[Telomerase activity in rat tongue carcinogenesis].

OBJECTIVE: To detect the telomerase activity in rat tongue cancer and precancerous lesions and to investigate the possibility of telomerase as oral cancer biological marker. METHODS: A total of 69 rat tongue carcinogenesis specimens induced by 4-nitroquinoline-1-oxide (4NQO) were examined for telomerase activity by PCR-telomeric repeat amplification protocol (TRAP) silver staining and ELISA. RESULTS: Sequential increase in telomerase activity in tongue carcinogenesis was found. The positive rate of telomerase in normal, HP, mmDP, sDP, ISC and SCC was 0.0%, 12.5%, 25.0%, 66.7%, 72.7% and 93.8%. CONCLUSIONS: Telomerase may be a tumor specific gene marker, and may play an important role in early diagnosis and treatment of oral cancer.

[Changes of blood circulation of the extremity with tibia shaft defects treated with external fixation].

OBJECTIVE: To study external skeletal fixation for long tubular bony defect by end to end compression with lengthening (EECL) and segmental bone transport by lengthening (STBL). METHODS: About 20% - 30% of the original length was resected from the middle shaft of the tibia in 10 goats of 1 - 2 years of age. The bony defect ends were shortened step by step with the sulcated half-ring external fixator. Changes of the blood flow of the distant extremity were measured with pulse-dopplar monitor, reference blood flow, and angiograph. RESULTS: The blood flow of the distant extremity was not affected when bony defect was less than 15% of the original length. Blood circulation disorder would appear in the distant extremity when bony defect ranged from 15% to 20% of the original length. The distant extremity would necrose because of obstruction of blood circulation when bony defect was more than 20% of the original length. CONCLUSIONS: EECL can be recommended if bony defect is less than 15% of the original length. SBTL might be the only feasible way if bony defect is more than 20% of the original length. If bony defect is within 15% and 20%, EECL should be applied with great care while watching the extremity circulation with pulse-Dopplar monitor.

[Activity of serum paraoxonase and its polymorphism in healthy Chinese of Shanghai].

OBJECTIVE: To study activity of serum paraoxonase and its polymorphic distribution in healthy people of Shanghai and to set a reference value for it. METHODS: Serum activities of paraoxonase, salt-stimulated paraoxonase and arylesterase were measured in 198 subjects (105 males and 93 female, aged 21 - 61 years) in Shanghai. Three phenotypes of serum paraoxonase could be distinguished based on the ratio of salt-stimulated paraoxonase activity and paraoxonase activity and the ratio of salt-stimulated paraoxonase activity and arylesterase. Its gene frequency was calculated according to Hardy-Weinberg equation. RESULTS: Activity of serum paraoxonase was 97.6 - 472.1 unit. There was no difference in serum paraoxonase activity between male and female, nor among different ages. It was found that 27 persons in low, 71 in medium and 100 in high activity. Gene frequency of low activity (PON * A) and high activity (PON * B) was 0.369 and 0.631, respectively. CONCLUSIONS: Distribution of serum paraoxonase in healthy people of Shanghai was different from that in Caucasians.

[Establishment and biological characteristics of a novel erythroleukemia cell line (HIE1)].

OBJECTIVE: To establish a novel leukemia cell line and characterize its biological characteristics. METHOD: The cell line was established by liquid cell culture. The genetic marker was analyzed by R-banding and reverse transcriptase-polymerase chain reaction (RT-PCR), cell morphology by microscopy, electron microscopy and histochemical staining, cell surface antigen by monoclonal antibody, hemoglobin by hyperomethemoglobin measurement and electrophoresis, erythroid differentiation by benzidine-staining, and monocyte-macrophage differentiation by cell morphology and phagocytosis. RESULTS: A novel erythroleukemia cell line (HIE1), with original cell genetic marker (Ph chromosome, bcr/abl fusion gene rearrangement), was established from a CML patient in blast crisis, and has been passaged for over 60 generations. Myelomonocyte marker and hemoglycoprotein A were found on the cell surface. HIE1 cells contained hemoglobin, the same HbA and HbA(2) bands as in normal individuals were displayed by Hb electrophoresis. The benzidine positive HIE1 cells were induced after exposure to 3.6 x 10(-4) mmol/L Ara-C. When HIE1 cells were treated with 100 ng/ml PMA for 3 days, one third of the cells became spindle in shape, and 6.5% of the cells exert phagocytosis. The cells were classified into two types with Wright-staining: one showing light blue cytoplasm and a few of cells with basophilic granules, the other showing dark blue cytoplasm with vacuoles and pseudopods without granules. In addition, POX, SB, CE stains were negative, and AE, PAS, ACP stains positive. Colony formation of the cells was 37%, the cell doubling time was 22 - 24 hrs, and EB virus detection was positive. CONCLUSION: A novel erythroleukemia cell line with bcr/abl fusion gene and characteristics of myelomonocytic and erythroid cells was established.

[Antiphospholipid thrombosis syndrome and acquired activated protein C resistance].

