Combinatorial networks.

Abstract

A novel approach for the analysis and virtual screening of large combinatorial libraries is presented. The method attempts to relieve the computational burden by computing the properties of the products in a way that does not require their explicit enumeration. In particular, a small subset of compounds from the virtual library is identified and their descriptors are calculated in a conventional manner. The resulting data is used as input to a multilayer perceptron, which is trained to predict the descriptors of the products from the descriptors of their respective building blocks. Once trained, the neural network is able to estimate the descriptors of the remaining members of the virtual library with remarkable accuracy, without ever, generating their connection tables. This method eliminates the two most time-consuming steps in virtual screening and allows the processing of very large combinatorial libraries that are intractable with conventional techniques.

Diagnostics for conformity of paired quantitative measurements.

Matched pairs data arise in many contexts - in case-control clinical trials, for example, and from cross-over designs. They also arise in experiments to verify the equivalence of quantitative assays. This latter use (which is the main focus of this paper) raises difficulties not always seen in other matched pairs applications. Since the designs deliberately vary the analyte levels over a wide range, issues of variance dependent on mean, calibrations of differing slopes, and curvature all need to be added to the usual model assumptions such as normality. Violations in any of these assumptions invalidate the conventional matched pairs analysis. A graphical method, due to Bland and Altman, of looking at the relationship between the average and the difference of the members of the pairs is shown to correspond to a formal testable regression model. Using standard regression diagnostics, one may detect and diagnose departures from the model assumptions and remedy them - for example using variable transformations. Examples of different common scenarios and possible approaches to handling them are shown.

Measurement of ultrasmall volumes using anodic stripping voltammetry.

This paper discusses a new and effective method for small volume determination. The benefits of working with small volumes are numerous and include using less material, generating less waste, increased functionality in less space, portability, and high throughput. As sample size decreases, measuring its volume becomes more challenging. The most prevalent method for small volume determination is visual inspection. The method presented in this paper is precise, more quantitative, and adaptive to many solution geometries, because it is based on a coulometric approach. Demonstration of this method is performed on volumes of approximately 1 nL containing silver and using a self-contained microcavity device that contains multiple electrodes. The silver is "exhaustively" deposited during a cathodic potential step, followed by anodic stripping voltammetry. It is expected that an even faster and more accurate analysis of volumes much smaller than those evaluated here is easily possible using this approach.
