The activity of cancer procoagulant in cases of uterine leiomyomas.

PURPOSE: It is currently believed that cancer procoagulant (CP), an enzymatic protein, is a product of malignant neoplastic cells. The present study was designed to test whether it is also synthesized by benign neoplastic cells, namely uterine leiomyomas. MATERIALS AND METHODS: We determined the activity of CP in the blood serum of women with uterine leiomyomas (N = 24), normal women (N = 15), and genital cancer patients (N = 6) by the coagulative method according to Gordon and Benson. Also, the CP activity in 10% tissue homogenates of uterine leiomyomas, normal uterine muscle and tissues of cervical and endometrial carcinoma was determined by the chromogenic method according to Colucci et al. RESULTS: The mean CP activity in the sera of women with uterine leiomyomas was 181.1 seconds (s) +/- 19.9 s, in healthy women--293.2 s +/- 33.8 s, and in genital cancer patients--78.8 +/- 18.5 s (all differences: p < 0.001). Similarly, in homogenates of uterine leiomyomas the CP activity was 19.6 +/- 3.8 nmoles pNa/ml, in normal uterine muscle it was 13.2 +/- 2.2 nmoles pNa/ml, and in cancerous tissue--28.0 +/- 6.6 nmol pNa/ml (all values being significantly different from each other). There was a strong correlation (r = -0.8122; p < 0.001) between the CP activity in uterine leiomyomas and serum activity, suggesting that the source of the serum CP activity was from the leiomyoma. The coagulation time of 120 to 240 s by the Gordon and Benson method supported the diagnosis of uterine leiomyoma, and a value below 120 s--the suspicion of genital cancer. CONCLUSIONS: Uterine leiomyomas, representing benign genital neoplasia, synthesize CP and are the likely origin of CP activity in blood, as has been described for malignant tumors, but to a lesser degree. There may be a role for CP as a tumor marker of genital neoplasia.

[Diagnostic value of testing the procoagulant activity of neoplasms in cases of stomach and esophageal neoplasms].

The cancer procoagulant (CP) activity has been evaluated in homogenates of stomach and esophagus cancer tissues and in patients serum. Activity of CP in homogenates of stomach and esophagus cancer tissues as well as in serum of examined cancer patients were statistically higher than in the control group. The data indicate that estimation of CP activity in the neoplasmic tissues homogenates and in serum may be suggested as an new biochemical marker of cancer process useful in oncological diagnosis.

[Cancer procoagulant activity in cases of esophageal, stomach and colorectal cancer considering progression degree and histological type of cancer].

The cancer procoagulant activity has been evaluated in homogenates of esophagal, stomach and colorectal cancer tissues and in the blood serum of patients with these neoplasms's. Activity of CP was significantly higher in examined material than in control. The correlation between CP activity and progression degree as well as histological type was affirmed. The higher activity of CP in homogenates as well as in serum was observed in cases with higher degree of clinical progression and smaller activity of this enzyme corresponded with lower degree of the cancer progression. The highest activity of CP was observed in the cases of adenocarcinoma whereas the lowest in cases of squamous cell carcinoma. Higher activity of CP in homogenates of examined tissues correlated with higher activity of this enzyme in the serum. Activity of CP depended on the tissue localisation of the cancer and the highest was in the cases of stomach cancers whereas the lowest was in the cases of esophagal cancer.

The influence of green tea on the activity of proteases and their inhibitors in plasma of rats after ethanol treatment.

Ethanol oxidation in the liver is accompanied by formation of acetaldehyde and free radicals. These compounds can react with biologically active proteins, including proteolytic enzymes and their inhibitors. The aim of this paper was to determine the influence of green tea on the activity of cathepsin G and elastase and their inhibitors such as alpha-1-antitrypsin and alpha-2-macroglobulin, total antioxidant status and lipid peroxidation in plasma of young rats chronically intoxication with ethanol. The activity of cathepsin G and elastase was increased, while the activity of their inhibitors was reduced after ethanol treatment. AT the same time, the total antioxidant status was significantly decreased while lipid peroxidation measured as malondialdehyde and 4-hydroxynonenal was significantly increased. Giving green tea to rats did not change the proteases and their inhibitors activity, but significantly increased total antioxidant status and decreased lipid peroxidation. Drinking green tea with ethanol partially prevents the changes observed after ethanol intoxication.

