Clinical value of urinary and serum pseudouridine in diagnosis and monitoring of primary liver cancer.

Urinary and serum pseudouridine concentrations were determined by high-performance liquid chromatography in 80 patients with primary liver cancer, 32 with benign space occupying lesions of the liver, 42 with liver cirrhosis and 40 healthy subjects. Their mean urinary and serum pseudouridine levels were 39.2 +/- 11.5 nmol/mumol creatinine and 3.4 +/- 1.3 mumol/L, 24.5 +/- 5.4 nmol/mumol creatinine and 2.5 +/- 0.5 mumol/L, 22.8 +/- 7.8 nmol/mumol creatinine and 2.3 +/- 0.4 mumol/L, 26.4 +/- 4.6 nmol/mumol creatinine and 2.3 +/- 0.4 mumol/L, respectively. Exceeding the mean plus 2SD of pseudouridine of healthy control was considered as positive value for the diagnosis of primary liver cancer. Thus the positivity of urinary and serum pseudouridine in hepatoma was 71.3% and 70.0%, respectively. The positive rate of combined pseudouridine and alpha-fetoprotein assay was 91.3% in patients with hepatoma. Besides, pseudouridine levels could elevate before positive localization and reduce to normal levels after tumor resection. The results showed that the determination of pseudouridine is of clinical significance in the diagnosis and monitoring of primary liver cancer.

An infectious cDNA copy of the genome of a non-cardiovirulent coxsackievirus B3 strain: its complete sequence analysis and comparison to the genomes of cardiovirulent coxsackieviruses.

The genome of the non-cardiovirulent coxsackievirus B3 (CVB3) strain CVB3/0 was cloned and sequenced to aid in the elucidation of the viral genetic basis for the CVB3 cardiovirulent phenotype. Reverse-transcribed sub-genomic complementary DNA (cDNA) fragments were enzymatically amplified using generic oligonucleotide primers and were assembled as a complete infectious genomic copy (pCVB3-0) downstream of the T7 RNA polymerase promoter. Positive-strand viral RNA transcribed from pCVB3-0 using T7 RNA polymerase and transfected into HeLa cells produced infectious virus (CVB3/0c). No differences in phenotype were observed comparing growth of CVB3/0c to the parental CVB3/0 in HeLa single-step growth curves, virus yields, or plaque size. When inoculated into C3H/HeJ mice, CVB3/0c achieved cardiac titers equivalent to the parental CVB3/0 and like the parental virus, demonstrated a non-cardiovirulent phenotype. The nucleotide sequence of the cloned CVB3/0 genome was determined and compared to the genomes of infectious cDNA clones of cardiovirulent CVB3 strains. Two consistent differences among nucleotides in non-translated regions and 8 amino acid differences relative to two well-characterized infectious cDNA copies of genomes from cardiovirulent CVB3 strains were identified.

Coxsackievirus B3 from an infectious cDNA copy of the genome is cardiovirulent in mice.

A coxsackievirus B3 (CVB3) cDNA clone, upon transfection of HeLa cells, produces CVB3 capable of induction of cardiac inflammation in C3H/He mice by day 8 post inoculation (p.i.). Liver and serum are cleared of CVB3 by day 8 p.i., but CVB3 persists in the heart through day 14. The nucleotide sequence and the predicted amino acid sequence of this clone have sites of divergence from 2 other completely sequenced CVB3 genomes although overall identity of the three CVB3 genomes is 99%.

Prospective study on the diagnosis of hepatocellular carcinoma by using alpha-fetoprotein reactive to lentil lectin.

In order to study the changes in AFP reactive to lentil lectin (AFP-R-L) during the development of hepatocellular carcinoma (HCC) and its clinical significance, AFP-R-L was monitored in 64 patients by using 20-400 ng/L AFP and negative imaging localization. AFP-R-L was determined by using affino-crossed-immunoelectrophoresis autoradiography. AFP-R-L above 25% was considered positive for the diagnosis of HCC. Over a follow-up of 3-31 months, 32 patients developed HCC. The positivity of AFP-R-L was 59.4% in the HCC group at the first assay. The accuracy of using positive AFP-R-L values to predict HCC was 95.0%. Increased AFP-R-L levels appeared 3-24 months earlier than did positive imaging location. The results suggest that positive AFP-R-L values can predict the development of HCC.

[Prospective study on the diagnosis of hepatocellular carcinoma by using alpha-fetoprotein reactive to lentil lectin].

