Tissue distribution of a novel cell binding protein, osteoadherin, in the rat.

Osteoadherin is a cell binding keratan sulfate proteoglycan which was recently isolated from mineralized bovine bone and subsequently cloned and sequenced. For studies of osteoadherin expression in rat tissues we isolated and sequenced a 1.3-kbp partial cDNA covering most of the coding region using a rat calvaria cDNA library. The most 5' end of the cDNA was obtained by reverse transcription PCR from the bone total RNA preparation. The deduced, translated protein sequence containing 423 amino acid residues shows high sequence identity to mouse, bovine and human osteoadherin except in the very acidic C-terminal region. However, the rat counterpart showed a similarly high content of acidic amino acid residues. Ribonuclease protection assay showed osteoadherin mRNA to be expressed in femoral bone and calvaria tissues, while no expression was detected in cartilage, tendon or liver. Using very sensitive nested RT-PCR, however, message was detected in femoral head, rib, tendon and bone marrow total RNA preparations. An antiserum specific for the rat C-terminal region of osteoadherin was generated and used for studies of protein distribution by immunohistochemistry during femoral head development. Osteoadherin was primarily present in bone trabeculae and no staining was seen in cartilage. In situ hybridization showed the strongest expression in osteoblasts close to the cartilage/bone interface of the growth plate and lower expression in diaphyseal osteoblasts. On maturation of the femoral head on day 60 some expression was detected immediately below the forming articular cartilage. Our data indicated that osteoadherin is primarily expressed by osteoblasts and might have a role in regulation of mineralization.

Cellular dysfunction of LQT5-minK mutants: abnormalities of IKs, IKr and trafficking in long QT syndrome.

Mutations in the minK gene KCNE1 have been linked to the LQT5 variant of human long QT syndrome. MinK assembles with KvLQT1 to produce the slow delayed rectifier K+ current IKs and may assemble with HERG to modulate the rapid delayed rectifier IKr. We used electrophysiological and immunocytochemical methods to compare the cellular phenotypes of wild-type minK and four LQT5 mutants co-expressed with KvLQT1 in Xenopus oocytes and HERG in HEK293 cells. We found that three mutants, V47F, W87R and D76N, were expressed at the cell surface, while one mutant, L51H, was not. Co-expression of V47F and W87R with KvLQT1 produced IKs currents having altered gating and reduced amplitudes compared with WT-minK, co-expression with L51H produced KvLQT1 current rather than IKs and co-expression with D76N suppressed KvLQT1 current. V47F increased HERG current but to a lesser extent than WT-minK, while L51H and W87R had no effect and D76N suppressed HERG current markedly. Thus, V47F interacts with both KvLQT1 and HERG, W87R interacts functionally with KvLQT1 but not with HERG, D76N suppresses both KvLQT1 and HERG, and L51H is processed improperly and interacts with neither channel. We conclude that minK is a co-factor in the expression of both IKs and IKr and propose that clinical manifestations of LQT5 may be complicated by differing effects of minK mutations on KvLQT1 and HERG.

Novel intestinal Helicobacter species isolated from cotton-top tamarins (Saguinus oedipus) with chronic colitis.

A disease similar to ulcerative colitis in humans has been identified in cotton-top tamarins (CTTs) in captivity. The clinical signs include weight loss, diarrhea, and rectal bleeding with the pathological features and biochemical abnormalities of ulcerative colitis. Approximately 25 to 40% of these animals develop colon cancer after 2 to 5 years of captivity. An infectious etiology has been proposed; however, no microbial agent to date has been identified. Helicobacter spp. have been associated with enterocolitis and inflammatory bowel disease (IBD) in humans and animals. Infection with Helicobacter pylori or Helicobacter mustelae is associated with an increased risk of gastric adenocarcinoma and lymphoma of the mucosa-associated lymphoid tissue. Helicobacter hepaticus causes hepatitis, hepatic adenomas, and hepatocellular carcinomas in susceptible strains of mice. The aim of this study was to assess a colony of CTTs with a high incidence of IBD and colon cancer for the presence of colonic Helicobacter spp. A fusiform, gram-negative bacterium with bipolar flagella and periplasmic fibers was isolated from the feces of CTTs. The bacterium grew under microaerobic conditions at 37 and 42 degrees C but not at 25 degrees C, did not hydrolyze urea, was positive for catalase and oxidase, did not reduce nitrate to nitrite, did not hydrolyze indoxyl acetate or alkaline phosphatase, and was resistant to nalidixic acid, cephalothin, and trimethoprim-sulfamethoxazole. On the basis of 16S rRNA gene sequence analysis, the organism was classified as a novel Helicobacter species. This is the first Helicobacter isolated from CTTs. Further studies are needed to elucidate the role of this novel Helicobacter sp. in the pathogenesis of ulcerative colitis and colonic adenocarcinoma in CTTs.

