[Study on antitumor effect and mechanism of aloe polysaccharides].

OBJECTIVE: To study the antitumor activity and mechanism of aloe polysaccharides (AP). METHODS: AP was administered i.p. or i.v. to Sarcoma 180(S180) bearing mice or Hepatoma22(H22) bearing mice solely or combining with CTX, FU and ADM respectively. 10 days later, for S180 mice, the blood was analyzed, the tumor was peeled off and weighed, and the spleen index, thymus index was calculated. For H22 bearing mice, the survival rate was observed or the IL-2, TNF content in serum was tested. RESULTS: 25 mg/kg.d or 50 mg/kg.d AP group could evidently reduce the tumor weight of S180 bearing mice and prolong the survival time of H22 bearing mice. AP also could improve the antitumor effects of CTX, ADM, FU, and lessen the chemotherapy side-effects. Furthermore, AP could improve the level of IL-2, TNF in the serum of mice bearing S180 or H22. CONCLUSION: AP has the effects of antitumor, enhancing the antitumor activity of chemotherapy drugs and lessening their side-effects. This effect was possibly derived from inducing IL-2 and TNF producing in body and improving the immunity activity.

[The effects of adenosine, interleukin-1 and theophylline on the expression of A2 adenosine receptor mRNAs in peripheral blood mononuclear cells from asthmatic patients].

OBJECTIVE: To explore the roles of A2a and A2b adenosine receptors(A2aAR and A2bAR) in the pathogenesis of bronchial asthma. METHODS: Eleven asthmatics and 11 healthy subjects were included in this investigation. Peripheral blood mononuclear cells(PBMCs) were obtained after density centrifugation on Ficoll-Hypaque. The expression of A2aAR and A2bAR mRNAs in PBMCs was examined by use of the reverse transcription-polymerase chain reaction (RT-PCR) and image pattern analysis semiquantitation method. RESULTS: The expression of A2aAR mRNA in PBMCs from asthmatics was not enhanced, compared with that of healthy subjects(P > 0.05). Adenosine or IL-1 or theophylline was not observed to have effect on A2aAR mRNA (P > 0.05). A2bAR gene expression in PBMCs was more markedly in patients with asthma than in normal individuals (P < 0.05). Adenosine and IL-1 significantly increased A2bAR mRNAs in asthmatics. Theophylline inhibited A2bAR gene expressions. The expression of A2bAR mRNA in PBMCs was positively correlated to serum total immunoglobulin E (TIgE) level in asthmatics (r = 0.72, P < 0.01). The expressions of A2bAR mRNA elevated by adenosine or IL-1 were correlated positively with serum TIgE(r = 0.78 or r = 0.61; P < 0.05) and negatively with the forced expiratory volume in first second to predicted value ratio (FEV1%) (r = -0.62 or r = -0.81; P < 0.05) in asthmatics. CONCLUSION: These results indicated that the expression of A2bAR mRNAs in PBMCs was more remarkable in asthmatics than in healthy subjects; that adenosine or IL-1 potentiated the mRNA expression of A2bAR in PBMCs in asthmatic patients, which was correlated with allergy state and the degree of airway obstruction; and that theophylline might antagonize adenosine by inhibiting A2bAR mRNA.

[The risk of sustained amenorrhea in patients with systemic lupus erythematosus receiving intermittent pulse cyclophosphamide therapy].

OBJECTIVE: To determine the risk of sustained amenorrhea in premenopausal women with systemic lupus erythematosus (SLE) receiving intermittent pulse cyclophosphamide (CTX) therapy. METHODS: Prospectively comparing the amenorrhea rate of 51 cases receiving intermittent pulse cyclophosphamide therapy versus that of 22 cases receiving intermittent pulse methylprednisolone (MP) therapy. RESULTS: The amenorrhea rate was higher in the CTX group (19.6%) than in the MP group (P = 0.025). In the CTX group, the amenorrhea rate of patients aged over 30 was higher than that of patients aged 30 or below 30 (P = 0.0018). CONCLUSION: Pulse CTX therapy in fertile women with SLE is associated with increased rate of sustained amenorrhea, and the older the patient is, the higher risk for sustained amenorrhea the patient runs.

