[Effects of aspirin and nifedipine alone or in combination on mesenteric microcirculation of rats].

AIM: To study the effects of the combination of aspirin (Asp) and nifedipine (Nif) on mesenteric microcirculation of rats. METHODS: Acute microcirculation disturbance (AMD) was produced by high molecular weight dextran (M(r) 480,000, 360 mg.kg-1 i.v.). Arteriole and venule blood flow velocity and diameter (ABFV, VBFV, AD, VD) and blood flow state (BFS) were observed by intravital microcirculation method. RESULTS: Asp 2.5, 5 mg.kg-1, Nif 0.05, 0.1 mg.kg-1, Asp + Nif (1 + 0.025), (2.5 + 0.05) mg.kg-1, i.v. had the significant increase of 11.1%, 31.3%, 18.5%, 19.3%, 30.5%, 39.8% of ABFV and 12.5%, 25.7%, 12.6%, 15.2%, 29.6%, 36.1% of VBFV respectively, the marked improvement of BFS, and the distinctive increase of 4.3%, 17.9%, 35.9%, 39.7%, 15.2%, 42.8% of AD and 2.2%, 4.2%, 26.2%, 27.4%, 3.4%, 28.9% of VD separately, and got a raise in the number of capillaries. Asp + Nif (1 + 0.025), 2.5 + 0.05) mg.kg-1 i.v. could reverse AMD. CONCLUSION: Asp was superior to Nif in the increase of BFV, but Nif was superior to Asp in expansion of blood vessel. Asp in combination with Nif produced marked synergistic action and protection againist AMD.

Localization and biodistribution of conjugate ATG-Dex-DNR in nude mice as models for human leukemia.

131I-labelled anti-thymoglobuline (ATG), 131I-labelled immunoconjugate ATG-Dex-DNR and 131I-labelled Ts-MoAb as control antibody, respectively, were injected by intraperitoneal (i.p.) administration into nude mice used as models for human T-cell leukemia. SPECT imaging was performed from day 1 to day 8 following i.p. injection. The results showed that radioimmunoimaging of human tumor xenografts was clearest day 3 after injection in both of ATG and ATG-Dex-DNR groups, whereas it's not the case in Ts-MoAb group. Nude mice were killed 8th day after injection with antibody or conjugate. The tumor, as well as different dissected normal organs including heart, liver, lungs, kidney, femur and intestine, were harvested, weighed precisely, and radioiodine-counted. T/NT ratios in experimental group was greater than 1.0 (ranged from 1.246-7.865), and in control group they were less than 1.0 (ranged from 0.263-0.757, except for tumor/femur ratio). Our results indicated that ATG and ATG-Dex-DNR had specific affinity to cell line of T-cell leukemia CEM.

[Pharmacokinetics of contragestazol (DL-111-IT), a new non-steroid antifertility agent in monkeys].

The pharmacokinetics of contragestazol, an early pregnancy temperating agent [3-(2-ethylphenyl)-5-(3-methoxyphenyl)-1H-1,2,4-triazole, DL-111-IT] was studied in Rhesus monkey. The blood concentration of DL-111-IT was determined by coupled column system HPLC method. Using an aqueous vehicle (20% cremophor EL in saline) DL-111-IT was injected intravenously to monkeys at doses of 25, 12.5 and 6.3 mg.kg-1. Blood drug concentration were measured. Using a programmable calculator the calculated pharmacokinetic parameters were as follows: alpha 1.83 h-1, 4.71 h-1 and 3.61 h-1, beta 0.15 h-1, 0.08 h-1 and 0.09 h-1; T1/2 beta 6.63 h, 10.2 h and 10.1 h; AUC 9.54 micrograms.h-1.ml-1, 3.94 micrograms.h-1.ml-1 and 3.75 micrograms.h-1.ml-1. An oil solution of DL-111-IT was injected intramuscularly in monkeys at doses of 50, 25 and 12.5 mg.kg-1. Its blood concentrations were determined at 0.08, 0.25, 0.5, 0.75, 1, 1.5, 2, 4, 8, 12 and 24 h after administration. From the time vs concentration curve, the pharmacokinetic parameters obtained were as follows: Ka 0.98 h-1, 1.03 h-1 and 1.45 h-1; Ke 0.42 h-1, 0.37 h-1 and 0.60 h-1; T1/2Ke 1.66 h, 1.90 h and 1.16 h; T (peak) 1.52 h, 1.57 h and 1.09 h; AUC 4.86 micrograms.h-1.ml-1, 5.61 micrograms.h-1.ml-1 and 1.74 micrograms.h-1.ml-1.

