[Effect of knockout AT1a receptor gene on transplasmamembrane calcium influx in aortic smooth muscle cells].

OBJECTIVE: To investigate the mechanism of angiotensin II receptor subtype (AT1a) mutant on angiotensin II (Ang II) mediated transplasmamembrane calcium influx in aortic smooth muscle cells (SMCs) from AT1a knockout and wild type mice. METHOD: Aortic SMCs were isolated and cultured. Intracellular free calcium concentration [Ca2+]i was measured using Fura-2/AM fluorescentic technique. RESULTS: Ang II caused a marked increase in [Ca2+]i in both cell types [AT1a group: (204 +/- 22) nmol/L vs (108 +/- 9) nmol/L]; CONTROL: [(194 +/- 19) nmol/L vs (110 +/- 7) nmol/L]. Administration of both calcium channel blocker, nifedipine and GTP-gamma s significantly inhibited Ang II effect, in contrast, application of pertussin toxin (PTX) activated the Ang II mediated calcium influx. The response of AT1a knockout cells was more sensitive to nifedipine and were enhanced by PTX. CONCLUSION: Calcium influx induced by Ang II can be regulated by PTX insensitive and non-Gi protein in AT1a knockout cells.

[Functional expression of human GM-CSF receptor in NIH3T3 cells].

OBJECTIVE: To characterize human GM-CSF receptor(GM-R) reconstituted in NIH3T3 cells. METHODS: Mouse NIH3T3 cells which do not express human GM-R were transfected with cDNAs encoding the alpha and beta subunit of the human GM-R, and then ligand-stimulated proliferative signal transduction and tyrosine phosphorylation of the positive transfectants were examined. RESULTS: The reconstituted functional receptor, GM-R alpha/beta, could mediate cell proliferation and colony formation in soft agar, and induce tyrosine phosphorylation of beta c, Jak2, Shc and Shc-associated P145(SHIP). However, human GM-CSF did not activate mitogenic signal expression of NIH3T3 cells transfected with GM-R alpha alone. CONCLUSIONS: Introduction of both alpha and beta subunit of human GM-R into NIH3T3 cells can result in ligand-dependent cell growth and colony formation via phosphorylation of beta c and activation of Jak2, Shc and SHIP signal transduction pathway.

Molecular mechanism of recessive congenital methemoglobinemia in Chinese pedigrees.

OBJECTIVE: To investigate the molecular mechanism of recessive congenital methemoglobinemia (RCM) in Chinese and to establish a gene diagnostic method of polymerase chain reaction-restricted fragment length polymorphism (PCR-RFLP). METHODS: Total RNA was isolated from the peripheral leukocytes of the propositus and b5R cDNA synthesized by reverse transcription-polymerase chain reaction (RT-PCR). The coding region of b5R cDNA was analyzed by sequencing of the RT-PCR products. RESULTS: Both propositi A and B were found to be homozygotes for a G to A transition at codon 57 in exon 3, changing a guanine to an adenine. This point mutation was not an artificial occurrence during polymerase chain reaction (PCR), as confirmed by Msp I restriction enzyme analysis of the genomic DNA. Propositus A's mother and propositus B's sister and her nephew were found to be heterozygotes for the mutation. CONCLUSION: A guanine-to-adenine transition at codon 57, replacing arginine with glutamine, was the molecular basis for RCM in two Chinese families.

[mRNA expression of basic fibroblast growth factor in the pulmonary tissues from rats repeatedly infected with mycoplasma pneumoniae].