[A kinetic study on the relationship between of IL-5, IL-10 and eosinophil apoptosis in asthmatic airway inflammation].

OBJECTIVE: To observe time-course profile of eosinophil apoptosis, in murine asthma models in airway explicate its implication for asthmatic inflammation remission, and investigate the role of IL-5 and IL-10 in modulating cell apoptosis. METHODS: An animal model of asthma was established by OVA sensitizing-challenging BALB/c mice. Histologic study of lung tissue was made with the use of electronmicroscope. At seven points (0 h, 8 h, 24 h, 48 h, 96 h, Day 7 and Day 14) in the time course after challenge, bronchoalveolar lavage (BAL) was performed obtain BAL cells and recover fluid. Levels of IL-5 and IL-10 were detected by ELISA. Cell apoptosis was assayed by PI stain on a flow cytometer. RESULTS: After challenge, apoptosis of eosinophils, lymphocytes, endothelial cells could be seen under the electronmicroscope. IL-5 in BALF elevated significantly whereas no significant difference was found between the levels of IL-10 in BALF before and after challenge. Eosinophil apoptosis was identified and a two-phase elevation of apoptosis percentages was observed, although no significant difference was found between average apoptosis in asthmatic group and that in control. Data analyses revealed a negative correlation between apoptosis percentages and the levels of IL-5, and a closer correlation between the percentages and IL-5/IL-10 ratio. CONCLUSION: IL-5 and IL-10 could up- and down-regulate asthmatic airway inflammation, respectively. EOS underwent apoptosis in situ and this was modulated by IL-5 and IL-10 interaction.

[Study on bolus cyclosphamide treatment for 64 cases of lupus nephritis].

OBJECTIVE: To report on the dosing, efficacy and side-effects of bolus cyclosphamide treatment for lupus nephritis (LN). METHODS: 64 consecutive cases of LN with 10 or more erythrocytes per high-power field, proteinuria (> 1 g of protein per day) and serum creatinine increased (> 133 mumol/L) were treated by bolus therapy with cyclophosphamide (CTX) given monthly for 6 months and then quarterly for 18 months. RESULTS: 49 patients had renal remission (defined as < 10 erythrocytes per high-power field, absence of cellular casts, excretion of < 1 g of protein per day and normal serum creatinine). The mean of doses was 1.1 g for each time (0.6-1.6 g), the mean of times of bolus CTX needed was 3.6 (1-8 times). The adverse events were amenorrhea (in 41% female patients), herpes zoster (in 13% patients) and hemorrhagic cystitis (in 1 patient). CONCLUSION: The results indicate that monthly bolus CTX therapy is effective and safe for patients with LN. Its adverse effect is relatively not a serious problem.

[Expression of bcl-2 in facial motoneurons and its ultrastructural localization following facial nerve injury].

OBJECTIVE: To explore the expression of bcl-2 in facial motoneurons and its subcellular distribution. METHODS: Wistar rats were used in this study. Facial nerve transection was performed at stylomastoid foramen or internal acoustic meatus. Facial nerve crush was made at stylomastiod foramen. The animal survived for 1, 3, 7, 15, 30 and 60 days respectively. Facial nucleus was treated with bcl-2 monoclonal antibody or bcl-2 DIG-labelling probe and studied with immunohistochemistry and in situ hybridization. The bcl-2 positive motoneuron was investigated with immuno-electron microscope. RESULTS: It was demonstrated that bcl-2 protein level was corresponded with bcl-2 mRNA expression. The level of bcl-2 expression in facial motoneurons was high in normal facial nerve. It increased on the first day and declined on the third day post-transection in facial motoneuron. It reached the lowest level on the 15th days following facial nerve injury (P < 0.05). The expression recovered to normal level in two months (P > 0.05). After facial nerve transected, the reduction of bcl-2 expression was more significant when facial nerve transected close to facial nucleus than that far from facial nucleus (P < 0.05). Comparing to facial nerve transection in stylomastoid foramen, there was more intensive bcl-2 expression following facial nerve crush (P < 0.05). Further study showed that bcl-2 primarily resided in the nuclear envelop, endoplasmic reticulum and mitochondrial membrane. CONCLUSIONS: These data indicated that high level bcl-2 protein may prevent facial motoneuron death following facial nerve injury. It is suggested that overexpression bcl-2 by transgene may prevent facial motoneurons death.