[A preliminary study of HCV infections with hemophilia and their family members].

28 cases of hemophilia were examined for HCV infection status by using the Kehua anti-HCV ELISA kit of second generation. It was found that the infection rate was 78.5% and the infection rate was even higher with patients who had received transfusions or preparations of coagulatory factors. 10 families of 15 patients were also investigated. It was found that of 15 hemophilia patients, 12 showed positive anti-HCV, while none of their 53 family members exhibited any positive anti-HCV. In 8 children of 9 couples no positive anti-HCV was found. Our results revealed that the hemophilia patient may get infected with HCV by receiving multiple transfusions or preparation of coagulatory factors. The risk of getting infected with HCV via daily-life contact including sexual contact is extremely low.

Pattern recognition analysis to the variation of nasal-pharynx cancer patients' trace element levels in samples of hair, whole blood, and tissue.

Pattern recognition has been used in this paper to analyze trace element levels in patients diagnosed with nasal-pharynx cancer (NPC) and in healthy control subjects. Trace elements such as Zn, Cu, Fe, Mn, and Mg have been tested in samples of hair and whole blood. Through Mahalanobis Distance Decision analysis, we have achieved good classification effects in whole blood samples: Efficiency for distinguishing patients in 96% and that of healthy controls is 90%. Classification hair samples is inferior to whole blood: Decision accuracy for patients is 58% and healthy controls is 90%. These results are also shown in a nonlinear mapping figure. At the same time, we have also determined 5 trace element levels in 16 other cancer patients' nonneoplastic and cancerous tissue, with no significant difference between them: Decision accuracy of cancerous tissue is 63%, and in nonneoplastic tissue is 50%, hence, we cannot identify them. It can be inferred that there is no idiosyncratic change of trace elements in cancer patients' neoplastic tissue, the change of a cancerous person may occur in the whole body.

Brain abscess due to Pasteurella multocida.

The patient was a 26-year-old man who complained of headache and vomiting. On examination, there was nothing abnormal, but the edge of the right optic papilla was not clear. His temperature was 38.5 degrees C, pulse 96/min, blood pressure 120/80 mmHg. A space-occupying lesion in his fronto-dextra cupular part was found by CT scanning. He had a 12-year history of chronic purulent otitis. The diagnosis was a brain abscess in the fronto-dextra cupular part. The brain abscess was extracted and Pasteurella multocida was isolated from the dark brown pus draining from the abscess. The patient recovered through proper antibiotic therapy based on a sensitivity test. Reports of infections caused by this organism in foreign countries very widely from local infections due to bites and scratches by cats, dogs etc. to general infections such as infections of the respiratory tract, sepsis and meningitis. However, Pasteurella multocida brain abscesses are rare. Pasteurella multocida is a Gram-negative short rod which is best known as part of the mouth flora and as a pathogen causing septicemia in many domestic animals, such as cats, dogs etc.. Infection in man results mainly from animal bites or scratches. It has been reported that Pasteurella multodida can cause human septicemia, meningitis, respiratory tract infection, conjunctivitis and other infections. We isolated a strain of Pasteurella multocida from the pus of a brain abscess following chronic purulent otitis on August 6, 1990.

[The treatment of borderline ovarian tumors and its prognosis].

