Expressions of IGF-1 and TGF-beta 1 in the condylar cartilages of rapidly growing rats.

OBJECTIVE: This study was designed to investigate the expressions of IGF-I and TGF-beta 1 in condyles and to determine if the special modeling and remodeling of the condylar cartilages are regulated by growth factors. METHODS: Fifty 5-weeks-old male rats were assigned to five groups randomly, and one group was sacrificed every week. The condyles were harvested from rats whose ages ranged from 5- to 9-weeks-old and they were inspected by using S-P immunocytochemical techniques. RESULTS: IGF-I and TGF-beta 1 are expressed during condylar cartilage development in a temporally and spatially regulated fashion, and different growth factors have different expressing characteristics. The expressions of IGF-I are more intense in germinal and transitional layers, while TGF-beta 1 expression was high in the mature and degenerated layers. In germinal and transitional layers, IGF-I expression was high at the beginning of rapid growth, while TGF-beta 1 expression was low. At the end of the rapid growth, IGF-I expression was lower and TGF-beta 1 expression was still high. In the mature and degenerated layers, TGF-beta 1 expression was very high, while IGF-I expression was lower throughout rapid growth. CONCLUSIONS: The growth factors are expressed differentially, and each shows specific or overlapping localization patterns depending on the stage of developmental process, meaning that IGF-I and TGF-beta 1 may regulate condylar cartilage proliferation and differentiation in an interrelated system. This study suggests that growth factors can be produced locally by condylar cartilages and play an important role in the metabolism of condylar cartilages.

[Determination of glycosylated hemoglobin in human erythrocytes by fast protein liquid chromatography system].

The chromatography method for analysis of glycosylated hemoglobin(HbA1c) in human erythocytes was established by using a fast protein liquid chromatography system(FPLC) and a Mono S column. The average intrabatch and interbatch coefficients of variations (n = 3) were 2.5% and 4.8%, respectively. There was a positive correlation between the results of FPLC and routine microcolumn method (r = 0.413, P < 0.02). Using FPLC method, the HbA1c levels in festing blood of 54 normal donors and 26 diabetics on admission were 4.6 +/- 1.19(%) and 10.2 +/- 3.45(%), respectively. A significant difference was found statistically between the results of two groups (t = 10.8, P < 0.01).

[Relationship between PGI2, TXA2 and threatened preterm].

The objectives of this study were to investigate the levels of 6-keto-PGF1 alpha, TXB2 and progesterone in plasma and cervical mucus of women with threatened preterm labor (Group I), term labor (Group III) versus their controls (Groups II and IV) respectively and to investigate the relationship between these substances and threatened preterm labor. One case was matched with one control (pregnant woman) by weeks of gestation. Radioimmunoassay was used to determine the levels of 6-keto-PGF1 alpha, TXB2 and progesterone. Statistical differences were assessed by student t test. The results showed that the levels of TXB2, 6-Keto-PGF1 alpha in plasm from women with term labor and threatened preterm labor were higher than those of the control groups (P < 0.001) respectively. The ratios of 6-keto-PGF1 alpha and TXB2 were lower than their controls (P < 0.001) respectively. No differences in the levels of progesterone were noted between the labor groups and their control groups (P > 0.05) respectively. The results observed in cervical mucus were in line with there in plasma. These results suggest that the levels of PGI2, TXA2, and PGI2/TXA2 ratio were associated with the onset of term labor.

[Identification and determination of Shenbaiye].

This study was amied at the method of identifying and determining Shenbaiye so that a ground can be provided for the quality requirements. Three Chinese materia medica (Radix Sophorae flavescentis, I; Cortex Phellodendri, II; Rhizoma Atractylodis, III) of Shenbaiye were identified by means of TLC successfully. The method in detail and the chromatogram were presented. The determination of I in Shenbiaye was carried out by solid phase cleaned-up and separated through neutral alumine column (10 x 1 cm i.d.) eluting with chloroform. This eluent was extracted with sulphuric acid (0.01 mol/L) and the excess of acids was titrated with sodium hydroxide (0.02 mol/L) using the methyl red as an indicator. The total alkaloid content of I was calculated as matrine of I in Shenbaiye. This method was established with a precision of 1.10% (n = 5) and the recoveries of 97.59 +/- 1.28% (n = 8). The results of determination of samples were reported.

[An immunohistochemical study of the proliferating activity of ameloblastic fibroma and ameloblastic fibrosarcoma].

