[Transsphenoidal microscopic removal of invading pituitary adenoma].

Eighteen cases of invading pituitary adenoma were removed via the approach of transsphenoidal microscopical operation. Among them 4 cases had insulin dependent diabetes. The pituitary adenoma were near-totally removed in 15 cases, partially removed in 3, and 10 cases were treated by supplement radiotherapy postoperatively and 5 cases by oral bromocriptin postoperatively. The average time of follow-up was 3.8 years. There were 3 cases(16.6%) had postoperative recurrence of the tumor which were finally cured by transfrontal craniotomy. The feasibility of the transsphenoidal microscopic approach for the invading pituitaty adenoma was discussed.

[The effects of menthol, borneolum and moschus on nasal airflow sensation and nasal resistance].

Menthol and borneolum have been used widely in traditional Chinese medicine for the treatment of nasal obstruction, but the mechanism has been still unknown. The effects of inhalation of menthol, borneolum, moschus on nasal resistance and airflow sensation were investigated in 52 subjects. All of these medicines could cause a highly significant enhancement of nasal airflow sensation but had no effect on nasal resistance. Therefore this sort of medicine is regarded as pseudo nasal decongestant.

[Scanning electron microscopic changes of maxillary sinus mucosa before and after functional endoscopic sinus surgery].

We attempted to evaluate postoperative maxillary sinus mucosal changes in 20 maxillary sinuses of 16 patients who underwent functional endoscopic sinus surgery (FESS). Ciliated area of the maxillary sinus mucosa and ostium of maxillary sinus were quantitatively observed by scanning electron microscope and image analysis before and after FESS. In the present study, the number of preoperative ciliated cells of maxillary sinus mucosa and ostium was significantly reduced, goblet cells and microvillous cells were increased, and squamous cell metaplasia observed in part of the maxillary sinuses. Many damaged ciliated cells tend to recover and ciliated area significantly increased at 6-12 months after the surgery. The results showed that the damaged ciliated epithelium could return to normal by the improvement of ventilation and drainage of the maxillary sinus following FESS.

[The effect of antibody against vascular endothelial growth factor (VEGF) on tumor metastasis].

OBJECTIVE: To study whether manipulation of angiogenesis with antibody against vascular endothelial growth factor (VEGF) may interrupt the tumor growth and metastasis. METHODS: IV TA2MA 891 is a murine spontaneous breast cancer with high rate of metastasis in lung in (TA2x615) F1 mice, and it was chosen as an animal model in this study because of the high expression of VEGF in primary tumor as well as in lung metastatic tumor. The mice were injected with anti-VEGF polyclonal antibody at a dose of 200 micrograms/mouse/day from day 9 to 23 following s.c. tumor inoculation and sacrificed on day 26. RESULTS: The growth of primary tumor was inhibited by 44.0% and the number and size of metastatic foci in the lungs were inhibited by 73.0% and 83.7% respectively. CONCLUSION: The antibody against human VEGF may be a potentially effective antimetastasis agent for the treatment of patients with lung metastasis.

[Prognosis of premenopausal patients with operable breast cancer related to the timing of operation].

OBJECTIVE: Determining the fact if the timing of mastectomy during the menstrual cycle influences the prognosis of premenopausal women with breast cancer. METHODS: 67 cases of breast cancer women with regular menstruation were reviewed and analyzed retrospectively by medical statistics in terms of 5-year overall survival, 5-year relapse-free survival and 5-year local-regional relapse survival. RESULTS: Even though the 5-year overall survival and 5-year local-regional relapse survival were not different statistically, timing of operation did have a tendency to influence outcome. Meanwhile, estrogen receptor played the most important role in influencing the above-mentioned aspects in comparison with other factors. CONCLUSION: We believe that operation during the last menstrual period might have influence on long-term survival of premenopausal breast cancer patients, but prospective studies are needed.

