Vascular endothelial growth factor gene expression regulated by protein kinase C pathway in endothelial cells during hypoxia.

Objective. To investigate the relationship between Vascular endothelial growth factor (VEGF) gene expression and protein kinase C (PKC) activity. Method. 1) Rat's primary pulmonary artery endothelial cells (PAEC) were cultured under hypoxia condition (1% O2). Changes of PKC activity and VEGF mRNA in PAEC were detected at 0 (control), 1, 3, 6, and 12 h in the hypoxic condition of culture. 2) After addition of PKC inhibitor (staurosporine) in culture medium and culturing PAEC in hypoxic condition immediately, PKC activity and VEGF mRNA in PAEC were measured at the same time. VEGF protein in culture medium under the two conditions above were also detected. Result. PKC activity in PAEC were obviously elevated at 1 h during hypoxia as compared with the control (P<0.05); VEGF mRNA expression in PAEC and VEGF protein level in culture medium were increased significantly (P<0.01) at 3 h and 6 h during hypoxia respectively as compared with the control (P<0.01); After addition of PKC inhibitor in culture medium and culturing cells in hypoxia condition immediately, PKC activity in PAEC decreased significantly as compared with that at 0 h (P<0.01), and there were no significant changes of VEGF mRNA in PAEC and VEGF protein level in culture medium at any time (P>0.05). Conclusion. The results demonstrate that hypoxia stimulates pulmonary arterial vascular endothelial cells to secrete VEGF, and PKC may be one of the factors that up-regulate VEGF gene expression during hypoxia.

[Retroviral vector-mediated HBsAg expression and its stability].

OBJECTIVE: To explore use of retroviral vector in gene therapy of hepatitis B. METHODS: The recombinant vector Plxsn-HBs was constructed by inserting HBV S gene into pLXSN. The pseudovirus, which was produced from PA317 after transferring with pLXSN-HBs by electroporation, were frozen at different temperature. The activities of the pseudovirus to infect eukaryotic cells and express antigen were determined by comparing the numbers of G418-resistant clones and assaying HBsAg in the supernatant of the cells with ELISA after infection HepG2, NIH3T3 and 293 cells. RESULTS: It was hard to find changes in HBsAg amount at different intervals and different temperatures. G418 resistant clones, however, were variable. When frozen at -20 degrees C, the numbers of clones were half less than that of the beginning after 6 months, few clones were formed after 12 months, and no clone was found after 24 months. When frozen at -40 degrees C, the numbers of clones were 121, 332 and 89 42, 137 and 43 for HepG2, NIH3T3 and 293 cell lines at 12 and 24 months, respectively. When frozen at -70 degrees C, the numbers of clones were 159 463 and 112 for HepG2, NIH3T3 and 293 cell lines at 24 months, respectively. There was no statistical difference compared to that of zero months. CONCLUSIONS: The activity of the peseudovirus to infect eukaryotic cells and expressed antigen was not changed after 2 years frozen at -70 degrees C.

[The inhibitory effect of anti-streptococcus mutans immunoglobulin of yolk on glucan synthesis of Streptococcus mutans].

OBJECTIVES: To observe the inhibitory effect of anti-Streptococcus mutans Hen-egg-yolk (IgY) on glucan synthesis of Streptococcus mutans. METHODS: Determine the glucan with anthrone method after using the different titer of IgY on S.mutans and S.sobrinus. RESULTS: The inhibitory effect of antibody on S.mutans and S.sobrinus synthesis glucan had a similar tendency. The effect of inhibit glucan synthesis was influenced by the titer of antibody. High titer with high inhibition. Begin with the group of 1:16 titer, the group of 1:2 titer showed the highest inhibition (P<0.05). CONCLUSION: The antibody can reduce synthesis of glucan from S.mutans and S.sobrinus.