Six cases of ameloblastic fibroma (AF) and three cases of ameloblastic fibrosarcoma (AFS) were immunohistochemically investigated with a proliferating cell nuclear antigen (PCNA) monoclone antibody. Data on the PCNA labeling index (LI) of both epithelial and mesenchymal components were analyzed. The results showed that the PCNA LI of mesenchymal component of AFS (40.8%) was significantly higher than that of AF (3.2%) (P < 0.01) and that of the epithelial component of AFS (5.3%) was significantly lower than that of the mesenchymal one of AFS (P < 0.01). The results suggested that PCNA LI be a useful marker for the differentiation diagnosis of AF and AFS.

Effect of gynostemma pentaphyllum mak on carcinomatous conversions of golden hamster cheek pouches induced by dimethylbenzanthracene: a histological study.

OBJECTIVE: To evaluate the effect of gynostemma pentaphyllum mak (GP) on carcinomatous conversions of leukoplasia of the golden hamster cheek pouch induced by dimethylbenzanthracene (DMBA). METHODS: 284 golden hamsters, ranging from 4 to 8 weeks of age, 80 to 100 grams in weight were randomly divided into three groups: control, model and experimental groups. Control group (8 hamsters) were sacrificed at week 0 and week 12 for control. Model group (123 hamsters) were pasted in bilateral cheek pouches with 0.5% DMBA, 3 times a week for establishment of experimental leukoplasia, and sacrificed at week 2 to week 9 (12-22 hamsters were sacrificed a week). Experimental group (153 hamsters) were divided into three subgroups. Subgroup A underwent simultaneous DMBA pasting and oral taking of GP, and were sacrificed at week 6 and week 9. Subgroup B underwent oral taking of GP for 12 weeks after completion of DMBA pasting. Subgroup C underwent DMBA pasting after completion of oral taking of GP for 12 weeks. RESULTS: The earliest epithelial dysplasia and carcinoma in situ occurred at week 3 and week 6 after DMBA pasting. There were significant differences between subgroup B and model group for 3 weeks' administration of DMBA (P < 0.05), and between subgroup C and model group (P < 0.01), while there was no difference between subgroup B and model group for 6 week's administration of DMBA (P > 0.05). CONCLUSIONS: GP could inhibit and reverse the carcinomatous conversions of leukoplasia of golden hamster cheek pouches, indicating its positive anticarcinogenic effect.

[Relationship between sleep apnea and cerebral blood flow].

OBJECTIVE: To study the cerebral blood flow in patients with and without sleep apnea syndrome (SAS). METHOD: Thirty patients with SAS were studied, with 32 patients without SAS as controls. The age, sex and baseline diseases were comparable between these two groups. Subjects were studied by polysomnography and transcranial Doppler ultrasonography. The systolic (VS) and mean (VM) cerebral blood flow velocities, resistent index (RI), and frequency spectral shape were determined. Anterior, middle and posterior cerebral arteries were examined by transcranial Doppler ultrasonography in all patients. RESULT: VS and VM were significantly lower in patients with SAS than those in the control subjects (P < 0.05 and P < 0.01). RI of the right PCA in SAS patients (0.56 +/- 0.06) was significantly higher than that in the controls (0.48 +/- 0.09, P < 0.05). Changes in the cerebral blood flow frequency spectral shape were remarkably increased in the SAS group as compared to control group. VS and VM were negatively correlated with apnea index (r = -0.413 and -0.628, P < 0.05 and 0.01), but positively correlated with SaO2(r = 0.435 and 0.712, P < 0.05 and 0.01). CONCLUSION: The results suggest that SAS patients have decreased cerebral blood flow velocities and more frequency shape abnormalities. These may decrease cerebral perfusions, and lead to cerebral atherosclerosis and stroke.

[Activation of insulin-like growth factor 1 gene expression in growing rat condyle by functional mandibular protrusion].

OBJECTIVE: We studied the changes of the IGF-1 mRNA expression in condylar cartilage to elucidate the biomolecular mechanism of mandibular advancement. METHODS: Sixty 5-week-old male rats were randomly divided into experimental and control groups. The mimic functional appliances were used in experimental group. The rats were sacrificed after 1, 3, 7, 14, 21 and 28 days to study the expression of IGF-1 mRNA by in situ hybridization. RESULTS: The results showed IGF-1 mRNA expression in the condylar cartilage. The transitional and maturational layers had the most abundant IGF-1 mRNA, and the IGF-1 mRNA abundance of germinal layer is higher than that of synovial and fibrous layers. Functional mandibular protrusion activated the IGF-1 mRNA expression and it began to increase after 3 days of the experiment, and the changes were most significant in 7-14 days. CONCLUSION: This suggested that the condylar cartilage growth and remolding after functional protrusion might be caused by IGF-1 gene expression activation.