[A research on real-time ventricular QRS classification methods for single-chip-microcomputers].

Ventricular QRS classification is key technique of ventricular arrhythmias detection in single-chip-microcomputer based dynamic electrocardiogram real-time analyser. This paper adopts morphological feature vector including QRS amplitude, interval information to reveal QRS morphology. After studying the distribution of QRS morphology feature vector of MIT/BIH DB ventricular arrhythmia files, we use morphological feature vector cluster to classify multi-morphology QRS. Based on the method, morphological feature parameters changing method which is suitable to catch occasional ventricular arrhythmias is presented. Clinical experiments verify missed ventricular arrhythmia is less than 1% by this method.

Radioimmunoimaging of human breast cancer xenografts in nude mice by using human anti-human breast cancer monoclonal antibodies.

In this study, the human breast cancer-bearing nude mice model has been established and the radioimmunoimaging was carried out by using human anti-human monoclonal antibodies. The results showed that the breast tumor-bearing nude mice received 131I-McAb-CM-1 had a clear image of the xenograft during 4 to 6 days after injection and at the same time T/NT all over 1.0. The highest T/NT reached 7.1. It demonstrates that McAb-CM-1 can specially combine with breast cancer tissues and hopefully it could be used clinically to improve accurate rate of the early breast cancer diagnosis.

[Multi-function ECG pre-amplifier for portable monitor].

In this paper, an ECG pre-amplifier portable monitor is given, which has many functions, i.e., 50 Hz notch, lead-off alarm and pacer pulse inhibition. The amplifier only consists of 3 chips of 8 pin ICs, and consumes current less than 1.4 mA.

[Cultivated technique of high yield for Polygonatum officinale].

By studying cultivated materials, hided crops, cultivated way, applied fertilizer about the cultivation of Polygonatum officinale, put forward following the methods of cultivation of P. officinale. Choose rhizome segments with top buds as plant materials, adopt level rectangular pieces of land in a field width 100 cm, cultivate late-maturing corn as hiding shady, adopt density of 1.5-2.0 ten thousand plants/mu, apply N and P fertilizers in the growth.

[Dynamic changes of serum antibody titer and liver ultrastructural pathology in acute sporadic hepatitis E virus infected patients].

7 acute hepatitis E patients were tested for anti-HEV IgM and IgG antibodies by EIA. It revealed that anti-HEV IgM and IgG antibody titers were the highest at day 10-45 of the illness, and anti-HEV IgM declined gradually and totally lost within 2 months. Anti-HEV IgG had a similar condition to anti-HEV IgM, but at 7th month there still had 3 cases (40%) remained to be antibody positive. One case was examined for ultrastructural pathology. The swelling of liver cells on the fortieth day of illness was still to be found, the vacuolation of cytoplasm and condensation of mitochondria were present in liver cells.

[Serum G-CSF levels in patients with chronic aplastic anemia].

OBJECTIVE: To explore the pathogenesis of aplastic anemia. METHODS: The spleen cells of BALB/c miceimmunized with rhG-CSF were fused with myeloma cell line SP2/0. Three murine hybridomas producing monoclonal antibody (McAb) against rhG-CSF were obtained. Serum levels of G-CSF in 54 cases of chronic aplastic anemia (CAA) were detected by ELISA method. RESULTS: Serum G-CSF levels in 70.4% (38 of 54 cases) of the CAA patients were increased (272.76 +/- 58.39ng/L). There was a negative correlation between blood neutrophil counts and serum G-CSF levels (r = -0.535, P<0.01). CFU-GM were detected in 12 cases of CAA, and all was decreased (17.92 +/- 10.28/2 x 10(5) nucleated cell). Serum G-CSF levels in almost 30% (12 of 54 cases) of the CAA patients were not increased. CONCLUSION: The hematopoietic progenitor cells of CAA patients were impaired, and some CAA could be treated with rhG-CSF and immunosuppressive agents.

