[A study of gene products encoded by Wilson disease gene].

OBJECTIVE: To investigate the pathogenesis of Wilson disease (WD) by detecting its gene products encoded by WD gene (ATP7B). METHODS: Patients diagnosed as WD were analyzed by SDS-PAGE in conjunction with Western blot. Two antibodies were used, which are specially against the sixth copper binding domain (Anti-CuBD) and ATP binding domain (Anti-ABD) of WD protein. RESULTS: The WD proteins were not expressed in two patients when using anti-CuBD, and poorly expressed when using anti-ABD. CONCLUSIONS: WD is highly heterogeneous in clinical manifestations and inheritance pattern. Two WD patients might simultaneously have exon 5 mutations and exon 8 mutations. The study of WD gene products would probe into the pathogenesis of WD.

[A study of Wilson's disease gene encoded products and gene mutations].

OBJECTIVE: To investigate the pathogenesis of Wilson's disease(WD). METHODS: Hepatocytes were isolated from WD patients' bioptic hepatic samples and cultured in vitro; WD proteins, the gene putative encoded products, were detected by SDS-PAGE in conjunction with Western blotting in liver samples of three patients and two controls. Their genomic DNAs were analyzed by means of direct DNA sequencing of WD gene (ATP7B) on exon 8. RESULTS: The WD proteins lanes from two WD patients were found to be much weaker than that from the control, from which one WD patient was proven as heterozygote of 778 position CGG-->CTG(Arg778Leu) and 770 position CTC-->CTG change of ATP7B. CONCLUSION: WD is highly heterogeneous in clinical manifestations and inheritance pattern. Abnormally expressed putative WD proteins in WD patients might be the results of ATP7B mutations, and the study of ATP7B products would help to probe into the pathogenesis of WD.

[Effect of radiation with 60Co on RBC membrane elastic shear modulus and membrane viscosity].

RBC membrane shear elastic modulus and membrane viscosity are two important indexes reflecting RBC membrane viscoelasticity. Their variation was investigated in this study after rabbits were radiated with 60Co. With a new ektacytometer, we measured the small deformation index (DId) and the half-time of deformation relaxation (t0.5) of RBC in flow field then we calculated RBC membrane shear elastic modulus and membrane viscosity. We found that the value of RBC membrane shear elastic modulus and membrane viscosity continuously increased from 0 to 16th day then continuously decreased and tended to be stable on 60th day or so. The reason may lie in the variation of proportion of new and old RBC in blood and variation of microconformation of RBC membrane after rabbits were radiated with 60Co.

[A study of association between atherothrombotic brain infarction and HLA-DQA1 peptide binding motifs].

OBJECTIVE: To analyze the hereditary susceptibility for HLA-DQA1 alleles according to trait of multiple genes leading to disease in atherothrombotic brain infarction (ABI). METHODS: HLA-DQA1 alleles in ABI patients (n=81) and healthy controls (n=99) were detected by PCR-SSP techniques. RESULTS: (1)The frequency of HLA-DQA1*0301 was significantly higher in ABI patients. (2)The frequency of HLA-DQA1*0301 was significantly higher in patients with family history of essential hypertension (EH). (3)The frequency of HLA-DQA1*0103 was significantly lower in ABI patients. CONCLUSION: HLA-DQA1*0301 allele might be a correlative gene with hereditary susceptibility of ABI and HLA-DQA1*0301 allele might be a correlative gene in ABI patients with the family history of essential hypertension, whereas HLA-DQA1*0103 allele might be a protective gene in the patients with ABI.

[Micropore filters for measuring red blood cell deformability and their pore diameters].

Micropore filters are the most important components in micropore filtration testes for assessing red blood cell (RBC) deformability. With regard to their appearance and filtration behaviors, comparisons are made for different kinds of filters currently in use. Nickel filters with regular geometric characteristics are found to be more sensitive to the effects of physical, chemical, especially pathological factors on the RBC deformability. We have critically reviewed the following viewpoint that filters with 3 microns pore diameter are more sensitive to cell volume than to internal viscosity while filters with 5 microns pore diameter are just the opposite. After analyzing the experiment results with 3 microns and 5 microns filters, we point out that filters with smaller pore diameters are more suitable for assessing the RBC deformability.