Isolation of Helicobacter canis from a colony of bengal cats with endemic diarrhea.

On the basis of biochemical, phenotypic, and 16S rRNA analyses, Helicobacter canis was isolated from Bengal cats with and without chronic diarrhea. Because the cats were coinfected with other potential pathogens, including Campylobacter helveticus, and because H. canis was isolated from nondiarrheic cats, the causal role of H. canis in producing the diarrhea could not be proven. Histologically, the colons of the four affected cats were characterized by mild to moderate neutrophilic, plasmacytic, and histocytic infiltrates in the lamina propria. Rare crypt abscesses were also noted for three cats but were a more prominent feature of the colonic lesions noted for the fourth cat. This is the first observation of H. canis in cats and raises the possibility that H. canis, like H. hepaticus and H. bilis in mice, can cause inflammation of the colon, particularly in hosts with immune dysregulation. Further studies are needed to determine the importance of H. canis as a primary enteric pathogen in cats and the role of cats in the possible zoonotic spread of H. canis to humans.

[Breast reconstruction using pedicled three-lobe transverse rectus abdominis musculocutaneous flap].

OBJECTIVE: An innovated transverse rectus abdominis musculocutaneous flap, the three-lobe TRAM flap was designed for breast reconstruction. METHODS: A triangle-shaped skin over the upper part of the rectus abdominis muscle was included in the conventional TRAM flap for the purpose of adding more skin for breast reconstruction and enhancing blood supply of the skin portion of the TRAM flap. RESULTS: In this group of six patients subjected to mastectomy and radiation therapy for breast cancer, complete survival of the three-lobe TRAM flap was obtained in 4 cases. Two patients had minor skin necrosis and fat liquefaction. Better breast contour and contracture release of the axilla were also achieved. CONCLUSION: This innovation of conventional TRAM flap for breast reconstruction has some advantages. It is especially useful for patients complicated with radiation lesions on the chest and axilla areas.

[Management of destructive electrical burns of wrist].

OBJECTIVE: To reduce amputation rate of most severe pattern (Type III) of electrical burns of wrist. METHODS: Early wrist decompression followed by selective transfusion of dextran, PGE1, low molecule heparin to prevent thrombosis of injured radial and ulnar arteries, vascular bridging across the distal forearm and wrist to the hand soon after the appearance of circulatory failure to the hand. Free tissue transfer, especially the use of greater omentum and the innovated abdominal flaps, to repair the circumferential wounds of the wrists are recommended. RESULTS: A group of 102 limbs of 90 patients were treated in the last 10 years, among them the amputation rate of most severe pattern (Type III) of electrical burns of wrists was significantly reduced from 80% to 38.9% compared with 10 years before. CONCLUSION: A comprehensive treatment including the use of anticoagulation drugs, vascular reconstruction, sophisticated surgical repair of the wound are effective in reducing the amputation rate of electrical burns of wrists.

[Human breast myoepithelial cell blocks estrogen receptor-negative breast carcinoma cell invasion].

OBJECTIVE: To investigate the effect of mammary myoepithelial cell on breast cancer cell infiltration. METHODS: By immunohistochemistry, Western Blot, Northern Blot and Matrigel invasion experiments, we detected the maspin in breast cancer cell and myoepithelial cell, and studied the machanism of breast cancer cell invasion inhibition by myoepithelial cell. RESULTS: Histologically, ducts and ductal carcinoma in situ (DCIS) were surrounded by an intact circumferential layer of myoepithelial cells. In invasive carcinoma, the myoepithelial cell layer appeared disrupted. In matrigel invasion experiments, myoepithel cell and its condition medium could block the ER-negative breast carcinoma cell line MDA-MB-231 cell invasion. The proteinase inhibitor most dramatically up-regulated in myoepithelial cells was the tumor suppressor maspin which was only highly expressed in myoepithelial cells, not in breast cancer cells. CONCLUSION: Mammary myoepithelial cells could inhibit tumor cell invasion, suggesting that myoepithelial cell might have antitumor effect in the regulation of breast cancer development and progression in vivo.