[Research of the chorda tympani nerve in cholesteatoma].

OBJECTIVE: To investigate the ultrastructure of the chorda tympani nerve and analyze the taste and facial nerve functions in patients with cholesteatoma. METHODS: 1. The tympanic segments of chorda tympani nerves were collected for ultrastructural investigations in 19 cholesteatoma cases who underwent canal-wall-down tympanoplasty. 2. All these patients received a spatial (regional) taste test preoperatively and postoperatively, respectively. Multiple loci were tested in a given order (front of the tongue, foliate papillae, circumvallate papillae, and soft palate). The solutions used in this study were 1.0 mol/L sodium chloride (salty), 1.0 mol/L sucrose (sweet), 0.032 mol/L citric acid (sour) and 0.001 mol/L quinine hydrochloride (bitter). The analyses of variance was used. 3. House-Brackmann facial nerve grading system was conducted to evaluate the facial nerve function preoperatively and postoperatively. RESULTS: 1. There were obvious ultrastructural damages in all the chorda tympani nerves, such as swelling (100%), disarrangement (100%), vacuoles formation of myelin (89%), edema of Schwann cells (95%), intracytoplasmic vacuoles in Schwann cell (89%) and proliferation of the collagen tissue (89%). 2. Two patients complained of change of taste after operation. The analyses of variance showed that the taste function of the ipsilateral side of tympanoplasty were not statistically altered for each stimulus at each locus (P > 0.05). 3. No facial palsy occurred postoperatively. CONCLUSION: This research suggested that the chorda tympani nerves underwent ultrastructural changes in patients with cholesteatoma. The dissection of chorda tympani nerve would not affect the taste and facial nerve functions.

Preservation of the hypothalamic structures in the total resection of craniopharyngioma.

OBJECTIVE: To explore an effective method of surgical management of craniopharyngioma. SUBJECTS AND METHODS: Fifty patients with craniopharyngioma had total and subtotal tumor ectomy. There were 29 males and 21 females, ranging in age from 15 to 56 years (mean 34.1 years). MR imaging showed that the tumors were located in the superior sellar region in 24 cases, in superior sella region and extended into the third ventricular floor in 19 cases, into parasella in 3 cases and down to intrasella in 4 cases. Complete cystic tumors were found in 5 cases, while the partial cystic tumor in 24 cases and complete solid tumors in 21 cases. Pterional approach was used in 48 patients and subfrontal approach in 2 patients. Great attention was paid to the preservation of the perforating arteries from the carotid, posterior and anterior communicating and anterior choroidal arteries to the hypothalamic structures. The clinical outcome was evaluated according to the GOS scale. RESULTS: Of the 50 patients surgically treated, 47 patients obtained total ectomy of the tumor and 3 patients with the secondary surgery had subtotal ectomy of the tumor. The pituitary stalk was preserved in 29 (58%) patients, severed in 14 patients and unidentified in 7 patients. Forty-six patients regained a normal life; one patient needed assist in life. Of the 3 deaths, one patient died of diabetes insipidus, one of inhalation asphyxia, and another one of water and sodium disorders. CONCLUSION: Avoidance of the injury to the neural structures in the third ventricular floor and preservation of the perforating arteries to hypothalamus are the key to achieve good surgical results in treating craniopharyngioma.