[Determination of demethylvancomycin in neonate serum by high performance liquid chromatography].

A rapid high performance liquid chromatography for determination of demethylvancomycin in neonate serum has been developed. The procedure involved a simple protein precipitation by acetonitrile-isopropanol (1:1) and then the sample was chromatographed on a reversed phase C18 column with UV detection at 236 nm. The mobile phase was CH3CN: 0.05 mol/L KH2PO4 = 8:92 (V/V). The calibration curve was Y = 35,721.89X - 13,031.54, r = 0.9998 and he detection limit was 0.3 mg/L. The average recovery was 94.7% +/- 1.2%. Intra-day and inter-day RSD were 2.23% and 2.62% respectively. It can be concluded that this method meets the requirement for routine clinical application. This method has been used to determine serum concentration of demethylvancomycin in neonates. The data obtained showed that the method was simple, rapid, sensitive and precise.

[Fine physical mapping of yeast chromosome V].

Electrophoretic karyotype of yeast strain A364a was obtained by pulsed field gel electrophoresis and the position of chromosome V on such karyotype was determined by means of dot hybridization with chromosome V-specific probe URA3. By cloning partially digested BamHI fragments of this chromosome DNA into integrative vector Yip5, a gene library specific to this chromosome was constructed. The number of the recombinants was much more than theoretically required. After screening probe-homologous fragments from this library and analysing such fragments with restriction enzymes BamHI, EcoRI, HindIII, PstI, and SalI, a fine physical map covering about 9.4% of A364a chromosome V (which was estimated as 620kb) was constructed. Further colony hybridization with boundary clones will enable us to "walk" throughout the whole chromosome.

Potential new insights into the molecular mechanisms of methamphetamine-induced neurodegeneration.

In the event that methamphetamine evokes HO. formation within serotonergic axon terminals, the resultant oxidation of 5-HT would be expected to generate not only 5,6-DHT but also T-4,5-D, 7-S-Glu-T-4,5-D, 6, 8, and 7,7'-D (figure 1), at least three of which (T-4,5-D, 7-S-Glu-T-4,5-D, and 6) are lethal in mouse brain. Furthermore, several intermediates/products formed in the in vitro oxidation of 5-HT by HO. are readily autoxidized (4,5-DHT, 5,6-DHT, 5, 7, and 9) or redox cycled (T-4,5-D, 6, 8, 7,7'-D, 7-S-Glu-T-4,5-D) in reactions that would be expected to yield O2-. and/or H2O2 as byproducts. These byproducts, in the presence of trace levels of transition metal ion catalysts, would be readily converted into HO. (Walling 1975; Halliwell and Gutteridge 1984). Together these putative aberrant oxidative metabolites of 5-HT and HO.-forming reactions might contribute to the degeneration of serotonergic nerve terminals. Similarly, the methamphetamine-induced intraneuronal formation of HO. in dopaminergic terminals might be expected to generate not only 6-OHDA (and 2-OHDA and 5-OHDA, figure 3) but also 5,-S-CyS-DA and 5-S-Glu-DA, precursors of DHBT 17 and other more complex dihydrobenzothiazines (figure 4). DHBTs 17 to 19 are lethal in mouse brain, although at this time the biochemical/chemical mechanisms underlying this toxicity and specific neuronal systems affected are unknown. However, 5-S-CyS-DA and 17 to 19 are much more easily oxidized than DA, and the latter DHBTs appear to be capable of redox cycling reactions (Zhang and Dryhurst 1994). Thus, the HO.-mediated oxidation of DA in dopaminergic nerve terminals induced by methamphetamine might be expected to generate aberrant oxidative metabolites that (as a result of autoxidation and redox cycling reactions) potentiate formation of O2-. and/or H2O2, and then HO. and neuronal damage. A number of lines of evidence, discussed previously, suggest that aberrant metabolite(s) of DA (other than or in addition to 6-OHDA) might contribute to the methamphetamine-induced degeneration of not only dopaminergic terminals but also serotonergic terminals. Similarly, aberrant metabolite(s) of 5-HT (other than or in addition to 5,6-DHT) might be involved in the degeneration of serotonergic and dopaminergic terminals and a subpopulation of cell bodies in the somatosensory cortex. Experimental evidence indicates that some of the neurodegenerative effects evoked by methamphetamine are mediated by NMDA and GABA receptors. Thus, it will be of considerable interest to investigate the neurotoxicity of putative aberrant oxidative metabolites of 5-HT (figures 1 and 2) and DA (figures 4 and 5) towards serotonergic, dopaminergic, and other neuronal systems and their interactions with NMDA, GABA, and other brain receptors. A central question relates to mechanisms by which methamphetamine might evoke the intraneuronal formation of oxygen radicals that appear to play important roles in the overall neurodegenerative processes evoked by this drug (DeVito and Wagner 1989; Cadet et al. 1994). Once putative oxidative metabolites of 5-HT such as T-4,5-D, 7-S-Glu-T-4,5-D, 5,6-DHT, 6, 8, and 7,7'-D (figure 1) are formed intraneuronally, autoxidation/redox cycling reactions should, in principle, be capable of generating O2-. and/or H2O2, the precursors of HO.. Similarly, intraneuronal formation of 6-OHDA, 5-S-CyS-DA, and DHBTs 17 to 19 and 22 would also be expected to potentiate elevated fluxes of O2-., H2O2, and HO. as a result of the facile autoxidation/redox cycling reactions of these putative aberrant metabolites. The presence of very low concentrations of 5-S-CyS-DA in DA-rich regions of human and other mammalian brains suggest that autoxidation (Rosengren et al. 1985; Fornstedt et al. 1986, 1989, 1990) or perhaps some other form of DA oxidation is a normal reaction in vivo. Furthermore, available evidence suggests that it is cytoplasmic DA that is oxidized to give 5-S-CyS-DA (Fornstedt et al. 1989; Fornstedt and