The relationship between the levels of granulocyte colony-stimulating factor and leukocytosis induced by all-trans retinoic acid in acute promyelocytic leukemia.

OBJECTIVE: To explore the mechanism of leukocytosis. METHODS: Enzyme linked immunosorbent assay (ELISA) method was used for detecting levels of serum granulocyte colony-stimulating factor (G-CSF) in 47 cases of acute promyelocytic leukemia (APL) during the treatment with all-trans retinoic acid (ATRA). RESULTS: The peak of increased serum G-CSF level occurred on the 9th day, and WBC number was the highest on the 11th day. After ATRA treatment, both serum G-CSF level and WBC number increased in 68.1% of the cases. In 19.2% of the cases treated, serum G-CSF level was increased but without obvious change in WBC number, and the reverse was true in 12.7% of the cases. CONCLUSION: Serum G-CSF level was statistically correlated to the number of WBC, promyelocytes and its late stage by Spearman's rank-order correlation coefficient.

Differentiation therapy for acute promyelocytic leukemia with all-trans retinoic acid: 10-year experience of its clinical application.

PURPOSE: To summarize the experiences in using all-trans retinoic acid (ATRA) for the differentiation therapy for acute promyelocytic leukemia (APL) since our introduction of its use in clinic in 1986. DATA RESOURCES: Data resources came from Chinese Journal of Hematology, English-language literature using MEDLINE (1988-1998), book entitled "Treatment of Malignancies by Inducing Differentiation and Apoptosis" published by Shanghai Publishing House on Sciences and Technology, and our recent data to be published. STUDY SELECTION: Thirty-five articles related to the purpose of this review were reviewed. DATA EXTRACTION: Data were checked for their quality, reliance and originality. RESULTS: ATRA combined with chemotherapy can decrease the incidence of retinoic acid syndrome and produce a very high remission rate (90%-95%). Post-remission treatment should include chemotherapy and ATRA, the 5-year survival probability was able to attain 0.71 +/- 0.06. The main problem in the treatment is early tolerance to ATRA and relapse of the disease. The most effective treatments for relapsed APL is to use arsenic trioxide. CONCLUSION: The combination of ATRA, chemotherapy and arsenic oxide in the treatment of APL for elevating the remission rate and prolonging survival time deserves further study.

[Mechanism of growth inhibition by genistein of human breast carcinoma].

OBJECTIVE: To study the mecnanism of inhibition of breast carcinoma cells by genistein. METHODS: Human breast cancer cell lines were treated in vitro with genistein. Northern blot, Western blot, plasmid transfection and apoptosis assay were used to evaluate the mechanism of cell growth inhibition. RESULTS: Genistein could inhibit proliferation of human breast carcinoma cells with different ER status and p53 status. This inhibition was associated with induction of p21WAF1/CIP1 mRNA and protein expression. Exposure of MCF-7 and MDA-MB-231 cells to genistein resulted in statistically significant increased p21WAF1/CIP1 mRNA stability, accompanied with increased p21WAF1/CIP1 protein expression. This increase in p21WAF1/CIP1 was subsequently followed by the onset of apoptosis. CONCLUSION: The inhibitory effect of genistein on cell proliferation of breast cancer appears to be due to p21WAF1/CIP1 expression and apoptosis through mechanism independent of ER and p53. The results provide evidence for the potential usefulness of genistein in the prevention and treatment of human breast cancer.

[Prognostic significance of cyclin-dependent kinase inhibitor p27kip1 expression in human breast cancer].

OBJECTIVE: To study the expression of p27kip1 in breast cancer in relation to biological behavior and prognosis. METHODS: p27kip1 expression at protein level was determined by immunohistochemistry technique in 181 patients with breast cancer. In addition, p27WAF1/CIP1, p53 and mdm2 were examined in 106 cases of this series. Data about apoptotic index, bcl-2 and bax protein expression were available in the other 91 patients. RESULTS: Of the 181 patients, p27kip1 expression was low in 121 (66.9%) and high in 60 (33.1%), respectively. p27kip1 expression level was correlated with histological grade, tumor size, axillary lymph node status, post-operative recurrence, invasion and metastasis. It was also correlated with bcl-2, bax, p53, p21WAF1/CIP1, and mdm2 expression levels. Univariate analysis demonstrated better disease-free survival in patients with higher p27kip1 expression than in those with lower expression. However, by Cox multivariate analysis, p27kip1 expression level was not found to be of independent prognostic significance. CONCLUSION: As an inhibitor of cyclin-dependent kinase, p27kip1 favors prognosis of human breast cancer not only by arresting cell cycle in G1 phase but also inducing cancer cell apoptosis.

