Studies of MAL gene in human esophageal cancer by RNA in situ hybridization.

OBJECTIVE: To observe the expression of MAL gene mRNA in esophageal cancer tissue and adjacent epithelial mucosa. METHODS: Non-radioactive RNA in situ hybridization. RESULTS: MAL gene was expressed in all epithelia of 33 matched samples but only in 8 of esophageal cancer tissues. Differential expression between esophageal cancer tissues and adjacent epithelia was not related to clinical parameters. CONCLUSION: Down-regulation of MAL may be an important event in the development of esophageal cancer.

Clinical observation on treatment of non-parvicellular carcinoma of the lung with jin fu kang oral liquid.

Jin Fu Kang Oral Liquid ([symbol: see text]), made of traditional Chinese drugs for supplementing qi and nourishing yin, was developed according to the common symptoms in lung carcinoma with deficiency of both qi and yin. Of the 96 cases in the Jin Fu Kang group, 1 case got complete remission (CR) after treatment, 8 cases partial remission (PR), 52 cases no change (NC), PR + NC covering 63.5%. Of the 52 cases in the group of Jin Fu Kang plus chemotherapy, 11 cases got PR after treatment, 26 cases NC, PR + NC covering 71.2%. Of the 25 cases in the chemotherapy group, 4 cases got PR after treatment, 11 cases NC, PR + NC covering 60.0%. The results show that the therapeutic effectiveness in the Jin Fu Kang group and the group of Jin Fu Kang plus chemotherapy was better than that in the chemotherapy group. The one-year survival rate and the two-year survival rate after treatment in the Jin Fu Kang group were 67.3% and 67.3% respectively; 66.7% and 66.7% in the group of Jin Fu Kang plus chemotherapy; and 40.3% and 0.0% in the chemotherapy group. The improvement of clinical symptoms, increase of body weight and improvement of health situation (KPS marks) after treatment in both the Jin Fu Kang group and the group of Jin Fu Kang plus chemotherapy were better than that in the chemotherapy group. Some indicators of immunology and hemogram after treatment were greatly improved in the Jin Fu Kang group, worse in the chemotherapy group, but no obvious improvement in the group of Jin Fu Kang plus chemotherapy.

[Protective effect of L-arginine on liver during ischemia-reperfusion injury].

OBJECTIVE: To explore the protective effect of L-arginine (L-Arg) on liver during hepatic ischemia-reperfusion injury (HIRI) and its mechanisms. METHODS: Changes of several parameters, including nitric oxide (NO), malondiadehyde (MDA), thromboxane B(2) ( TXB(2) ), 6-keto-prostaglandin F(1-alpha) (6-keto-PGF(1-alpha) ), TXB(2) / 6-keto-PGF(1-alpha) ( T / K ) and alanine aminotransferase (ALT) as well as liver morphological changes, and the effect of L-Arg on them were observed during HIRI in 20 rabbits and 18 patients who were scheduled for elective hepatic surgery. RESULTS: The levels of NO and 6-keto-PGF(1-alpha) decreased remarkably. MDA concentration, TXB(2) content and T / K ratio as well as ALT activity increased significantly. The morphological changes of hepatocytes were obvious during HIRI. After treatment with L-Arg, the variations of all the parameters were markedly alleviated. CONCLUSION: L-Arg possess preventive effects on HIRI by raising NO and reducing MDA as well as correcting TXA(2) / PGI(2) imbalance after hepatic ischemia-reperfusion.

[A quantitative study of poincare dispersed-dot plot for heart rate variability].

Poincare dispersed-dot plot is an important nonlinear dynamics method that can effectively describe the attractor shape. Yet studies in the past generally processed the plot in a graphic mode without further discussion on the quantitative indices. That was not suited for clinical application and studies. So, based on the shape of Poincare dispersed-dot plot, we propose a group of quantitative parameters of Poincare dispersed-dot plot: plot square(SQ), long axis (LA), short axis(SA), and angle related to long-axis and short-axis(ALS). According to the characters of the heart rate Poincare dispersed-dot plot, we have posed the 'tortoise crawling and counting' algorithm to get these four quantitative parameters. We have tested these four quantitative parameters with clinical and animal experiment data. The results demonstrate that these four parameters are of certain specificity to heart diseases, indicating that these parameters might be useful for clinical diagnosis.