[Clinicopathological and diagnostic study of amyloidosis of oral maxillofacial area].

OBJECTIVE: To study the clinicopathological features of amyloidosis studied in oral maxillofacial area and the diagnostic methods. METHODS: HE staining and staining of alkaline Congo red, Immunohistochemical staining, Sections examined by transmitted and polarized light microscopy, Transmitted electron microscopy. RESULTS: 14 cases of the disease were reported. Clinically, the patients usually had progressive enlargement and solidness of the tongue (macroglossia, 12 cases), hard nodules or cords of the cheek and lip mucosa (8 cases), single or multiple hard masses of submandibular and submental areas (6 cases), and ecchymosis or petechiae of the face, neck, chest or oral mucosa. Microscopically, the disease is characterized by the even pink amyloid protein deposit in the soft tissues and the wall of blood vessles, which shows positive staining of alkaline Congo red, Immunohistochemical staining can differentiate the primary amyloidosis from secondary one. Ultrastructurally, Amyloid protein appeares as felt-like finely filament. CONCLUSION: Amyloidosis involving in oral maxillofacial area mainly arise in tongue, less frequently, in buccal and lip mucosa, salivary glands. It is not difficulty to dignosis the disease by HE staining combining with staining of alkaline Congo red and other specific staining methods. Ultrastructural and immunohistochemical study is helpful to make show the dignosis.

[Dynamic change of androgen-receptor in tumor and surrounding liver tissue of nude mice model with transplanted human HCC].

OBJECTIVE: To research into the law of androgen-receptor (AR) dynamic change in developing course of human HCC. METHOD: Dynamic measurement of AR in tumor and peritumorous tissue was performed in nude mice model with orthotopic transplanted human HCC. RESULT: Tumor nodules were present in 90% nude mice 4 weeks after transplantation, tumor necrosis was found in the 10th week, and remarkable necrosis occurred in the 12th week. The quantity of AR in tumor and peritumorous tissue was highest in the early stage of tumor (102.32 +/- 21.42 fmol/mg protein and 72.45 +/- 10.11 fmol/mg protein respectively), and then decreased progressively with time to the 10th week (40.98 +/- 21.11 fmol/mg protein and 53.39 +/- 7.01 fmol/mg protein respectively). The difference in AR between each two weeks was significant. CONCLUSION: Androgen promotes the development of tumor because of existence of AR, and its action may decline after establishment of tumor. Therefore, antiandrogen treatment may be ineffective after establishment of tumor.

[Induction of insulin-like growth factor I expression to mandibular advancement in growing rat condylar cartilage].

It was studied effects of functional mandibular advancement on IGF-I peptide of condylar cartilage of 60 5-week-old male rats. Animals were randomly divided into the experimental and control groups, and the mimic functional appliances were used in experimental group. The rats were killed after 1, 3, 7, 14, 21 and 28 days. The results showed that the condylar cartilage of growing rats expressed IGF-I the strongest in the germinal layer, medial in the transitional layer, and the least in the maturational layer. IGF-I positive cells and their immunoreactive levels increased after 1 week of functional mandibular advancement, and reached the peak level at 2 weeks. The results suggest IGF-I in mandibular condyle of rat is related to its growth and differentiation activity, and changes of condylar IGF-I after functional protrusion are relevant to the histologic changes and cellular functions which indicate their involvement with active bone growth and remodeling.

[Differential expression of IGF-I and its mRNA in mandibular condylar cartilage of rat--direct evidence for servosystem theory of facial growth].

It was studied the expression of IGF-I and its mRNA in the condylar cartilage of 10 7-week-old SD male rats by using in situ hybridization and immunohisto-chemistry technique. The results showed both IGF-I and its gene expressed in growing rat condyle. IGF-I peptide was abundant in germinal zone, and positive reaction of its mRNA was strongest in transitional and maturational zones. These indicate that condylar cartilage has the capability of local production and secretion of IGF-I, mediating the command effect of STH, and differential expression of IGF-I and its mRNA might establish the local feedback loop, which supply a direct evidence for servosystem theory of facial growth.