Levels of alpha-fetoprotein reactive to lentil lectin (AFP-R-L) were monitored in 27 patients with 20-400 micrograms AFP/L and negative imaging. These cases finally were proven to be affected by hepatocellular carcinoma. AFP-R-L above 25% was used to define the positive result for hepatocellular carcinoma. The positivity of AFP-R-L was 63%. At the same time, AFP-R-L was detected in 71 cases of chronic liver diseases. All but one had negative AFP-R-L values. The results suggested that AFP-R-L could make a dissection 3-28 months earlier than positive imaging. An accuracy of predicting hepatoma with positive AFP-R-L was 94%. So the diagnosis of hepatoma with negative imaging could be early made by measurement of AFP-R-L.

Mass treatment of filariasis using DEC-medicated salt.

Diethylcarbamazine (DEC)-medicated salt was used to control bancroftian filariasis in 20 endemic counties and cities of Fujian Province in China. The population was 5,189,126 and the microfilaria (mf) rates were between 1.56 and 11.81%. The total dose of DEC was 6-10.5 g per person over a period of 2-4 months. Six to 9 months after treatment the mf rate was reduced to 0-0.57% with a mean of 0.07% (621/823,796). In 184 villages sampled one year post treatment the mf rate was 0.22% (371/171,067) and in 29 villages 2-4 years after treatment the mf rate was 0.05% (31/56,459). In a region of endemic Malayan filariasis, with a population of 67,778 and mf rate of 3.82-4.36%, treatment with DEC-medicated salt to a dosage of 3.2 g DEC per person was administered for 2 months. Six months after treatment the mean mf rate was reduced from 4.08 to 0.26% (10/3841). Filariasis in all the villages controlled with DEC-medicated salt reached the basic criteria of elimination (mf rate below 1%).

[The effects of microinjection of TRH or morphine into unilateral nucleus septum on pain threshold and electroacupuncture analgesia in rats].

Experiments were performed on awake rats. The pain threshold (tail flick latency, TFL) was measured with radiant heat tail-flick method. Microinjection of thyrotropin releasing hormone (TRH 2.5 micrograms/1 microliter) into septal area can increase remarkably and last 40 min, but the analgesic effect can not be reversed by the opioid receptor blocker naloxone (NAL) into the septal area; injection 10 micrograms of morphine into unilateral septal nucleus, the pain threshold had no obvious change; the after-effect of electroacupuncture (EA) was remarkably prolonged injecting TRH (1.5 micrograms) into unilateral septal nucleus. The after-effect of EA caused by TRH could abolish by NAL. The results imply that (1) microinjection of TRH into the unilateral septal nucleus could raise TFL, intraseptal opiate receptors may not be involved in analgesic effect elicited by TRH. (2) morphine was injection into unilateral septal nucleus having no effect on TFL, suggesting that endogenous opioid peptides in the septal nuclei might not be the major role in anti-nociceptive reaction in rats. (3) Microinjection TRH into the intraseptal can obviously prolong the after-effect of EA, NAL could abolish it, indicating that endogenous opiate peptides in the septal nuclei may play a role partially in mediating EA analgesia.

[Effect of intracerebroventricular injection of thyrotropin releasing hormone on electro-acupuncture analgesia and the content of cerebral cAMP in rats].

Experiments were performed on 42 female albino rats weighing from 180-250 gm. The pain threshold (PT) was determined by radiant heat induced tail-flick method. The cAMP contents in various regions of brain were measured by radio-immunoassay. Electro-acupuncture (EA) at bilateral "Zusanli" and "Sanyinjian" in rats significantly elevated PT, increased the content of cAMP in the hypophysis, while decreased that of septal nucleus. No effects were found in the basal PT and the cAMP contents of the hypophysis and septal nuclei after intraventricular injection of TRH. After TRH administered intraventricularly, however, EA didn't elevate PT, but still increased hypophysial cAMP with a decrease in the level of cAMP in the septal nucleus. These results suggest that the cAMP may be related to analgesic effect induced by EA in some brain regions, and TRH injected into cerebral ventricles can antagonize the analgesic effect caused by EA, but this effect seemed to have nn relation to the contents to the contents of cAMP of hypophysis, septal nucleus, caudatum, hypothalamus and thalamus.

A novel alteration in the structure of an activated c-myc gene in a variant t(2;8) Burkitt lymphoma.

We have characterized a variant Burkitt lymphoma in which translocation joins the immunoglobulin kappa locus on chromosome 2 to the c-myc gene on chromosome 8. This Burkitt lymphoma is especially interesting because, in contrast to the more common lymphomas that carry 8;14 translocations, it carries a translocation that involves a light chain locus and occurs 3' to and at least 20 kb downstream of the c-myc gene. Furthermore, the c-myc gene from the translocated chromosome is abnormally expressed in that there is a characteristic shift in c-myc promoter utilization and an increase in c-myc transcript. These disturbances could be explained by novel structural alterations that occur in the c-myc gene and include a duplication of a 2.5 kb segment of DNA containing the two c-myc promoters and their untranslated leader exons. Interestingly, these alterations arise at a considerable distance from the translocation breakpoint.