OBJECTIVE: To investigate changes in mRNA expression of basic fibroblast growth factor (bFGF) in pulmonary tissues from rats repeatedly infected with Mycoplasma pneumoniae (MP). METHODS: Rats were infected with MP for 9 times during a period of 24 weeks by ultrasonic nebulizing inhalation to establish animal models of chronic pulmonary MP infection. In situ hybridization was performed with bFGF cDNA probe and results were quantitatively analysed to measure the changes of bFGF mRNA expression in lung tissues. RESULTS: (1) MP polymerase chain reaction (PCR) tests showed positive results in the bronchoalveolar lavage fluid (BALF) from MP infected animals (n = 4) while results were negative for BALF from the control rats (n = 4) and rats infected with MP but treated with erythromycin (n = 4). Bacterial cultures of the bronchial and the lung tissues were negative in all three groups. Observation with transmission electron microscope showed that interalveolar septa were widened with increased amount of collagen in the MP infected rats while there were no obvious abnormalities in the other two groups. (2) Positive expression of bFGF mRNA were found in alveolar walls of MP infected rats. No expression of bFGF mRNA was found in control animals. In the rats infected with MP but treated with erythromycin positive expression of bFGF mRNA was found to be scarcely distributed in alveolar walls. Quantitative analysis showed that the optical densities of bFGF mRNA expression were 41.32 +/- 10.44 in the MP infected rats (n = 4), which were significantly higher than those in the control group (0.30 +/- 0.13, n = 4, P < 0.01) and those in the MP infected and erythromycin treated rats (6.03 +/- 2.41, n = 4, P < 0.01). CONCLUSIONS: Repeated pulmonary infection with MP can lead to up-regulation of bFGF mRNA expression, which suggests that bFGF might play a role in the pathogenesis of pulmonary interstitial fibrosis after repeated MP infection.

[Changes of transmembrane ion transport in essential hypertensives with familial histories].

OBJECTIVE: To investigate the effects of heredity on transmembrane ion transport in essential hypertension (EH). METHODS: 52 essential hypertensives were divided into two groups, 23 with positive family history (FH(+)) and 29 with negative family history (FH(-)). The rate constant of ouabain-sensitive sodium efflux ((o)K(os)) was measured as the half of the increase in erythrocyte Na(+) concentration during incubation with ouabain for 2 hours, (o)K(os) was calculated by an equation and the maximal rate (Vmax) of red cells Na(+)/H(+) exchange was determined as the Na(+) influx promoted by an outward H(+) gradient. Intracellular Na(+) concentration was measured with flame photometry and (45)Ca(2+) influx in ATP-depleted red cells with liquid scintillation counting. RESULTS: The red cell Na(+)/H(+) exchange and (45)Ca(2+) in-flux were significantly higher but the (o)K(os) lower in both the FH(+) and FH(-) groups than in the controls. FH(+) group demonstrated a significantly higher (o)K(os) and lower Ca(2+) influx as compared with those in FH(-) group. In FH(+) group, systolic blood pressure was positively correlated with age, while diastolic blood pressure positively correlated with (45)Ca(2+) influx. CONCLUSION: Our data show a genetic contribution to abnormal membrane transport in hypertension. There is a defect in membrane permeability to Ca(2+) and a greater exchangeable pool of cytosolic free Ca(2+) in FH(+) group that participate in the pathogenesis of inherited hypertension.

[The effect of blood-pancreatic juice barrier on 5-fluorouracil in post-pancreatoduodenectomy patients].

OBJECTIVE: To observe the distribution and the relationship of 5-Fu in the plasma and the pancreatic juice in post-pancreatoduodenectomy patients after 5-Fu given intravenously. METHODS: After 5-Fu (1.0 g/m(2)) was given intravenously in post-pancreatoduodenectomy patients, blood and pancreatic juice were collected. The 5-Fu concentrations were determined by HPLC and at last the penetration ratio (PR) of 5-Fu was studied with PCNONLIN. RESULTS: 5-Fu in the plasma penetrated the pancreatic tissue and crossed the blood-pancreatic juice barrier (BPJB) in post-pancreatoduodenectomy patients (PR = 1.01 +/- 0.49). The concentration of 5-Fu in pancreatic juice was much higher than that in plasma. CONCLUSIONS: 5-Fu is suitable for adjuvant chemotherapy of pancreatic adenocarcinoma after pancreatoduodenectomy.

[Comparison of subclinical infection rates between vaccinated group with type I inactivated vaccine against hemorrhagic fever and controls].