[Expression of recombinant human soluble Flt3 ligand and its in vitro effects on malignant hematopoietic cells].

OBJECTIVE: To express human soluble Flt3 ligand in a yeast expression system. Pichia pastoris, and investigate the effects of recombinant human soluble Flt3 ligand (rhFL) on the proliferation of malignant hematopoietic cells(MHC) and the effect of dexamethasone (DXM) on the expression of Flt3 receptor (Flt3R) and the rhFL induced proliferation of those cells. METHODS: An artificial gene for rhFL was synthesized by using favored genetic codons of the yeast Pichia pastoris. Flt3R was determined by flow cytometry. Proliferation of MHCs was measured by microculture tetrazolium (MTT) assay. RESULTS: 1. An expression vector, pPICZ alpha A-FL, was constructed and electroporated into Pichia pastoris. A yield of over 30 mg/L of yeast culture supernatant was obtained. The rhFL could stimulate the proliferation of Raji and HL-60 cells at concentrations of 10-100 ng/ml, but did not for most of other MHCs. 2. Flt3R was expressed in 5 of 18 MHC lines and the expression in leukemic blast cells was widespread and extremely heterogeneous. 3. The presence of the receptor on the surface of MHC did not necessarily imply a significant ligand-induced response, at least in terms of proliferation. By contrast, some Flt3R-negative MHC responded to rhFL. 4. DXM down-regulated the expression of Flt3R and inhibited the proliferation induced by rhFL in some MHC lines and fresh leukemia cell samples. CONCLUSION: A yield of 30 mg/L of rhFL was obtained. rhFL stimulated the proliferation of Raji, HL-60 cells and some fresh leukemia cells, which could be inhibited by DXM through down-regulation of Flt3R. A combination of FL and DXM might possibly serve to control hematopoietic defects in malignant hematopoietic diseases.

[Experimental study on endothelialization of autologous pericardial heart valves in vitro].

OBJECTIVE: To investigate a method of autologous pericardial heart valves (APHV) treatment beneficial to endothelialization. METHODS: Bovine aortic endothelium cells (BAEC) were cultured and seeded in autologous pericardial pieces treated with different methods in vitro. The influence of different treatment on cellular growing, amount of 3H-TdR incorporation and PGI2 releasing, contents of intracellular cAMP was studied by means of transmission electron microscopy, 3H-TdR incorporation experimentation and radioimmunity method. RESULTS: The BAEC seeded in bovine pericardial pieces treated with three different concentrations of GA 10 minutes could alive. In the group with method of treatment with 0.2% GA, the growing of BAEC was best, the cellular density and the amount of 3H-TdR incorporation at different time spots were higher than that of the other groups and it did not enhance the abnormal synthesis of cAMP and PGI2 in BAEC. CONCLUSION: The pericardium treated with 0.2% GA 10 minutes has little toxicity to endothelium cell, which indicates that this method is beneficial to endothelialization of APHV postoperatively.

[Effect of hypoxic hypercapnia on expression of endothelin-1 mRNA of pulmonary arterioles in rats].

OBJECTIVE: To investigate the effects of chronic hypoxic hypercapnia and ligustrazine on expression of endothelin-1 (ET-1) mRNA and the ultrastructure of pulmonary arterioles in rats. METHODS: Thirty rats were randomly divided into three groups: control group (A), hypoxic hypercapnic group (B), hypoxic hypercapnia + ligustrazine (lig.) group (C). ET-1 mRNA was observed in arterioles from rats by the technique of in situ hybridization. The average value of integral light density (LD) of ET-1 mRNA in pulmonary arterioles was detected by an image analysor and the relative content of ET-1 mRNA was calculated. RESULTS: (1) mPAP was significantly higher in rats of B group than that of A group (P < 0.01) and it was much lower in rats of C group than that of B group (P < 0.01). Differences of mCAP were not significant in three groups (P > 0.05); plasma ET-1 concentration was significantly higher in rats of B group than that of A group (P < 0.01), plasma ET-1 concentration was significantly lower in rats of C group than that of B group (P < 0.01); (2) Light microscopy showed that WA/TA (vessel wall area/total area) and SMC (the density of medial smooth muscle cells) were significantly higher in rats of B group than those of A group (P < 0.01). WA/TA and SMC were significantly lower in rats of C group than those of B group (P < 0.01). Electron microscopy showed proliferation of medial smooth muscle cells and collageous fibers of pulmonary arterioles in rats of B group, and ligustrazine could reverse the changes mentioned above; (3) Hybridization in situ showed that LD of ET-1 mRNA in pulmonary arterioles was significantly higher in rats of B group than that of A group (P < 0.01); LD of ET-1 mRNA in pulmonary arterioles was significantly lower in rats of C group than that of B group (P < 0.01). CONCLUSIONS: Increase of expression of ET-1 mRNA in pulmonary arterioles contributes to the development of pulmonary hypertension and structural remodeling of pulmonary arteries in chronic hypoxic hypercapnic rats. Ligustrazine can inhibit pulmonary hypertension by decreasing the expression of ET-1 mRNA in pulmonary arterioles.