[A randomized, double-blind, placebo-controlled, multicenter study on sibutramine in over-weighted and obese subjects].

OBJECTIVES: To assess weight loss efficacy, safety and tolerability of sibutramine in simple obese subjects. METHODS: Randomized, double-blind, placebo-controlled clinical trial. Four hospital outpatient clinics in Shanghai, Chongqing, Shandong and Tianjin, respectively. PARTICIPANTS: 233 men and women, 18-65 years old, with body mass index (BMI) ranging from 27 to 40 kg/m2 were randomly divided into an intervened group and a placebo control group. Sibutramine 10 mg or placebo once a day. MAIN OUTCOME MEASURES: Body weight, routine laboratory and clinical safety monitoring. RESULTS: Of 233 eligible patients, 120 received sibutramine and 113 placebo. Weight reduction was significantly greater in the intervented group (6.8 +/- 3.1) kg than the placebo control group (0.48 +/- 2.6) kg from week 4 onwards to week 24 (P < 0.001). Some minor side effects were noticed in the subjects who took sibutramine. But the symptoms were light and short term. Sibutramine was well tolerated. CONCLUSIONS: Sibutramine 10 mg once a day is an effective and safe therapy for weight reduction in simple over-weighted and obese subjects.

[Imaging evaluation and interventional therapy of hepatic metastases from small intestinal leiomyosarcoma].

OBJECTIVE: To evaluate the imaging appearances and the short-term curative effect of hepatic metastases from small intestine leiomyosarcoma treated by interventional methods. METHODS: Seven cases with 46 lesions of hepatic metastases from small intestinal leiomyosarcoma were analyzed retrospectively. The diameters of the metastases were from 2 cm to 18 cm. CT triphasic scan and hepatoarterial angiography were performed in all cases. The interventional therapy methods included transcatheter arterial chemoembolization (TACE) (all 7 cases), percutaneous left subclavian artery port-catheter system implantation (PCS) (1 case). Three cases were also treated with percutaneous tumor necrosis drainage (PTND) because of obvious liquefaction necrosis within the tumor. RESULTS: The lesions were substantial occupation and some were mixed density with cystic component. The cystic component increased in the larger tumor. The hepatic metastases were enhanced remarkably during the hepatic arterial-dominant phase scan and had abundant tumor vascular and tumor stain on hepatoarterial angiography. The size of tumor in 4 cases was reduced after TACE. The tumor in the patient who was treated with PCS had no changed. The size of the tumor in 3 cases was slightly enlarged. The tumors in the 3 patients treated with PTND were reduced remarkably. CONCLUSIONS: The predominant blood-supply of the hepatic metastases from small intestinal leiomyosarcoma is the hepatic artery abundant in blood vessels. TACE, PCS and PTND are effective methods to treat hepatic metastases from small intestinal leiomyosarcoma.

Effect of 60Co irradiation on characteristics of hemorheology in rabbits.

A high-dose (7 Gy) whole-body 60Co irradiation for a short period caused disturbances of hematopoietic function. A decrease in the hematocrit of the circulating blood lasted for about 15 days, thus forming an anemic animal model. We studied the influence of high-dose 60Co irradiation on hemorheologic parameters: percentage of reticulocytes, RBC deformability, sedimentation rate and plasma fibrinogen concentration in the rabbit. It was found that the plasma fibrinogen concentration increased to twice more than control level and that percentage of reticulocytes in circulation disappeared immediately after irradiation. The deformation index of RBCs in shear flow decreased from a value of 58% down to a value of 42% in the first two weeks and gradually returned to control levels about 40 days after 60Co irradiation. Our results showed that a short period of high-dose 60Co irradiation caused severe and relatively long-lasting damage of hematopoietic system in animals' body.

[The expression of red blood cell deformability in micropore filtration tests].

For the purpose of evaluating and comparing RBC deformability quantitatively with micropore filtration method, the precise meanings and applicability of physical parameters and defined indices expressing RBC deformability are discussed and analyzed in detail. The mean transit time (or the relative transit resistance) and the proportion of pore-clogging cells are found to be appropriate for representing the average deformability and the clogging characteristic of RBCs respectively. To eliminate the effects of clogging on filtration, previous studies were conducted filtration with as less clogging as possible. Investigations on experimental results from various filtration methods, however, reveal that the clogging property of RBCs contains important information on the RBC deformability. It reflects a different aspect of the RBC deformability besides the average deformability and is more sensitive to the abnormal deformability induced by physical, chemical, especially pathological factors.

