Soft lining materials: composition and structure.

Abstract

An analysis of five proprietary acrylic soft lining materials revealed that they can be classified into two groups. The first of these groups (Soft Oryl and Coe Soft) resemble the tissue conditioners in composition in that their liquid component contains no monomer. A study of their mechanical properties may reveal that these materials should, in fact, be classified as tissue conditioners. The second group (including Palasiv, Virina and Coe Super Soft) were more conventional acrylic materials which differed in the amount of plasticizer and the type of acrylic monomer used in the liquid component and in the nature of the polymer component.

Short-chain and medium-chain aliphatic-ester synthesis in Saccharomyces cerevisiae.

In the yeast Saccharomyces cerevisiae, the enzymes which catalyse the synthesis of ethyl acetate, ethyl n-hexanoate and isoamyl acetate were partly resolved from a fraction containing slowly sedimenting lipoproteins released during cell disruption with glass beads. Solubilization with detergents and fractionation by affinity chromatography have demonstrated the presence of at least three, and probably four, ester synthases which differ in their catalytic properties. Isoamyl-acetate synthase was solubilized and extensively purified to apparent homogeneity by successive chromatographies on various columns. On the basis of its specific activity in cell-free extracts, the enzyme was purified 19,000-fold with a 5% activity yield. As judged by SDS/PAGE, it consists of a single polypeptide chain with a molecular mass of 57 +/- 3 kDa and its apparent pI is 5.5. The enzyme acetylates isoamyl alcohol, ethanol and 12-DL-hydroxystearic acid from acetyl-CoA but is unable to use n-hexanoyl-CoA as a cosubstrate. This enzyme, defined as an acetyl-CoA: O-alcohol acetyltransferase, could be the product of one of the anaerobically induced genes in S. cerevisiae.

Dominant role of glucagon in the initial induction of phosphoenolpyruvate carboxykinase mRNA in cultured hepatocytes from fetal rats.

The injection of streptozotocin to 18-day-old rat fetuses induced, 2 days later, a 50% fall in plasma insulin and a twofold increase in plasma glucagon concentrations and liver cAMP levels. Phosphoenolpyruvate carboxykinase mRNA that were undetectable in the fetal rat liver, accumulated 48 h after streptozotocin injection, their concentration being 30% of that found in the liver of 1-day-old newborn rats in whom liver phosphoenolpyruvate carboxykinase gene expression is maximal. Physiological concentrations of glucagon (0.7 +/- 0.2 nM) induced, within 2 h, phosphoenolpyruvate carboxykinase mRNA accumulation in cultured hepatocytes from 20-day-old fetuses. The addition of insulin (0.01-100 nM) inhibits, by no more than 30%, the glucagon-induced phosphoenolpyruvate carboxykinase mRNA accumulation. Exposure of fetal hepatocytes to insulin for 24 h did not change the glucagon dose/response curve and did not lead to a more efficient inhibition of the glucagon-induced phosphoenolpyruvate carboxykinase mRNA accumulation, despite a clear stimulatory effect on the rate of lipogenesis. In contrast, when hepatocytes were cultured in the presence of dexamethasone, the glucagon-induced phosphoenolpyruvate carboxykinase mRNA accumulation can be totally inhibited by pharmacological concentrations of insulin (10 nM). From these in-vivo and in-vitro studies, it is concluded that, under physiological conditions, the postnatal rise in plasma glucagon concentration is more important than the fall in the plasma insulin concentration for the primary induction of liver phosphoenolpyruvate carboxykinase gene expression.

In vitro and in vivo effects of ammonia on glucose metabolism in the astrocytes of rat cerebral cortex.

Effects of 1 and 5 mM ammonium acetate on glucose metabolism were studied in astrocytes. But for an elevation in the levels of fructose-6-phosphate, phosphoenol pyruvate, and pyruvate, glucose metabolism was unaltered in the presence of 1 mM ammonium acetate. With 5 mM ammonium acetate, but for unaltered lactate, ADP, ATP and decreased aspartate, levels of several intermediates were elevated. Similar results were obtained when astrocytes isolated from hyperammonemic rats were incubated with glucose except for an enhanced production of 14CO2 from [U-14C]glucose. It is suggested that glucose metabolism of astrocytes may not be severely affected in astrocytes of cerebral cortex in acute hyperammonemic states.