[Beta-diversity of Stipa communities in Inner Mongolia Plateau].

The beta-diversity characteristics of four Stipa communities (S. baicalensis, S. grandis, S. krylovii and S. klemenzii) were studied using comparable plots methods. As a measurement of species turnover within communities, Whittacker's index(beta ws) was size-dependent, and reduced gradually with the increasing quadrat size. Under the same scale, beta ws indices of the communities were similar. Cody's index (beta c) was also affected by sampling size. beta c increased initially as quadrat size enlarged, then stabilized over the size of 0.5 m2 for S. baicalensis and S. grandis communities, and 2 m2 for S. krylovii and S. klemenzii communities. S. baicalensis community had the highest Cody index, while S. klemenzii community had the lowest one. In most cases, the within-community distance interval had a relatively high effect on Cody index; From S. baicalensis to S. klemenzii community, the functional group diversities of forbs, perennial bunch and rhizome grasses decreased greatly, the diversity of intermediate xerads reduced significantly, and the diversity of xerads increased firstly, and then declined for S. klemenzii community. The changes in species composition of four Stipa communities could give a reflection to the degrees of community replacement along ecological gradients, and Cody indices between the communities reduced gradually. The Morisita-Horn similarity coefficient between S. baicalensis and S. grandis community was significantly higher (0.72) than those between S. grandis and S. krylovii communities (0.55), and between S. krylovii and S. klemenzii communities(0.42).

Relationship between metabolic phenotype of N-acetylation and bladder cancer.

OBJECTIVE: To study the relationship between metabolic phenotype of acetylation and bladder cancer. METHODS: Totally 203 healthy volunteers and 67 patients with bladder cancer were investigated with caffeine as a metabolic probe. Urine samples were collected in 2-6 hours after a cup of 140 mg coffee was taken, and the caffeine metabolites, 5-acetylamino-6-formylamino-1-methyluracil (AFMU) and 1-methylxanthine (1X) were analyzed by high performance liquid chromatography (HPLC). The frequency histogram and probit plot were constructed to select the critical value which was used to assess slow and fast acetylation status both in healthy volunteers and patients with bladder cancer. RESULTS: The peak height ratios of AFMU and 1X (AFMU/1X) were from 0.06 to 6.50 for healthy volunteers and 0.10 to 6.31 for patients with bladder cancer, both with the critical value of 1.10. Of 203 healthy volunteers involved in this study, 26.3% were slow acetylacors, as compared to 46.3% with slow acetylacors in patients with bladder cancer. The odds ratio is 2.376, and the gene frequency for healthy volunteers and patients with urinary bladder cancer were 0.5218 and 0.6804, respectively. CONCLUSIONS: N-acetylation status in the Chinese population is polymorphic and completely concordant with that determined with other metabolic probes. Slow acetylators are significantly associated with bladder cancer.

[Management of the neck for patients with thyroid papillary carcinoma].

OBJECTIVE: To search an optimal management of the neck for patients with thyroid papillary carcinoma. METHODS: Clinical data of 424 cases with thyroid papillary carcinoma treated in our hospital from Jan, 1965 to Jan, 1987 were analyzed retrospectively. Patients with positive cervical lymph nodes (N+) were treated with radical excision of the primary tumor and neck dissection. For patients without cervical lymph node involvement (N0), neck dissection was spared. On follow-up, whenever cervical lymphatic node metastasis occurred, neck dissection was performed. All patients were followed up for more than 10 years. RESULTS: The 5- and 10-year survival rate of the 258 N+ patients was 84.3% and 80.4% respectively, and that of the 166 N0 patients was 94.1% and 91.3% respectively. Twenty-three N0 cases later developed cervical lymph node metastasis. Their 5- and 10-year survival rate was 91.4% and 82.2% respectively after neck dissection. CONCLUSION: Since only 13.9% of N0 thyroid papillary carcinoma patients who did not receive neck dissection develop cervical lymph node metastases later, and their survival rate was just as good where neck dissection is postponed until metastases occur, prophylactic neck dissection seems unnecessary.

