[Relationship between obese gene expressive product and obesity].

In order to study the relationship between obese protein and obesity. A radioimmunoassay for human plasma obese protein (OP) was established using human OP (57-92) and its specific antiserum. We observed the distribution and the characteristics. Of OP in normal human and mouse tissue and changes of plasma OP in obese patients. We also observed the tissue distribution after intravenous injection of 125I-labeled OP fragments (116-167) and (93-105) in rats. The results were as follows. The contents of OP were much higher in brain tissue than those in abdominal adipose tissue; levels of OP in liver and in skeletal muscle tissue were zero; the contents of abdominal adipose tissue were higher in female mice than those in male mice; the contents of plasma OP in normal human beings were 194.3 +/- 17.7 ng/L, while those in mice were 2257.8 +/- 171.9 ng/L. It was also found that the administered OP fragments were widely distributed in various tissue and organs including the brain. Kidney was the richest in the OP fragments; liver and lung ranked second. The half time of the OP fragments in plasma clearance was about 4.5 min. The contents of OP in obese adults and in obese children were much lower than those in normal control group. There was significant negative correlation between OP levels and body mass index, serum concentration of glucose, cholesterol and triglycerides. So the present study indicates the important relationship between the changes of OP contents and the pathogenesis of obesity.

[The effect of minimal dexamethasone on the rate of growth of mouse embryonic fibroblasts in vitro].

OBJECTIVE: To study of the relationship between inhibition of the growth rate of mouse embryonic fibroblasts (MEF) and the dosage of dexamethasone. METHOD: Cell culture. RESULTS: The effect was observed from 48 to 72 hours after addition of 50 micrograms/ml (0.005%) dexamethasone. In comparison with the control, the number of MEF was cut down by 16%-26%, whereas the addition of dexamethasone between 1 microgram/ml and 10 micrograms/ml (0.0001-0.001%) had no significant effect on the rate of growth of MEF. CONCLUSION: The minimal dosage of dexamethasone has no significant inhibitory effect on the growth rate of fibroblasts and wound healing.

[The reverse medial plantar island flap: anatomy and clinical applications].

A new flap, the reverse medial planter island flap based on the distal end of the medial plantar artery (MPA) is introduced. It is found that the MPA is connected with the plantar profund artery via the first plantar metatarsal artery or the big lateral branch of the MPA, joining the common digital artery that comes from the arch of the plantar artery. This flap was used in 3 cases of electric injury with satisfactory results. It suggests that the reverse medial plantar island flap is an ideal flap for reconstruction of the defects in the weight-bearing area of the forefoot.

[The structural changes of cortical bone beneath plate after rigid platefixation and removal].

We investigated the structural changes of bone under the plate after plate fixation and removal. Twenty-four New Zealand white rabbits were plated on their intact left tibiae with stainless steel plates and 4 animals served as controls. The plates were removed 2 months after implantation in 20 plated animals, of which, 4 were sacrificed immediately after plate removal and the other were killed in successive groups of 4 each at 1, 2, 3 and 4 months after plate removal respectively. The remaining 4 plated animals were killed at 6 months after implantation. Bone samples were harvested and prepared for scanning microscopic observation. Internal fixation with a rigid plate may lead not only to osteopenia of the cortex under the plate, but also to disorganization of the cortex, in which the mineral column and collagen fibers were oriented in a randomized pattern. The regional osteoporosis could recover gradually after removal of the rigid plate. However, the restoration of normal bone structure occurred later than that of the bone mass. Delayed restoration of bone structure might be one of the potential causes of refracture of the plated bone.

[The etiologic role of thermal injury on the induction of NO and NOS in plasma, burned wounds and visceral organs].

Using a 35% TBSA burned animal moedl, we investigated the levels of NO contents (nitritc concentration assay) and NOS (hemoglobulin absorbent optical density test) in the burned wound, circulation, as well as five visceral organs which inluded heart, lung, liver, kidney and intestine mucosa. Samples were obtained from scalded male Wistar rats at 1 hour postburn (PBh1)PBh3, PBh8, PBh12, PBh24, PBh48 and PBh72, respectively. Samples of sham burned rats were obtained as controls. The plasma level of NO concentration was decreased within 72 hours postburn especially at PBh3, 8, 12, 24, 48 (P < 0.05 to P < 0.01). This was in agreement with our findings in the ESR assay. The cutaneous NO cotents was 3.8 to 57.8 times higher than that of plasma and other visceral organs. It is thus possible for burned skin tissue to be an important sites in the production of NO. However, the cutaneous NO might have little systemic influences and can only be a local fator due to its short half-life. The close relationship of changes in the NO and NOS in five visceral organs postburn provided strong evidence on the function of NOS as NO premerase.

[Pharmacological effects of zisu and baisu].

The extract and volatile oil of zisu (Perilla frutescens) has shown significant antipyretic effect in rabbits and antiemetic effect in pigeons. The fatty oil extracted from its seeds has significant antitussive effect in mice and anti-asthmatic effect in guinea pigs. The extract, volatile oil and fatty oil of Baisu, which is of the same genus as Zisu have the same effects as those of Zisu. The acute toxicities of the extract and fatty oil of Zisu and Baisu, whether by peroral or by intraperitoneal, are similar to each other. These results indicate that Baisu has the same pharmacological effects as Zisu, and thus can be used as a substitute for Zisu.

