A decrease in Sendai virus infection potency by interactions with cationic thermo-responsive polymers.

Abstract

Three types of synthetic cationic polymers were incubated with Sendai virus (HVJ), and virus infection of Rhesus monkey kidney cells, LLCMK2, was measured by the plaque assay method. One composition poly(N-isopropylacrylamide-co-N,N-dimethylaminopropyl acrylamide-co-butylmethacrylate) of (P(IP-27DA-16BM)) showed substantial decreases in the infection potency of the virus over a wide range of polymer concentrations. This polymer expressed a significant virus titer decrease even at a very low concentration (2 x 10(-4) wt%), and its virus titer reduction was found to be incubation temperature-dependent. As the virus was incubated with this polymer above the phase transition temperature of this polymer over a long period, the virus titer reduction was larger. Selectivity to the virus titer reduction in comparison with cytotoxicity to LLCMK2 cells was evaluated. This P(IP-27DA-16BM) polymer was revealed to possess a higher selectivity to virus reduction than sodium dodecyl sulfate. This indicates the feasibility of this polymer for a virus-specific inactivating agent.

Mucoadhesive properties of carbopol or chitosan-coated liposomes and their effectiveness in the oral administration of calcitonin to rats.

Mucoadhesive liposomes were prepared by coating multilamellar liposomes with Carbopol (CP) in a similar manner to that used in the preparation of chitosan-coated liposomes (CS-Lip) previously reported [Takeuchi et al., Chem. Pharm. Bull. 42 (1994) 1954-1956; Pharm. Res. 13 (1996) 896-901]. The positively charged liposomes containing dipalmitoylphosphatidylcholine (DPPC) and stearylamine at an optimum formulation ratio (40:1) could be coated with CP without aggregation of the liposomal particles during the coating process. The mucoadhesive properties of both the resultant polymer-coated liposomes (CP-Lip and CS-Lip) and the positively or negatively charged noncoated liposomes (Non-Lip) were evaluated using the rat intestine. The order was CS-Lip> or = CP-Lip>positively charged Non-Lip>negatively charged Non-Lip. The adhesive property of CP-Lip decreased on increasing pH of the dispersing medium from pH 5.0 to 7.4, probably owing to the electric repulsion between CP-Lip and the mucus layer. The effectiveness of the liposomal formulations in oral administration of calcitonin was evaluated by measuring the change in the blood calcium concentration of rats. Administration of CP-Lip and CS-Lip containing calcitonin showed an enhanced and prolonged reduction in blood calcium concentration. The overall pharmacological effect of CP- and CS-Lips evaluated by means of the area under the plasma calcium concentration curve was 2.4 and 2.8 times higher than that of negatively and positively charged Non-Lips, respectively.

Patterned monolayer/polymer films for analysis of dilute or salt-contaminated protein samples by MALDI-MS.

This paper describes a surface science/mass spectrometry effort to develop and characterize a patterned gold surface that serves as a MALDI sample platform capable of concentrating and purifying proteins. Using microcontact printing, small (200-microm diameter) hydrophilic spots of bare gold or chemically anchored poly(acrylic acid) (PAA) are patterned at 5-mm intervals in a hydrophobic field consisting of a self-assembled monolayer of hexadecanethiol. Building on recent innovations by others, the small hydrophilic spots concentrate the sample to achieve good reproducibility and high sensitivity in the MALDI signal. One of the key features in this work is the combination of the high density of carboxylate groups in PAA with a small spot size to afford both concentration and purification of proteins via ionic interactions. This translates into detection limits for salt-contaminated proteins that are 20-100 times lower (low femtomole) than those reported for previous polymer- or monolayer-modified MALDI probes (using proteins in the 3-15-kDa range). Reflectance FT-IR spectroscopy and ellipsometry were used to determine the amount of protein adsorbed to a PAA-modified sample plate as a function of pH and salt concentration. Amide absorbances in IR spectra correlate well with MALDI-MS signals measured after addition of 2,5-dihydroxybenzoic acid as a matrix.