[Relationship between structural and functional changes of islets, and abnormal metabolism of glucose and lipids in OLETF rats with impaired glucose tolerance].

OBJECTIVE: To investigate the level of blood glucose, cholesterol (Cho), triglycerides (TG), insulin secretion and the efficacy of acarbose in preventing impaired glucose tolerance (IGT) in Otsuka Long-Events Tokushima Fatty (OLETF) rats. METHODS: 10 OLETF rats with IGT, 8 IGT OLETF rats with acarbose treatment, and 36 LETO rats with normal glucose level were observed for their blood glucose, Cho, TG, insulin secretion and histological changes in the pancreas and skeletal muscles. RESULTS: OLETF rats with IGT were mild obese, with fat infiltration in the islets and skeletal muscles. Their blood Cho and TG were higher than those of LETO rats (2.5 mmol/L +/- 0.3 mmol/L vs 2.1 mmol/L +/- 0.3 mmol/L, P < 0.01; 0.7 mmol/L +/- 0.2 mmol/L vs 0.4 +/- 0.2 mmol/L, P < 0.01), but their TG was lower than that of diabetic rats (1.8 +/- 1.0). The Cho, TG and post-challenge glucose levels were normal in the IGT rats with 4-week acarbose treatment, with their insulin secretion and glucose tolerance improved and fat infiltration decreased. CONCLUSION: Acarbose can prevent obesity and fat infiltration in islets and muscles, improve glucose and lipid metabolism, and insulin secretion in OLETF rats with IGT.

[Primary study on immunologic effect of live attenuated hepatitis A vaccine (H2 strain) after booster dose].

OBJECTIVE: To study the immunologic effect of live attenuated hepatitis A vaccine (H(2) Strain, 10(7.0)TCID(50)) after booster and to compare with the results of 1 dose live attenuated hepatitis A vaccine (H(2) Strain, 10(7.0)TCID(50)). METHODS: 42 susceptibles with negative anti - HAV were selected in Zhengding, Hebei province. Each subject received 3 doses live attenuated hepatitis A vaccine at 0, 2, 6 months and was bled at 1, 2, 6, 7, 9, 12 months after vaccination. RESULTS: The seroconversion rate at 1 month after the first dose was 81.4% and reached 100% after the second dose. GMT arrived the peak 2,739 mIU/ml at one month after the third dose, before stared declining. The seroconversion rate kept 100% at 12 months after the first dose, but GMT decreased to 979 mIU/ml. CONCLUSION: The first dose worked as the base of boostering. A booster dose of live attenuated hepatitis A vaccine could induce secondary immune response well. The immunologic effect after booster dose could match the effect with inactivated vaccine and was better than the results of 1 dose live attenuated hepatitis A vaccine. The program seemed to be useful to the protective effect and the immuno - persistence.

[A case-control study on the risk factors of epidemic hemorrhagic fever].

OBJECTIVE: To study the risk factors of epidemic hemorrhagic fever (EHF). METHODS: A 1:1 matched case-control study was conducted to analyse EHF risk factors. RESULTS: One hundred and eighty-five matched pairs were investigated. Five factors were associated with risks of the disease, including rodent activity in house, farm working, travel, annual average income of an individual and ground material of the house, when chi(2) test and logistic regression model were processed respectively. The coefficients of rodent activity in house, farm working, travel, annual average income of an individual and ground material of the house in multivariable Logistic regression model were 1.377, 1.687, 1.630, -1.216 and -0.888, respectively, when P value < 0.05 was observed. CONCLUSIONS: Factors as high average income of individuals, brick or cement used as material of the ground in the house, seemed to be protective factors of EHF, which were important to the control and prevention of EHF.

[Safety and immunogenicity of inactivated bivalent EHF vaccine in humans].

