The anxiogenic beta-carboline FG-7142 inhibits locomotor exploration similarly in postweanling and adult rats.

The beta-carboline FG-7142 exerts many stress-like effects in the adult rat, including the inhibition of locomotor exploration. However, comparable effects in immature animals have not been reported. Stress-like effects of FG-7142 are mediated via its inverse agonist actions on gamma-aminobutyric acid (GABA) receptors. GABA systems change considerably with development and GABA agonists such as diazepam have opposite behavioral actions in immature and adult rats. Accordingly, we compared FG-7142 effects on locomotor exploration in postweanling and adult rats. Postweanling male rats (24 days) and adult rats (90 days) received a single injection of vehicle or FG-7142 (15 mg/kg i.p.) and 15 min later were placed in photocell monitors for 1 h. Although postweanling animals traveled a smaller distance overall than adults, FG-7142 inhibited locomotor exploration to a similar degree in both groups. We conclude that FG-7142 exerts stress-like effects in postweanling rats and may be considered for use as a model of childhood stress.

Developmental changes of eukaryotic initiation factor 2B subunits in rat hippocampus.

Regulated protein synthesis is critical for neural development, such as the formation of synapses and neural circuits and the modulation of synaptic plasticity. Protein synthesis is controlled by translation factors, including initiation, elongation and release factors. Here we investigated the developmental changes of eukaryotic initiation factor 2B (eIF2B) subunits in rat hippocampus. The eIF2B beta, gamma, delta and epsilon subunit protein levels were maximal at embryonic day 18 and then decreased during development. Aged hippocampus contained only trace amounts of these subunits. The finding that eIF2B subunit levels are high in developing hippocampus suggests that regulated protein synthesis is active in young, highly plastic brain.

Lactate dehydrogenase isoenzymes in dental pulp of rats according to stage of root development.

The objective of this study was to present a classification of the root development stage of female rat molar teeth and to evaluate the variation in the lactate dehydrogenase (LDH) activity and electrophoretic isoenzyme profile according to the stage of root development of the molar teeth. We also studied the LDH activity and isoenzymes of the pulp of incisor teeth. The stage of development of the rat first molar at the age of 15 days and that of the second molar at the age of 18 days was classified as the beginning of root formation. At the age of 15 days, the electrophoretic profile of the isoenzymes for the first molar showed a prevalence of LDH-1 followed by LDH-2. However, for the maxillary second molar there was a prevalence of LDH-4 followed by LDH-1, while for the mandibular second molar LDH-1 predominated followed by LDH-2 and LDH-4. From 18 days of age, the prevalence was always of LDH-1. The electrophoretic profile of LDH isoenzymes from the pulp of the incisor teeth at the ages studied (25 and 60 days) showed the following order of prevalence: LDH-1 > LDH-2 > LDH-3 > LDH-4 > LDH-5. These results suggest that there are variations in the prevalence of the various forms of LDH isoenzymes in the dental pulp of rats according to the developmental stage of the root.