Cancer procoagulant as a marker in monitoring the therapy in cases of oesophageal, stomach and colorectal cancer.

Cancer procoagulant activity in the blood serum of patients with oesophagal, gastric and colorectal cancer was evaluated before and after the tumour removal. Cancer procoagulant activity was significantly higher before the operation in comparison to the control group and was reduced after a total operative procedure, whereas it was kept on a high level after a non-radical procedure or in cases of metastases. Examination results point to the possibility of using the evaluation of cancer procoagulant activity in monitoring the course of treatment of patients with oesophagal, gastric and colorectal cancer.

[Diagnostic significance of cancer procoagulant activity in colorectal cancer].

This study aimed at evaluating diagnostic significance of cancer procoagulant (CP) activity in the homogenates of colon cancer tissues and in blood serum of patients with this neoplasm. Procoagulant activity, depending of specific cancer procoagulant, has been found in all examined tissues as well as in blood serum of cancer patients. CP activity in homogenates of colon cancer tissues as well as in blood serum of the examined cancer patients has been markedly higher than in the normal subjects. These data indicate that CP activity in the neoplastic tissue homogenates and in blood serum may be of value in the diagnosis of cancer.

[Cancer procoagulant (CP): the new biochemical marker in oncologic diagnosis].

This review describes the physicochemical and biological properties of cancer procoagulant (CP). This procoagulant is cysteine protease which directly activates coagulation factor X to factor Xa. CP is found in the tissues and blood of subjects suffering from cancer so its determination could be very useful in diagnostics and prognostics of cancer disease.

Evaluation of CA 15-3 serum levels in breast cancer patients.

The usefulness of post operative serum CA 15-3 determination was evaluated in a group of 57 breast cancer patients. CA 15-3 was assayed at baseline and every 3 months for a median follow-up of 34 months. Recurrence-free survival probability for patients with elevated serum CA 15-3 levels before surgery was 53.5% versus 88% for patients with normal serum CA 15-3 levels (p < 0.201). In 12 of the 15 patients with increased trend for CA 15-3 during follow-up, elevated CA 15-3 levels preceded (10 patients) coincided (2 patients) with the clinical detection of tumour recurrence, providing a predictive value of an increased trend of 80%. In the group of 10 patients who showed elevated serum CA 15-3 levels prior to the detection of recurrence, the median load-time was 5 months/(range 3-11 months). We concluded that CA 15-3 had prognostic value in breast cancer. Serial determinations of CA 15-3 was consistent with the treatment results.

Activity of cancer procoagulant (CP) in serum of patients with cancer of lung, breast, oesophagus and colorectum.

Activity of cancer procoagulant (CP) was studied in blood serum of 90 patients with cancer of lung, breast, oesophagus and colorectum, and of 15 healthy people. The activity of CP was determined by the coagulation method. Sera of patients with cancer showed higher mean activity of CP than sera of healthy control. Of the 90 cancer patients 78 were identified correctly by this test as having cancer (sensitivity 85%). In the case of lung and colorectal cancers the higher CP activity was observed the more advanced was the clinical stage of cancer, and the test was positive in 100%. After radical removal of malignant tumor of lung, decreased CP activity was found.

[Cathepsin D activity in amniotic fluid and serum during labor and puerperium in normal cases and those complicated by EPH-gestosis].

Cathepsin D activity during labour and puerperium in normal cases and those complicated by mild or moderate EPH-Gestosis was examined. Sera from maternal and umbilical cord blood, as well as the extraplacental blood and amniotic fluid were used for determinations. The gestational age of physiologic pregnancies and pregnancies complicated by EPH-Gestosis was 40 weeks. The first stage of labour was from 4 to 6 hours. The second stage of labour was over 10 minutes. In the group of women with mild and moderate EPH-Gestosis, the maternal serum activities on the third and fifth day of puerperium were significantly lower than in nonaffected subjects. In patients with moderate EPH-Gestosis, the activities in the maternal sera on the first day of puerperium, and in the umbilical blood were decreased when compared to controls. Uniformly, activities of the studied enzyme have been found to be lower in the umbilical cord blood sera than in the sera of the parturients. In cases of mild and moderate EPH-Gestosis the enzyme activity in the amniotic fluid was much lower than in controls, whereas in the extraplacental blood it was much higher.