OBJECTIVE: To explore the correlation between antiphospholipid antibodies (APA) activated protein C resistance (APCR) and antiphospholipid thrombosis (APL-T) syndrome and investigate the mechanism of thrombosis in APL-T syndrome. METHODS: ELISA, APTT-LA and APTT+/-APC methods were used to detect anticardiolipin antibodies (ACA), lupus anticoagulants (LA) and APCR in 20 APL-T syndrome patients respectively. RESULTS: Twenty patients were diagnosed as APL-T. The patients were classified in three ways. According to etiology, 14 of 100 systemic lupus erythematosus (SLE) patients were diagnosed as "secondary" APL-T, and 6 of 16 "unexplained" thrombosis and spontaneous abortion as "Primary" APL-T syndrome. According to antibodies, 12 of 20 patients with positive LA were diagnosed as lupus anticoagulant thrombosis syndrome (LA-T), 12 of 20 patients with positive ACA as anticardiolipin antibodies thrombosis syndrome (ACA-T). According to clinical presentation: 7 of 20 patients with deep vein thrombosis were classified as type I APL-T, 3 of 20 patients with cerebrovascular thrombosis as type II, 1 of 20 patients with vein cerebrovascular thrombosis and spontaneous abortion as type IV (mixing), and 9 of 20 patients with spontaneous abortion as type V (fetal wastage type). The ACAs and LA are strongly associated with APL-T. There is close relationship between APCR and APL-T. CONCLUSION: The acquired APCR might be an important reason for thrombosis in APL-T caused by APA and LA.

[Detection of soluble endothelial protein C receptor (sEPCR) in patients with CHD, DM and SLE].

OBJECTIVE: To evaluate the clinical significance of plasma soluble endothelial protein C receptor (sEPCR) level in patients with coronary heart disease (CHD), diabetes mellitus (DM) and systemic lupus erythematosus (SLE). METHODS: ELISA of antibody-sandwiched principle was used to detect sEPCR in 34 patients with CHD, 42 with DM-II, 63 with SLE and 20 normal controls (NC). RESULTS: The plasma level of sEPCR in patients with DM was higher than that in NC group (P < 0.05). The sEPCR level in SLE patients with thrombosis and non-thrombosis are higher than that in NC group (P < 0.005 and P < 0.001, respectively). There is no significant difference between the two groups of SLE (P > 0.05), and between the CHD and NC groups. CONCLUSION: sEPCR level increased in DM and SLE, it seems related to the process of inflammatory reaction and damage of endothelial cell.

[Clinical and experimental study on treatment of bronchial asthma with pingchuan oral liquid].

OBJECTIVE: To observe the clinical effect of Pingchuan oral liquid, including Hanchuan oral liquid (HCOL) and Rechuan oral liquid (RCOL), and to explore its mechanism. METHODS: HCOL and RCOL were prepared based on the therapeutic principles of eliminate phlegm, remove blood stasis, lower reversed Qi flow and relieve asthma. HCOL was used to treat 50 patients of bronchial asthma in Group A and RCOL for 55 patients in Group B, and they were compared with 52 patients treated with Guilong Kechuanning capsule in the control group. The relevant animal experiment was also conducted. RESULTS: The total effective rate of HCOL was 88.0%, that of RCOL was 90.9%, they were significantly higher than that of the control (73.1%, P < 0.05). Moreover, the two new preparations showed the effects in easing main symptoms and signs of asthma, decreasing peripheral eosinophilic granulocyte count and immunoglobulin level, and improving pulmonary function superior to those of Guilong Kechuanning capsule (P < 0.05, P < 0.01). Experimental study showed that they could lower blood thromboxane B2 and endothelin, raise blood nitric oxide and 6-keto-prostaglandin F1 alpha. CONCLUSION: Pingchuan oral liquid is obviously effective in treating bronchial asthma, the mechanism possibly lays on adjusting immune function, anti-allergy and antagonizing inflammatory media.

[Ion channels in rat pancreatic beta cells].

Using perforated and cell-attached patch clamp techniques, the characteristics of ATP sensitive K(+) channels (K(ATP)), delayed rectifier K(+) channels (K(DR)), Ca(2+) and Na+ channels on single rat pancreatic beta cell membranes were studied. The results showed that (1) the efflux and influx conductance of K(ATP) channels was about 31 and 65 pS respectively, and the reversal potential of K(ATP) was about 60 mV; (2) K(DR) was activated completely after a latency of 20 ms, and K(DR) was about 1/3 of K(ATP); (3) whole cell Ca(2+) current reached a peak (40 60 pA) at 0 mV; L-type Ca(2+) channel was the main Ca(2+) channel in beta cells, but other types of high voltage activated Ca(2+) channels existed as well; and 4) whole cell Na(+) current reached a peak (200 400 pA) at 10 mV; but the expression level of Na(+) channel in beta cells varied among the cells. About half of the beta cells virtually had no Na(+) currents.

[Changes of lipoprotein lipase and hepatic lipase and their significance in gallstone formation in rabbit model].