OBJECTIVE: Safety and immunogenicity of inactivated bivalent EHF vaccine in humans were evaluated in the epidemic area of Zhejiang province, China. METHODS: Susceptible persons with negative anti-EHF were selected in Jiande county, Zhejiang province to receive 3 doses of inactivated bivalent EHF vaccine at 0, 7, 28 days. A booster injection was given one year after the primary immunization. Antibody responses were measured in human volunteers by IFA and MCPENT. Local and general reactions were recorded within 72 hours after each vaccination by physicians. RESULTS: Two weeks after the primary vaccination, 99.04% of the subjects developed significant hantavirus antibody titre measured by IFA which had a 37.34% drop one year after the primary vaccination. Seroconversion rate increased to 100% two weeks after the booster dose. Neutralising antibody titres paralleled this trend with 100% of vaccine recipients producing neutralising antibody two weeks after the primary doses. However, it dropped to 80% one year after the primary vaccination. One hundred percent of the vaccine recipients started to respond two weeks after boosting. The geometric mean titre (GMT) of neutralising antibody against 76 - 118 and UR were 18.27 +/- 2.21 and 12.47 +/- 2.16 respectively after the primary injections, but it increased to 37.09 +/- 2.24 and 32.61 +/- 2.05 respectively after the secondary immunization. General and local reaction rates were 0.46% and 1.98%, with no severe side effects observed in the vaccinees. CONCLUSION: The vaccine was well tolerated and could induce good humoral immune response.

[Protective efficacy of H2 strain live attenuated hepatitis A vaccines in an outbreak of hepatitis A].

OBJECTIVE: To investigate the protective efficacy of H(2) strain live attenuated hepatitis A vaccine (H(2)-strain vaccine) locally produced in an outbreak of hepatitis A (HA). METHODS: A nonrandomized controlled design was used in a trial for HA vaccination. Five thousand five hundred and fifty-one children at preschool classes and at grades 1 - 3 of primary schools were selected in 14 villages. Epidemiological and serological methods were used to evaluate its protective efficacy of the vaccines and serum immunoglobulin M against hepatitis A virus (IgM anti-HAV) was detected with ELISA for 271 subjects in vaccinated group and 341 in the control one. RESULTS: An outbreak of HA occurred one year after the vaccination with 26 cases in the vaccinated group, including one vaccine-related case with an attack rate of 0.28% (1/356), and 25 cases in the control group with an attack rate of 5.92% (25/422). The protective efficacy of vaccines was 95.27% (95% confidence interval of 85.83% to 104.72%). Positivity of IgM anti-HAV, i.e., newly-infected cases, was 4.43% (12/271) in the vaccinated group and 13.38% (42/314) in the control group, respectively. Clinically apparent cases accounted for 8.33% (1/12) and 50.00% (21/42) in newly-infected cases in vaccinated and control groups, respectively. CONCLUSION: Vaccination with H(2) strain vaccines locally produced was effective in reducing occurrence of HA during its outbreak, but not so effective in preventing its infection. A booster dose of HA vaccine should be vaccinated for children in adequate time.

[Determination of volatile halohydrocarbons in drinking water by gas chromatography with direct aqueous injection].

The volatile CHCl3, CCl4, CHBrCl2, CHBr2Cl, CHBr3 in drinking water have been determined by wide bore capillary gas chromatography/electron capture detector(GC/ECD) with direct aqueous injection method. The method is simple, fast and highly sensitive. It has better accuracy and precision. When the average mass concentration of five halohydrocarbons ranged from 9.03 to 20.19 micrograms/L, the relative deviations ranged from 5.7% to 9.1%, RSD ranged from 2.9% to 7.5%. The standard deviations ranged from 0.3 to 0.9. Correlation coefficients were 0.998 and 0.999. The detection limits ranged from 0.1 to 0.7 microgram/L and were lower than those in other work when the temperature of ECD was 350 degrees C.

[Screening and fermentation conditions of a bacterial strain over-producing xylanase].

A xylanase producer WXULI-11 was isolated and identified as Pseudomonas sp. By combination treatment of WXULI-11 with UV and NTG, a mutant WLUN024 was obtained and its enzyme activity reached 354 IU/mL in 36 h by shaking flask. The mechanism of its xylanase production was primarily studied. The results showed that this enzyme was induced by xylan, xylose and some other xylosic material, while xylose had "two-way effect" on xylanase synthesis. Effects of nitrogen and other factors on enzyme production were also investigated.