OBJECTIVE: To compare subclinical infection rate in the vaccinated group with type I inactivated vaccine against hemorrhagic fever with renal syndrome (HFRS) with that in the controls and to understand its enhancement. METHODS: A trial field was selected in Jiande County, Zhejiang Province during July 1974 to November 1997. Paired-sera before and after vaccination were collected from 401 vaccinee and 360 controls, respectively. Serum titer of indirect immunofluorescent IgG antibody (IFAT) against HFRS was detected for each of them, and its cut-off value depended on the distribution of serum antibody titer in the second determination in the controls, which could be used to evaluate subclinical infection before vaccination. RESULTS: There was no significant difference in subclinical infection between those with positive and negative IFAT before vaccination, with different cut-off values for identifying subclinical infection. In both vaccinated and control groups with negative IFA before vaccination, subclinical infection rate was 7.62% in the controls, and 2.17% and 1.63% in the vaccinated ones, with cut-off values of 1:160 and 1:320, respectively, significantly different from that in the former. Subclinical infection rate was 11.38% and 6.78% in the vaccinated ones, as cut-off values of 1:20 and 1:40, respectively, without significant difference from the controls. CONCLUSION: No increase in subclinical infection in the vaccinated group with type I inactivated vaccine against HFRS was found.

[Expression in Lactococcus lactis of catalytically active phenylalanine ammonia-lyase from parsley].

The phenylalanine ammonia-lyase (PAL) cDNA of parsley (Petroselinum crispum) was subcloned into constitutive expression vector pMG36e downstream of the P32 promoter. The resulting plasmid pMG36ePAL was introduced into Lactococcus lactis subsp. lactis MG1363 by electroporation. The recombinant strain showed its PAL activity conversing the L-phenylalanine in the culture medium into trans-cinnamic acid. A new secretory vector pXHS was constructed by recombination of pMG36e with a Lactococcal usp45 secretion leader coding sequence and a translational coupling sequence. Then the pXHSPAL was constructed and used for expression of PAL in L. lactis, the PAL activity was also detectable. The L. lactis dnaJ promoter sequence was cloned and used to construct a heat inducible vector pXHJ. PAL cDNA was cloned into pXHJ and the L. lactis IL1403 was transformed with the recombinant plasmid pXHJPAL. After a heat shock from 30 degrees C to 37 degrees C, the PAL activity of the pXHJPAL strain could increase approximately onefold. The prospect of using these engineering L. lactis strains for PKU therapy was also discussed.

Molecular basis for interprotein complex-dependent effects on the redox properties of amicyanin.

The quinoprotein methylamine dehydrogenase (MADH), type I copper protein amicyanin, and cytochrome c-551i form a complex within which interprotein electron transfer occurs. It was known that complex formation significantly lowered the oxidation-reduction midpoint potential (Em) value of amicyanin, which facilitated an otherwise thermodynamically unfavorable electron transfer to cytochrome c-551i. Structural, mutagenesis, and potentiometric studies have elucidated the basis for this complex-dependent change in redox properties. Positively charged amino acid residues on the surface of amicyanin are known to stabilize complex formation with MADH and influence the ionic strength dependence of complex formation via electrostatic interactions. Altering the charges of these residues by site-directed mutagenesis had no effect on the Em value of amicyanin, ruling out charge neutralization as the basis for the complex-dependent changes in redox properties. The Em value of free amicyanin varies with pH and exhibits a pKa value for the reduced form of 7.5. The crystal structure of reduced amicyanin at pH 4.4 reveals that His95, which serves as a ligand for Cu2+, has rotated by 180 degrees about the Cbeta-Cgamma bond relative to its position in oxidized amicyanin and is no longer in the copper coordination sphere. At pH 7.7, the crystal structure of reduced amicyanin contains an approximately equal distribution of two active-site conformers. One is very similar to the structure of reduced amicyanin at pH 4.4, and the other is very similar to the structure of oxidized amicyanin at pH 4.8. Potentiometric analysis of amicyanin in complex with MADH indicates that its Em value is not pH-dependent from pH 6.5 to 8.5, and exhibits an Em value similar to that of free amicyanin at high pH. The structure of reduced amicyanin at pH 4.4, with His95 protonated and "flipped", was modeled into the structure of the complex of oxidized amicyanin with MADH. This showed that in the complex, the redox-linked pH-dependent rotation of His95 is hindered because it would cause an overlap of van der Waals' radii with residues of MADH. These results demonstrate that protein-protein interactions profoundly affect the redox properties of this type I copper protein by restricting a pH-dependent, redox-linked conformational change of one of the copper ligands.