This experiment was made to investigate the changes of lipoprotein lipase(LPL) and hepatic lipase (HL) activity and their effects on gallstone formation in rabbit model in which the stones were induced by high cholesterol diet. Activities of plasma LPL and HL were determined; other data including concentration of plasma lipoprotein cholesterol, concentration of bile cholesterol and bile acids were also obtained. The results showed that with the rabbits continuously fed on high cholesterol diet, LPL activity heightened markedly (P < 0.05), and HL activity increased gradually (3 and 4 weeks groups vs control group, P < 0.05). The changes of concentration of plasma VLDL-C and LDL-C were the same as that of LPL activity, but the concentration of plasma HDL-C, HDL2-C, HDL3-C and bile acids showed no significant changes (P > 0.05). These results suggest that the heightened activities of LPL and HL might make the liver take up more cholesterol, secrete it into the bile duct and hence accelerate gallstone formation.

[Study on IGF-I regulation the proliferation of rat condylar chondrocytes in vitro].

This study was aimed at the mechanism for the changes in the proliferation of cultured rat mandibular condylar chondrocytes. Flow cytometry was used to explore the changes of cellular DNA content and cell cycles of passaged condylar chondrocytes. The results showed that when the mandibular condylar chondrocytes were cultured with IGF-I for 24 hours and 96 hours, the cellular proliferation in the experimental group enhanced higher than that in the control group. It was implied that IGF-I may be a mitogen on rat mandibular condylar chondrocytes. The mitogenic effect of IGF-I had a lag period.

[Construction of a novel Bm NPV Bac to Bac system].

A Bi-Shuttle vector Bm-Bacmid was constructed by co-infecting Bm N cells with wild type genomic DNA from BmNPV and Ac-Bacmid DNA. It could not only replicate in E. coli cells as a large plasmid and but also remain infectious when induced into Bm N or Sf9 cells. Recombinant virus rBmHBe was obtained after transposition of a donor plasmid carrying Hepatitis Be antigen gene (HBeAg) into att Tn7, and was demonstrated by Southern blotting. SDS-PAGE analysis showed that HBeAg gene were highly expressed in Bm N cells. By ELISA testing, the highest antigenecity titer of HBeAg protein in cell cultural medium was up to a dilution of 1:32,000. Although HBeAg protein also presented in the Bm N cells the titer was only 1:2000. The HBcAg protein was much fewer than HBeAg (< 1:160) whatever in culture medium and in cells. The results showed that Bm N cells was able to recognize the signal peptide sequence and cut it correctly for HBeAg protein's excreting production.

[Studies on the toxicity of polymer of glycolide and lactide(PLGA) and the anti-fertility effect of levonorgestrel(LNG)].

PLGA(9:1), a chemical degraded from biomaterials, has been subjected to the tests of acute toxicity, irritations on conjunctiva and muscle, haemolysis in vitro and allergic reaction of mice and rats. The anti-fertility effect of PLGA-LNG microphere composite on the same animals also has been tested. The studies demonstrated that PLGA had fine biological compatibility without side effects such as toxicity, irritations on conjunctiva and muscle, pyrogenicity, haemolysis in vitro and allergic reaction. The contraception period of mice lasted 11.5, 34.6 and 81.2 days respectively after the subcutaneous (s.c.) injection of PLGA-LNG microphere which contains LNG 5, 10 and 20 mg/kg separately. The intramuscular(i.m.) injection of PLGA-LNG microphere containing LNG 13.6, 19.8 and 30 mg/kg respectively could keep the estrus of rats supressed for 76.3, 104.1 and 131.8 days. The injections of PLGA-LNG microphere through s.c. or i.m. showed more significant effects than the same dosages of PLGA-LNG micro-crystal. These results suggest that PLGA-LNG microphere injection has long-term anti-fertility effect.

[Histological study of the local-regional invasion of pyriform sinus carcinoma].

OBJECTIVE: In order to offer the basis for the operation of the pyriform sinus carcinoma, the local-regional invasion patterns of the pyriform sinus carcinoma were investigated. METHODS: Twenty-six surgical specimens obtained from patients who undergone laryngopharyngectomy were subjected to a whole organ section study. RESULTS: Tumors located in the lateral wall of the pyriform sinus spread mainly towards the lateral wall of the hypopharynx. Tumors located in the inner wall of the pyriform sinus spread mainly towards the larynx and opposite side pyriform sinus, the paraglottic space and thyroid cartilage were particularly frequent invaded. It was rare of the invading of the cricoid cartilage. The invasion of the epiglottis and preepiglottic space across the midline had not been found. There were two invading routines, the paraglottic space and preepiglottic space. The invasion of the paraglottic space was through the aryepiglottic fold and the inner surface of the thyroid cartilage. The invading of the preepiglottic space was through the aryepiglottic fold and the upper part of the inner surface of the thyroid cartilage. CONCLUSIONS: The invading of the preepiglottic space was not the contraindication of the partial laryngectomy. It is practicable to preserve the laryngeal function in most of the patients while tumors located on the lateral or inner wall of the pyriform sinus. There was a tendency to the invading to the contralateral side in the inner wall of the pyriform or postcricoidarytenoid region. Attention should be paid to the submucosal spreading in the postcricoidarytenoid region.