Kallikrein gene therapy in newborn and adult hypertensive rats.

The tissue kallikrein-kinin system has been postulated to play an important role in blood pressure regulation. Kallikreins are serine proteinases that release potent vasodilating kinin peptides from precursor kininogens by limited proteolysis. Our recent studies show that systemic delivery of the human tissue kallikrein gene into adult spontaneously hypertensive rats (SHR) results in a sustained reduction of blood pressure for several weeks. The goal of this study is to evaluate whether early delivery of the kallikrein gene into newborn SHR could exert a suppressive effect on blood pressure phenotype during rat growth and development. A human tissue kallikrein cDNA construct, under the control of cytomegalovirus promoter (CMV-cHK), or vector DNA was injected subcutaneously into the necks of 2-day-old SHR. Blood pressures were monitored biweekly from 3 to 19 weeks by the tail-cuff method. A single injection of the human kallikrein cDNA construct caused a significant reduction of blood pressure (n = 6, p < 0.001) from 11 to 17 weeks after injection compared with control rats receiving vector DNA. Intravenous delivery of the human tissue kallikrein gene into adult SHR produced blood pressure lowering effects (n = 6, p < 0.001) that lasted for 6 weeks in male but not in female rats. The expression of human tissue kallikrein in rats was identified by reverse transcription polymerase chain reaction followed by Southern blot analysis and an ELISA specific for human tissue kallikrein. Kallikrein gene delivery did not cause any changes in body weight, urine volume, or water intake in the experimental animals compared with the control group. No antibodies to either human tissue kallikrein or its DNA were detected in rat sera 19 weeks postinjection. These results show that delivery of the kallikrein gene at an early stage of life has a protective effect against development of hypertension in adult SHR and that gender differences could be a factor in kallikrein gene therapy for the treatment of hypertensive disorders.
