An efficient protocol for solution- and solid-phase end-group differentiation of spermidine.

Abstract

The end-group differentiation of a selectively protected spermidine was achieved through a short sequence of steps. The functionalization of spermidine in solid-phase was monitored by FT-IR.

Using trifluoroacetic acid to augment studies of potato suberin molecular structure.

Systematically varied reaction times and concentrations of trifluoroacetic acid (TFA) have been used to remove polysaccharides associated with suberin isolated from potato wound periderm, thereby augmenting spectroscopic determinations of the molecular structure of this protective plant polyester. Treatments with dilute TFA left a residual insoluble material for which both solid-state 13C and 1H NMR spectra displayed significant improvements in resolution without compromising the integrity of the protective plant polyester, whereas higher concentrations of TFA made it possible to achieve controlled hydrolysis of the suberin aliphatic or aromatic domains. Among the isolated fragments were two hydroxyphenyl derivatives reported previously in lignins and a novel aliphatic-aromatic ester trimer that is identified provisionally. Together these protocols help to characterize the carbohydrate types that are bound covalently to the suberin polyester and to identify the interunit covalent linkages among the aliphatic ester, phenolic, and carbohydrate moieties in suberized potato tissue. The strategies described herein may also advance molecular-level investigations of lignocellulosic materials or vegetable tissues that exhibit strengthened intercellular adhesion.

Efficient asymmetric synthesis of novel 4-substituted and configurationally stable analogues of thalidomide.

The preparation of new thalidomide derivatives 4-methyl-(3S,4R)-3a and 4-phenyl-(3S,4S)-3b starting from pyroglutamic acids (2R,3R)-7a and (2R,3S)-7b, possessing an inappropriate stereochemistry, was successfully realized due to stereochemically complete epimerization at the alpha-stereogenic center upon formation of the corresponding N-phthaloyl anhydrides 9a,b. The demonstrated conformational stability of these new thalidomide derivatives provides solid experimental evidence for practical feasibility of the approach described here to overcome the inherent problem of configurational instability of thalidomide by introducing an alkyl or aryl group in the C4 position. TMSCH2Li-induced regioselective lithiation of (S)-nicotine.

The first regioselective C-4 lithiation of (S)-nicotine has been realized using TMSCH2Li as basic reagent in toluene. The reaction proceeded under mild conditions with a small excess of electrophile. The 4-chloro derivative was subsequently metallated at C-5 with the same basic reagent in THF at -78 degrees C. This methodology opens a straightforward access to functional diversity in (S)-nicotine chemistry.