Twenty patients with ovarian borderline epithelial tumors were analysed. The tumor in 9 patients were serous and in 11 patients mucinous in nature type. 15 cases were classified as of stage I, 3 cases of stage II and 2 cases of stage III, according to the FIGO staging. All pathologic slides were re-reviewed to confirm the diagnosis and no patients were lost to follow-up. Total hysterectomy and unilateral or bilateral salpingo-oophorectomy were done in 12 patients, among whom, 2 died of tumor recurrences. Unilateral adnexectomy was done in 8 patients, and none died of tumor recurrence. Fourteen patients received post operative chemotherapy 1-8 courses with PAC regimen, 12 patients were alive up to the present. Six patients did not receive chemotherapy and among these 3 with stage Ia were alive and 1 with stage Ib died of tumor recurrences. The survival rate of stage I was 92.3%. Our results indicated that the prognosis was closely related to clinical staging. We suggested that patients of stage I who wish to preserve childbearing function may be treated with unilateral salpingo-oophorectomy, and chemotherapy should be given to the patients with high risk factors. Because of the indolent nature of these tumors, long-term follow-up at least 10 years is necessary.

[Prognostic significance of nuclear DNA content of colonic carcinoma].

In this study nuclear DNA content of the cancer tissue was analysed retrospectively by microspectrophotometry in two groups of colonic cancer patients: Group I, 19 cases who died within two years after radical surgery and Group II, 24 cases with a survival of more than 5 years. The other clinicopathological findings including location of tumor, grade of histological differentiation, depth of invasion, lymph vessel and venous invasion, lymph node metastasis, histological stage, and preoperative CEA level were similar between the groups. The DNA histograms were classified into two types as following: Type I: in which the ratio of over 3C cells was less than 15% type II: in which the ratio of over 3C cells was 15% or more. It was found that the percentage of type I DNA histogram was significantly higher in the long-term-survivors, while type II histogram was predominant in the short-term-survivors (P < 0.05). These results suggest that the nuclear DNA content of the cancer cells may bone of prognostic value in colonic carcinoma.

[Sister chromatid exchanges in lymphocytes of patients with gynecological malignancies].

The frequency of SCE in the peripheral blood lymphocytes of 30 patients with gynecological malignancies was investigated. The frequency of SCE was found to be 5.37 +/- 0.44, 5.30 +/- 0.35 and 3.76 +/- 0.38 respectively in the patients before and after chemotherapy and in the control cases. The SCE values deviated significantly from those of control, and were not related to the staging of the tumor. The study suggested the possibility of utilizing SCE as a cytogenetic indicator in some malignant tumors.

[Cisplatin nephrotoxicity: clinical observation and animal experiment].

Animal experiment and clinical observation were carried out to study the nephrotoxicity of cisplatin in order to give protective measures. In animal experiment it had been demonstrated that the nephrotoxicity induced by cisplatin was mainly related to the intraperitoneal concentration of cisplatin. The nephrotoxicity was permanent in rats treated with cumulative cisplatin. The main pathologic lesions were characterized by interstitial hyperemia, degeneration of the tubular cells and formation of tubular casts. In a Clinical study to compare two preventive methods (sodium thiosulfate and hydration) from cisplatin induced nephrotoxicity, the BUN and serum Cr were measured in 28 patients. The results revealed that there was no significant difference between the two groups (P greater than 0.05). The two protective measures had similar effect in reducing the nephrotoxicity of cisplatin. Five of 6 patients treated with cisplatin developed hypo-magnesemia but only one had symptoms.

[Pharmacokinetics of norethindrone-3-oxime and norethindrone in rhesus monkeys].

After intravenous administration or nasal feeding of (6, 7-3H)-labelled norethindrone-3-oxime (NETO) or norethindrone (NET) to rhesus monkeys, the serum concentrations were determined by measuring the radioactivity after separation with HPLC. The serum concentrations of total extractable radioactivity were also measured without HPLC separation. In cases of nasal feeding, NETO and NET were quickly absorbed, and almost all of them were eliminated within 24 hours. NETO, when given via both routes, was partly metabolized to NET and partly remained in original form. The blood concentration--time curves for NETO and NET were adequately fitted to two compartment models. No significant difference in pharmacokinetic parameters between the two drugs was observed. The absolute bioavailability for NETO and NET were found to be 64.46 +/- 34.60% and 35.02 +/- 26.49% respectively.