A practical synthesis of 3-substituted delta 3,5(6)-steroids as new potential 5 alpha-reductase inhibitor.

A new and practical synthetic approach to 3-substituted delta 3,5(6)-Steroids, as potential 5 alpha-reductase inhibitor, is described. The key step involves Pd-catalyzed coupling reaction of steroid 3-enol 5H-3-oxo-octafluoropentanosulfonates. 3-Phenylacetylenyl substituted delta 3,5(6)-steroid 3g and 3-phosphate substituted delta 3,5(6)-steroid 3f in our synthesized compounds exhibited high 5 alpha-reductase inhibitory activity in vitro assay.

Inhibition of vascular endothelial growth factor-induced receptor activation with anti-kinase insert domain-containing receptor single-chain antibodies from a phage display library.

A single-chain antibody phage display library was constructed from spleen cells of mice immunized with a soluble form of a human vascular endothelial growth factor (VEGF) receptor, kinase insert domain-containing receptor (KDR). After two rounds of biopanning, >90% of the clones recovered were specifically reactive to KDR. Subsequent selection identified two clones that blocked VEGF binding to KDR. The clones were expressed in Escherichia coli and purified as soluble single-chain Fv (scFv) antibodies. The affinities of the scFv for binding to KDR were determined by BIAcore analysis (2.1 x 10(-9)-5.9 x 10(-9) M). One scFv, p1C11, was shown to inhibit VEGF-induced KDR phosphorylation and VEGF-stimulated DNA synthesis in human umbilical vein endothelial cells. There is much experimental evidence to suggest that the VEGF/KDR/Flk-1 pathway plays an important role in tumor angiogenesis, a process that is essential for tumor growth and metastasis. The antibodies discussed here, which block VEGF binding to KDR, have potential clinical application in the treatment of cancer and other diseases where pathological angiogenesis is involved.

Redox properties of tryptophan tryptophylquinone enzymes. Correlation with structure and reactivity.

The pH dependence of the redox potentials for the oxidized/reduced couples of methylamine dehydrogenase (MADH) and aromatic amine dehydrogenase (AADH) were determined. For each enzyme, a change of -30 mV/pH unit was observed, indicating that the two-electron transfer is linked to the transfer of a single proton. This result differs from what was obtained from redox studies of a tryptophan tryptophylquinone (TTQ) model compound for which the two-electron couple is linked to the transfer of two protons. This result also distinguishes the redox properties of the enzyme-bound TTQ from those of the membrane-bound quinone components of respiratory and photosynthetic electron transfer chains that transfer two protons per two electrons. This difference is attributed to the accessibility of TTQ to solvent in the enzymes. One of the quinol hydroxyls is shielded from solvent and thus is not protonated. The unusual property of TTQ enzymes of stabilizing the anionic form of the reduced quinol is important for the reaction mechanism of MADH because it allows stabilization of physiologically important reaction intermediates. Examination of the extent to which disproportionation of the MADH and AADH semiquinones occurred as a function of pH revealed that the equilibrium concentration of semiquinone increased with pH. This indicates that the proton transfer is linked to the semiquinone/quinol couple. Therefore, the quinol is singly protonated, and the semiquinone is unprotonated and anionic. It was also shown that the oxidation-reduction midpoint potential for AADH is 20 mV less positive than that of MADH over the range of pH values that was studied and that the TTQ semiquinone of AADH was less stable than that of MADH. This may be explained by differences in the active site environments of the two enzymes, which modulate their respective redox properties.