[Immunoregulation of Youguiyin, Sijunzitang, Taohong Siwutang in treating patients with deficiency of kidney, spleen and blood stasis syndrome].

OBJECTIVE: To study the different immunoregulatory effects of Youguiyin, Sijunzitang, Taohong Siwutang to patients with deficiency of Kidney, Spleen and blood stasis Syndrome respectively. METHODS: The treated group, 40 patients in each with deficiency of Kidney, Spleen and blood stasis were treated with Youguiyin, Sijunzitang and Taohong Siwutang respectively. Three control groups (20 patients each) were established respectively. Peripheral venous level of complement C3, circulating immunocomplex (CIC), interleukin-2 (IL-2), interferon-gamma (IFN-gamma), lymphocyte transformation rate (LTR), the activity of natural killer (NK) cell, and the phagocytic function of neutrophil were measured before and after treatment. RESULTS: Sijunzitang could reduce the level of CIC (P < 0.01), raise the activity of NK cell and LRT (P < 0.01) of the patients with deficiency of Spleen syndrome; the same did the Youguiyin to that of patients with deficiency of Kidney syndrome. Also, Sijunzitang could increase the level of IL-2 in blood and the phagocytic function of neutrophil (P < 0.05); the Taohong Siwutang could lower the level of CIC to patients with blood stasis syndrome (P < 0.01), increase the activity of NK cells (P < 0.05). CONCLUSION: Youguiyin, Sijunzitang could enhance the immunoregulatory function of the patients with deficiency of Spleen and Kidney, particularly the cellular immune functions, in which the Sijunzitang effect is more complete and significant, while Taohong Siwutang eliminated the effect of CIC in blood better.

[Comparative study of the regulation of kidney tonic and promoting blood circulation recipe on apoptosis in T cells of old rats].

OBJECTIVE: To investigate the mechanism responsible for the observed reduction in T cell function of the Kidney Deficiency Syndrome. METHODS: After 18th activation with anti-CD3 mAb, the qualitative and quantitative analysis of apoptoses in T cells were performed by using DNA gel electrophoresis, TdT-mediated dUTP nick end labelling (TUNEL) and flow cytometry. RESULTS: The percentage of apoptotic cells was 47.0% in the old and 22.2% in the young group (P < 0.01). In the Kidney tonic group, the mean percentage of apoptotic cells was 31.2%, significantly lower than that in the aged group (P < 0.05). The mean percentage of apoptotic cells in the promoting blood circulation group was 43.3% and no significant difference was seen in the percentages of apoptotoic T cells from the promoting blood circulation group compared with the old group. CONCLUSION: Activation-induced excessive apoptosis of T cells is one of the mechanisms elucidating decreased T cell function of the Kidney Deficiency Syndrome and aging, the retardation of immune senescence by Kidney tonic recipe could be accounted for by down-regulation of activation-induced apoptosis in T cells.

[Effect of three composite recipes on content of monoamine transmitters in hypothalamus induced by intracerebroventricular injection of interleukin-1 of rats].

OBJECTIVE: To explore the different effects of Youguiyin decoction, Sijunzi decoction, Taohong Siwu decoction on the content of monoamine transmitters in the hypothalamus induced by cytokines. METHODS: A reactive status of hypothalamic monoamine transmitters to cytokine in rat was built by observing the content change of monoamine transmitters in hypothalamic homogenate after intracerebroventricular (icv) injection of human recombinant interleukin-1 beta(IL-1 beta). On this basis, the different effect of three composite recipes on this status was observed. RESULTS: The content of norepinephrine (NE) declined significantly compared with control group after icv injection of IL-1. Compared with IL-1 group, Youguiyin further reduced the content of NE induced by icv injection of IL-1, while Taohong Siwu decoction and Sijunzi decoction did not show any effect on this status. CONCLUSIONS: It seems that Youguiyin for tonifying Kidney could increase the reactivity of hypothalamic NE neuron, Sijunzi decoction for invigorating Spleen and Taohong Siwu decoction for promoting blood circulation did not show this effect.