[Extraction and sequencing of trace DNA from cornu Cervi pantotrichum].

The trace DNA extraction method was used to extract DNA from the blood, hair, Cornu cervi pantotrichum, hairy antler of the sika deer, Cervus nippon, and penis of Bulbalus bublis and Asinus vulgaris. A 307 bp fragment of mitochondrial DNA cytochrome b gene was amplified with primers L14841 and H15149 for these extractions. The purified PCR products were sequenced by the dideoxy method. DNA sequence obtained from the Cornu cervi pantotrichum is the same as that obtained from the fresh blood and hair of the sika deer, but the sequence of the so-called hairy antler is quite different, implying that it is not from the sika deer. In addition, the phylogenetic relationships among different species are congruent with the traditional known phylogeny.

[The effect of cryotreat on the size of the medium and high melting-point castable alloys].

Castable alloys are widely used in prosthodontics. The properties of alloys can be improved by cryotreat. This method may cause a size alteration of materials. This study revealed the effects of different cooling rates on the size of the castable alloys. The results showed: 1. The rapid cooling rate caused significant deformation of the alloys. 2. The 3 degrees C/min rate and the 1 degree C/min rate did not cause any significant deformation of the alloys. 3. The 3 degrees C/min rate and the 1 degree C/min rate did not show any difference in their deformation causing ability. 4. The rapid cooling rate and the slow cooling rate (3 degrees C/min and 1 degree C/min) showed significant difference in their deformation causing ability. These results suggested that 3 degrees C/min was recommendable as a cooling rate of the cryotreat of the medium and high melting-point castable alloys.

[The effect of cryotreat on the mechanical properties of the medium and high melting-point castable alloys].

Two mechanical properties that are closely related to the clinical practice, i.e. the surrendering toughness and the hardness, of the medium and high melting-point castable alloys, were tested before and after the cryotreat. The results showed: 1. Cryotreat significantly increased the surrendering toughness of the medium and high melting-point castable alloys (40.00%); 2. Cryotreat significantly increased the surrendering toughness of the medium and high melting-point castable alloys (25.45%); 3. Cryotreat significantly increased the surrendering modulus of the high melting-point castable alloys (25.77%); 4. Cryotreat significantly increased the hardness of the medium melting-point alloys(24.40%); 5. Cryotreat significantly increased the hardness of the medium melting-point alloys (7.77%). These results suggest that cryotreat is an effective procedure in enhancing the toughness and hardness of the medium and high melting-point castable alloys.

[Applications of computer image processing and graph processing technologies in digital modelling of 3-D shapes of teeth, jaw and prosthesis].

In this paper, the images of sliced sections or CT sections of the specimen of teeth, jaw and prosthesis were processed by a computer image analysing system, and the plane shape of the 3-D objects of interest on the sections was converted into digital images using a special software module for contour analysing. Then the images were adjusted in scale, modified in coordinates and smoothed in shape, and the data format was transferred for finite-element analysis by a specially designed software. In such ways a method for the digital modelling of the 3-D shape of teeth, jaw and prosthesis was successfully established.

[Arginine-glutamine replacement at residue 57 of NADH-cytochrome b5 reductase in Chinese hereditary methemoglobinemia].

OBJECTIVE: To elucidate the molecular mechanism of NADH cytochrome b5 reductase (Cytb5R) deficiency in hereditary methemoglobinemia. METHODS: Cytb5R cDNA was cloned from white blood cells from patient with hereditary methemoglobinemia by RT-PCR method, and the genomic DNA from 3 pedigrees with hereditary methemoglobinemia were analyzed by restriction enzyme analysis. RESULTS: On sequencing the cDNA, two missense mutation were found. One is CGG-->CAG at codon 57 of exon 3, caused Arg-Gln replacement. The other is GAG-->GGG at codon 222 of exon 8. The former mutation abolishes the Msp I recognition site which was confirmed in two of three hereditary methemoglobinemia family. The latter mutation generates a recognition site for Bsi Y I. Amplification of exon 8 by PCR followed by digestion with Bsi Y I revealed no mutation in all patients from the three families. CONCLUSION: Arg57-Gln replacement is responsible for Cytb5R deficiency in the two Chinese pedigrees.

[Determination of elemental sulfur in liquefied gas by high performance liquid chromatography].

A method for the determination of elemental sulfur in liquefied gas has been developed in this work. First, the elemental sulfur existed in the liquefied gas was extracted by chloroform and the volatile petroleum hydrocarbons vaporized with a rate of ca. 500mL/min at low temperature in an ice water bath. The elemental sulfur dissolved in chloroform was then determined by high performance liquid chromatography. The operating conditions were: 300 x 4mm i.d. mu-Bondapak C18 column; 0.8mL/min methanol-water, 95/5 (V/V), as eluent and UV-254nm detector. The average recovery of elemental sulfur was 98.3% and the minimum detectable limit was 0.1ng/g.

Legal issues in health care administration: what is important to the administrator?

Many legal questions are presented to hospital administrators daily. To determine the relative importance of their concerns, a questionnaire covering 57 legal issues was submitted to the administrators of all 196 Louisiana hospitals. The survey indicated that every issue was considered important, but some issues were considered very important. Sometimes the level of concern was influenced by the characteristics of the hospital, for example, size and ownership.
