy of synthetic biology and artificial intelligence-driven design holds the key to unlocking the biotechnological potential of Candida yeasts.

R-stereopreference analysis of lipase Novozym 435 in kinetic resolution of flurbiprofen.

Immobilized lipase from Candida antarctica (Novozym 435) was employed in the kinetic resolution of racemic flurbiprofen by enantioselective esterification with methanol. It was found that the lipase has the R-stereopreference and the reaction matches Bi Bi Ping Pong mechanism with dead-end inhibition of methanol. Furthermore, the R-stereopreference was analyzed in details from the aspects of enzymatic kinetic mechanism and reaction activation energy of both enantiomers. The R-enantiomer shows lower activation energy and higher maximum reaction rate than the S-enantiomer, which implies the R-stereopreference of the lipase and makes the kinetic resolution of flurbiprofen via enzymatic reaction feasible.

Seven-membered cyclic sulfite eudesmane derivatives: partial synthesis, structural determination, and enzymatic resolution.

Two chromatographically inseparable, diastereomeric eudesmane cyclic sulfites have been semisynthesized from alpha-santonin. This diastereomeric mixture was resolved by enzymatic acetylation with CCL. Each cyclic sulfite derivative was individually characterized on the basis of its spectroscopic and crystallographic properties.