The management of epithelial ingrowth after laser in situ keratomileusis.

OBJECTIVE: . To investigate the treatment of corneal epithelial ingrowth with flap melt after LASIK. PATIENTS AND METHOD: Thirteen eyes of 13 patients with corneal epithelial ingrowth and flap melt after LASIK were treated based on the thickness and the extent of melting of the flap. Nine eyes were treated by lifting the flap and scraping the epithelium in the interface. Three eyes underwent allogenic keratomileusis. The flap was removed in one eye. RESULTS: Recurrence occurred in 2 eyes treated by lifting the flap. The flaps, however, healed normally after retreatment. The grafts healed normally in the allogenic keratomileusis group, except for one eye in which the flap was lost. The corneal epithelium healed over the stromal bed after the flap was removed. Corneal haze of grade 2 was present at 6 months. The uncorrected visual acuity was between 0.3 and 1.0, and the best-corrected visual acuity lost 1 to 3 lines. CONCLUSIONS: Corneal epithelial ingrowth with flap melt is a severe complication after LASIK. However, with proper treatment, vision can be salvaged in most cases.

[IR studies on hydrogen bonding in hard segment domains of segmented polyester polyurethaneurea].

Hydrogen bonding in hard segment domains of segmented polyester polyurethaneurea (PUU) based on 4,4'-diphenylmethane diisocyanate (MDI), ethylene diamine (ED) and polybutylene adipate (PBA) (M.W.1,975 and M.W.1,228) was studied by means of IR spectroscopy. It was found that the double peak absorption of urea C=O groups occurred in the studied PUU with the increase of hard segment content. Two hydrogen bonding models of urea C=O groups in different hard segment sequence lengths of PUU were proposed. Three dimensional hydrogen bonding in hard segment domains can be formed by further bonding of the above planar hydrogen bondings.

[IR studies on deuterated polyester polyurethaneurea elastomers].

Segmented polyester polyurethaneurea (PUU) based on 4,4'-disphenylmethane diisocyanate (MDI), ethylene diamine (ED) and polybutylene adipate (PBA) elastomers and their deuterated polymers were studied by means of IR spectroscopy. The results indicated that IR bands of hydrogen bonded N-H vibrations involved in PUU were greatly affected by deuteration. The bands which were sensitive to deuteration were those near 3,332, 3,190, 1,600, 1,539, 1,317, 1,257 and 1,230 cm-1, respectively. In addition, it was also found that PUU samples with different hard segment contents and polyester soft segment molecule weights were similarly affected by deuteration in degree.

[Direct determination of rhizoma cimicifuga by FTIR spectroscopy].

Fifteen samples of original plants of Rhizoma Cimicifuga were determined quickly and directly with the method of FTIR and comparing software. The results showed that the spectra of these samples were characteristic relying on the difference of chemical components and their content of related components. The difference of spectra among samples belonging to different families was distinct, and in different species with same genus too. To the same species, between two samples with different producing areas, or with same producing area but different collection date, the spectra were not obvious. This approach provide a quickly and accurately determination method of crude drug without extraction and separation.

[IR studies on segmented polyester polyurethaneurea elastomers].

The studies segmented polyester polyurethaneurea (PUU) elastomers based on 4,4'-diphenylmethane diisocyanate (MDI), ethylene diamine (ED) and polybutylene adipate(PBA) (M.W.1,975 and M.W.1,228) were studied by means of IR spectrascopy. The major IR bands of PUU were first assigned and the views were proposed for the bands at 1,600, 1,317, 1,520 and 1,230 cm-1 assigned by other authors. The reliable theoretical basis for the assignments of IR bands and the studies of hydrogen bonding in these elastomers has been proposed by use of deuteration and varying temperature. The results indicated that substantial hydrogen bonding mainly occurred in the aggregate region of hard segments, a small amount of hydrogen bonding can be formed at the interface between the soft and hard segments. It can be concluded from this work that the studied PUU materials were phase segregation.