[The design and evaluation of horizontal pipe mini-flame atomization and ionization synchronous detector in GC/AAS].

Gas chromatography/atomic absorption spectroscopy(GC/AAS) is a good method for the species analysis of organometallic compounds. But the traditional atomizers are not very suitable for this technology and all of them response only to one kind of signal--the concentration of the metallic atoms of the ground state. They can not give any information about the organic group of organometallic compounds and organic compounds which coexist with the former. For GC/AAS we want to design and manufacture a new kind of detector which is much more sensitive and has a much smaller dead volume and will sensitively and synchronously response to the atomization signal of organometallic compound and ionization signal of organic compound. The authors have noticed that the atomization of organometallic compound and ionization of organic compound have been existing in the same hydrogen flame. The question is how to gain and exchange and output the two signals which are completely different in characters. For this purpose we designed and manufactured a new type of horizontal pipe mini-flame atomization and ionization synchronous detector. The key part is a T type glass tube (80 mm x 13 mm x 10 mm i.d.) which covers horizontally on the jet of the mini-flame atomizer and a long pipe stainless steel collector (70 mm x 9.5 mm o.d. x 9 mm i.d.) is tightly inserted in the tube. The light beam of the hollow cathode lamp passes through the hydrogen flame along the axial center of the glass tube and the ground state metallic atoms in the flame diffuses to both ends of the tube along the axial center of the glass tube too. This process enriches the depth of the light absorption and then increases the sensitivity of atomization. At the same time the long pipe collector can enrich the collection efficiency of the ionization and increases the sensitivity and widens the linear range. So the detector can synchronously and sensitively detect the organometallic and organic compounds. The detection limit for atomization of the organometallic compound(diethylmercury) is 2.5 x 10(-11) g.s-1 and for ionization of the organic compound (benzene) is 1.0 x 10(-11) g.s-1. The linear range is 7.0 x 10(2) for atomization of diethylmercury (r = 0.9989) and 2.5 x 10(5) for ionization of benzene (r = 0.9992). The selectivity of the detector to the organic solvent under the optimum operating condition of atomization is 10(4)-10(7). The relative standard deviation (RSD, n = 11) of the peak area in the optimum operating conditions of atomization for the organometallic compound (diethylmercury) and of ionization for the organic compound (benzene) is 1.8% and 1.5% respectively. Under the same operating condition, the atomization and the ionization can not reach the optimum performance at the same time. If the atomization is in the optimum performance state, the performance for ionization is like a chromatographic thermal conductivity detector (TCD). The optimum performance of the atomization and ionization can be quickly adjusted by changing the flow rates of the corresponding gases such as H2, N2 and air.

[Diversity of metabolites and their bio-activities in myxobacterium Sorangium cellulosum].

Different Sorangium cellulosum strains not only showed diversity in their cell and fruiting body morphologies, but also differences of bio-activities and components of the metabolites. All the Sorangial strains studied in this paper had no activity on Gram-negative bacteria, some were able to repress Gram-positive bacteria. However, all the strains were able to repress growth of fungi and tumor cells strongly and widely. Thin layer chromatography assay of metabolites showed multi-components in the metabolites, and most of them have abilities of repressing fungi and tumor cells in different degrees. Four strains were found to be able to produce compounds with activity of promoting polymerization of microtubule. Based on Rf value on TLC, the bio-active component produced by So33-1 strain was similar to Epothilone A, while that of So81 was much different. The results of this paper suggested that Sorangium cellulosum is a much beneficial resource for screening nature compounds with bio-activities against eukaryotes.

[Preliminary study on isolation, purification and hydrolytic activity of cysteine proteinases in Entamoeba histolytica].