Cancer procoagulant--CP.

A review of literature concerning cancer procoagulant (CP) has been carried out. This procoagulant directly activates coagulation factor X to factor Xa. Possibilities of utilising determinations of this activator in diagnostics and prognostics of the cancerous disease are discussed.

Tissue factor pathway inhibitor (TFPI).

Tissue factor pathway inhibitor (TFPI) is a plasma proteinase inhibitor. It is a 42 kD glycoprotein that consists of 276 amino acid residues which sequence is known. TFPI is synthesized by vascular endothelial cells and part of it is associated with glycosaminoglycans of these cells. In blood TFPI is found in a free-form (active) and in an associated with lipoprotein form (nonactive). TFPI directly inhibits activated factor Xa and then factor VIIa/TF complex. Decreasing TFPI activity facilitates an activation of blood coagulation and fibrin forming, increasing TFPI activity inhibits these processes.

Cancer procoagulant (CP) in lung cancer.

Lung cancers (squamous cell carcinoma, microcellular carcinoma, macrocellular carcinoma and adenocarcinoma) show procoagulant activity. It mainly depends on the presence of cancer procoagulant (CP) in lung cancer cells.

Activity of liver proteases in experimental methanol intoxication.

Intoxication of rats with methanol (1.5 and 3.0 g/kg body weight) led to a significant, time- and dose-dependent decrease in the activities of cathepsins A, B and C, while the activity of cathepsin D was unaffected. The decrease was associated with a different partial release of individual cathepsins to the post-lysosomal fraction.

[Activity of cathepsin D in some neoplasms of the gastrointestinal tract].

In the present paper we analyzed cathepsin D activity in digestive tract cancers. Cathepsin D activity was estimated in 10% homogenates of oesophageal cancer, gastric cancer and colon cancer tissues and in the blood serum and expressed as the amount of liberated tyrosine which was assayed acc. to Folin-Ciocalteau. Mean cathepsin D activities in neoplastic tissues and normal counterparts were as follows: oesophaged cancer (218.5 mM Tyr/1/2 h vs 145.0 mM Tyr/1/2 h), gastric cancer (285.4 mM Tyr/1/2 h vs 142.3 mM Tyr/1/2 h) and colon cancer (233.7 mM Tyr/1/2 h vs 159.5 mM Tyr/1/2 h). In all examined neoplastic tissues cathepsin D activity was almost too-fold higher than in the normal counterparts. Cathepsin D activity in the sera of cancer patients was too a lesser degree higher than in the sera of normal subjects. The data indicate that estimating of cathepsin D activity in the neoplastic tissues homogenates and in the blood serum may be of diagnostic value and may constitute an information which is complementary to the analysis of other tumor markers and histopathologic examination.

[Tissue factor pathway inhibitor (TFPI) and its role in pathology].

Tissue factor pathway inhibitor (TFPI) is an important regulator of blood coagulation. It was described as antyconwertin (AC), extrinsic pathway inhibitor (EPI) and lipoprotein-associated coagulation inhibitor (LACI). TFPI is a glycoprotein, which is mainly synthesized by vascular endothelial cells. The concentration of this enzyme in plasma is low (100 ng/ml). TFPI inhibits activity of factor Xa and complex FVIIa/TF. Changes TFPI activity play an important role in coagulation disorders and presently the function of TFPI in various kinds of diseases is discussed.

Complexoproductive and antiheparin properties of low density lipoproteins (LDL). III. Interaction of low density lipoproteins (LDL) with synthetic polyanionic compounds.

Low density lipoprotein (LDL) form insoluble complex bonds with polymetacrylic and polyvinylsulphonic acids in the presence of calcium chloride. Quantity of arising complexes depends on the concentrations of LDL, acids and calcium chloride and on pH. These complexes dissociate solutions of sodium chloride, urea and guanidine hydrochloride.