OBJECTIVE: To explore the invading mechanism of amebae in lamina porpria and observe the interaction between the cysteine proteinase (CP) of Entamoeba histolytica and laminin. METHODS: CP was identified by laminin-sepharose affinity chromatography, followed by isolation, purification and inhibitor experiment. The hydrolytic activity was measured by gelatin electrophoresis. RESULTS: Purified CP of E. histolytica showed a strong affinity with laminin. The molecular weight of CP is 27 kDa. It can be inhibited by EC-64 and exhibited a protein hydrolytic activity. CONCLUSION: The specific affinity and hydrolytic activity of CP might play an important role in its invasion to the basement membrane of intestinal mucosa.

[Cytotoxic effect of Acanthamoeba trophozoite on HeLa cells].

OBJECTIVE: To investigate the cytotoxic effect(CTE) on human cervix cancer HeLa cells induced by five strains of pathogenic free-living Acanthamoeba. METHODS: The cytotoxic effect of five isolates of Acanthamoeba on HeLa cells was investigated by light microscopy and MTT method. RESULTS: The photomicrographs of HeLa cells showed a sequence of cardinal morphological features of apoptosis when HeLa cells were exposed to Acanthamoeba in a time-dependent manner at a ratio of 1:1 for 12 h. MTT method showed more than 50% of tumor cells underwent cytolysis following exposure to A. lugdunensis trophozoites, and only 18% of cells treated with A. polyphaga underwent CTE. The CTE produced by A. lugdunensis and A. quina trophozoites was more rapid than the others, beginning as early as 6 h after coincubation and resulting in cytolysis by 72 h. CONCLUSION: These five strains of Acanthamoeba exhibit cytotoxic effects of varying degrees on HeLa cells, inducing apoptosis.

Frequent amplification of the telomerase reverse transcriptase gene in human tumors.

Activation of telomerase is a crucial step during cellular immortalization and malignant transformation of human cells and requires the induction of the catalytic component, human telomerase reverse transcriptase (hTERT), encoded by the hTERT gene. It is poorly understood how the hTERT gene is activated in human cancer cells. In the present study, we examined the hTERT gene copy number in human cancer cell lines and in primary tumor tissues. Amplification of the hTERT gene was observed in 8 of 26 (31%) tumor cell lines and 17 of 58 (30%) primary tumors examined (8 of 21 lung tumors, 3 of 10 cervical tumors, 5 of 19 breast carcinomas, and 1 of 8 neuroblastomas). In addition, 13 of 26 (50%) cell lines and 13 of 58 (22%) primary tumors displayed gain of hTERT gene copies with 3-4 copies/cell. The present findings imply that the hTERT locus may be a frequent target for amplification during tumorigenesis and that this genetic event probably contributes to the dysregulation of telomerase activity occurring in human tumors.

Characterization of the repressor function of the nuclear orphan receptor retinoid receptor-related testis-associated receptor/germ cell nuclear factor.

Retinoid receptor-related testis-associated receptor (RTR)/germ cell nuclear factor is a nuclear orphan receptor that plays an important role in the control of gene expression during early embryonic development and gametogenesis. It has been shown to repress transcriptional activation. In this study, we further characterize this repressor function. We demonstrate that RTR can suppress the transcriptional activation induced by the estrogen receptor related-receptor alpha1 through its response element. The latter is at least in part due to competition for binding to the same response element. In addition, RTR inhibits basal transcriptional activation, indicating that it functions as an active repressor. Mammalian two-hybrid analyses showed that RTR interacts with the co-repressor nuclear co-repressor (N-CoR) but is unable to interact with the co-repressor SMRT or RIP140. Pull-down analyses with glutathione S-transferase-RTR fusion protein demonstrated that RTR physically interacts with N-CoR in vitro, suggesting a potential role for N-CoR in the transcriptional repression by RTR. To identify the regions in RTR essential for the binding of RTR to N-CoR, the effect of various deletion and point mutations on this interaction was examined. This analysis revealed that this interaction requires the hinge domain, helix 3 as well as the helix 12 region of RTR. The residues Ser(246)-Tyr(247) in the hinge domain, Lys(318) in helix 3, and Lys(489)-Thr(490) in helix 12 are identified as being critical in this interaction. Our results demonstrate that RTR can function as an active transcriptional repressor and that this repression can be mediated through interactions with the co-repressor N-CoR. We show that this interaction exhibits several characteristics unique to RTR. Through its repressor function, RTR can suppress the induction of transcriptional activation by other nuclear receptors. These repressor activities may provide important mechanisms by which RTR regulates gene expression during development and gametogenesis.

Dietary arginine alters time of symptom onset in Huntington's disease transgenic mice.

Recent neuroimaging studies reported complex changes in cerebral blood flow (CBF) in early-staged Huntington's disease (HD) patients. Deckel and co-workers [Deckel and Duffy, Brain Res. (in press); Deckel and Cohen, Prog. Neuro-Psychopharmacol. Biol. Psychiatry 24 (2000) 193; Deckel et al., Neurology 51 (1998) 1576; Deckel et al., J. Nucl. Med. 41 (2000) 773] suggested that these findings might be accounted for, in part, by alterations in cerebral nitric oxide (NO) and its byproduct, peroxynitrite. The current experiment tested this hypothesis by altering NO levels via manipulations of dietary L-arginine (ARG), the dietary precursor of NO, in mice transgenic for HD. Seventy-one mice were assigned at 12 weeks of age to one of three isocaloric diets that varied in their content of ARG. These diets included: (a) 0% ARG, (b) 1.2% ARG (i.e. typical mouse chow), or (c) 5% ARG. The 5% ARG diets in HD mice accelerated the time of onset of body weight loss (P<0.05) and motor impairments (P<0.05), and increased resting CBF in HD relative to control (P<0.05). Conversely, the 0% ARG diet demonstrated no loss of body weight and had no changes in CBF relative to controls. However, the 0% ARG HD group continued to show significant deficits on motor testing (P<0. 05). The 1.2% ARG HD group showed reduced body weight loss, better motor functioning, and fewer changes in CBF compared to the 5% ARG HD group. Immunocytochemistry analysis found greater deposition of nitrotyrosine in the cortex, and vasculature, of HD+ mice, 5% and 1. 2%>0% arginine diets. When collapsed across all conditions, CBF inversely correlated (P<0.05) both with the body weight and motor changes suggesting that changes in CBF are associated with behavioral decline in HD mice. Collectively, these findings indicate that dietary consumption of the NO precursor ARG has a measurable, but complex, effect on symptom progression in HD transgenic mice, and implicates NO in the pathophysiology of HD.

Spinophilin regulates the formation and function of dendritic spines.

Spinophilin, a protein that interacts with actin and protein phosphatase-1, is highly enriched in dendritic spines. Here, through the use of spinophilin knockout mice, we provide evidence that spinophilin modulates both glutamatergic synaptic transmission and dendritic morphology. The ability of protein phosphatase-1 to regulate the activity of alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) and N-methyl-D-aspartate (NMDA) receptors was reduced in spinophilin knockout mice. Consistent with altered glutamatergic transmission, spinophilin-deficient mice showed reduced long-term depression and exhibited resistance to kainate-induced seizures and neuronal apoptosis. In addition, deletion of the spinophilin gene caused a marked increase in spine density during development in vivo as well as altered filopodial formation in cultured neurons. In conclusion, spinophilin appears to be required for the regulation of the properties of dendritic spines.

Effect of temperature on viscosity properties of some alpha-amino acids in aqueous urea solutions.

Viscosities for solutions of glycine, DL-alpha-alanine, DL-alpha-amino-n-butyric acid, DL-valine, DL-leucine and L-serine in 5 mol kg(-1) aqueous urea have been determined at 278.15, 288.15, 298.15 and 308.15 K. The viscosity B-coefficients for the amino acids in the aqueous urea solution have been calculated at different temperatures. The effect of temperature on the B-coefficients is discussed on the basis of the Feakins equation. The contribution of solute to the activation parameters (delta mu0*2, deltaH0*2, deltaS0*2) for viscous flow of the solution have been calculated, together with the Gibbs energy, enthalpy and entropy of transfer for the amino acids from the ground-state solvent to the hypothetical viscous transition state solvent. The contributions of the charged end group (NH3+, COO-) and CH2 groups of the amino acids to B-coefficient and delta mu0*2 have been also estimated using the linear correlations between B-coefficient or delta mu0*2 and the number of carbon atoms in the alkyl chains of the amino acids. All the activation parameters are discussed in terms of the solute-solvent interactions in the ground and transition states.
