p53 independent G1 arrest and apoptosis induced by adriamycin.

The biological activity of adriamycin was investigated in human breast carcinoma (HBC) cells, Adriamycin inhibited the growth of a number of HBC cell lines and induced G1 arrest followed by apoptosis. In MCF-7 cells that harbor wild-type p53, adriamycin-induced G1 arrest and apoptosis was accompanied by p53-independent regulation of WAF1/CIP1 as well as bax mRNA levels. In MDA-MB-231 cells which possess a mutant p53, adriamycin-induced G1 arrest and apoptosis was also associated with a concomitant up-regulation of WAF1/CIP1 mRNA while these cells did not express bax or bcl-2 messages. Thus, adriamycin induces G1 arrest and apoptosis via a unique pathway which appears to involve activation of downstream effectors of p53-independent manner.

[The preparation of poly (DL-lactide) hollow fiber and the release of drug in vitro].

The biodegradable hollow fibers of DL-PLA were prepared by a dry-wet phase inversion spinning process. By using four different spinning systems, DL-PLA hollow fibers with varying asymmetric membrane structure were obtained. The structure of hollow fiber wall was examined by SEM. In vitro release of norethisterone from the DL-PLA hollow fibers was also investigated. The hollow fibers were filled with a 50 wt% NET in castor oil. The DL-PLA hollow fibers NET release was found to be dependent on the membrane structure of the hollow fiber wall. Possible release mechanisms were discussed.

[Measurement and evaluation of radiation dose distribution of gamma-ray altimeter in static test for recovery capsule during simulated landing].

The gamma-ray altimeter, containing a gamma-radioactive source 137Cs with activity of 2.96 x 10(10) Bq, was used to detect the height of the spacecraft recovery capsule during landing. To measure the gamma-dose distribution near to the guidance section, especially at position where the useful load is located in recovery capsule, gamma-dose field at the test-site for one machine test was measured by means of a gamma-dose rate meter FJ-317C and a gamma-dose meter ANRI-01-02. The result showed that the absorbed dose rate at 5 meters from gamma-source on the ground is as low as the radiation protection limit when the altimeter is down to 0.16 m from the ground. The ground reflection effect for gamma-ray decreases as height of the recovery capsule increases. Therefore, the gamma-dose level at useful load in the recovery capsule will meet the requirements of flight safety.

[Investigation on the elimination of organic substances in urine by supercritical water oxidation].

Application of Supercritical Water Oxidation (SCWO) to processing of human metabolic wastes of Controlled Ecological Life Support System was studied for organic substance elimination in urine and recover potable water. The SCWO of simulated urine was investigated with oxygen as the oxidant at 400-500 degrees C and 24-30 MPa in a continuous flow reactor. The organic substance elimination efficiency was obviously effected by temperature, pressure and reaction time. It is lower than 10% at 400 degrees C, but more than 95% at 550 degrees C with reaction time of 2 min.

[Preliminary study on the arsenic trioxide-induced NB4 cell apoptosis and its molecular mechanisms].

OBJECTIVE: To illustrate the possible cellular and molecular mechanisms of arsenic trioxide (As2O3) in the treatment of acute promyelocytic leukemia (APL). METHODS: APL cell line NB4 was used for in vitro studies. The effect of As2O3 on APL was studied by using flow cytometry, DNA electrophoresis, Narthern blotting and Western blotting. RESULTS: As2O3 induced NB4 cell apoptosis, while not inhibiting the growth and survival of two other leukemic cell lines (HL-60 and U937). Furthermore, As2O3 effectively down-regulated the expression of bcl-2 gene without changing the mRNA levels of other apoptosis-associated genes (including p53, c-myc, bax and bcl-XL). CONCLUSION: These might be one of the molecular mechanisms of As2O3 induced NB4 cell apoptosis.

[Effects of arsenic trioxide on the subcellular localization of PML/PML-RARalpha protein in leukemic cells].

OBJECTIVE: In order to illustrate the possible roles of PML-RARalpha protein in arsenic trioxide (AsO3)-induced NB4 cell apoptosis. METHODS: Effects of As2O3 on the subcellular localization of PML-RARalpha in NB4 cells were studied. RESULTS: (1) Anti-PML serum staining was reduced and PML granules emerged in the perinuclear cytoplasm in a diffuse pattern in HL-60 cells under As2O3 treatment; (2) abnormal PML/PML-RARalpha granules were decreased; (3) NB4 cells accumulated anti-PML serum staining granules in the cytoplasms were increased and similar accumulation also found in apoptotic cells; and (4) pretreatment with all-trans retinoic acid (ATRA) for 24 or 48 hours did not alter the As2O3 effects. CONCLUSION: As2O3-induced apoptosis was independent of the retinoic acid signal pathway, and it might be regulated by PML/PML-RARalpha and/or other related genes.

[Pharmacokinetics of intravenous arsenic trioxide in the treatment of acute promyelocytic leukemia].

OBJECTIVE: In order to study the pharmacokinetics and metabolism of arsenic trioxide (As22O3), and its major side effects. METHODS: The clinical pharmacokinetics of intravenous As2O3 in 8 relapsed acute promyelocytic leukemia (APL) patients were studied by Gas-phase chromotography. RESULTS: (1) After 2 hour intravenous drips of 10mg As2O3, drug plasma maximal concentration (Cpmax) was 0.94 +/- 0.37mg/L(x +/- s), time to peak concentration (Tp) was 4hrs, T1/2 alpha and T1/2beta were 0.89 +/- 0.29hrs and 12.13 +/- 3.31hrs, respectively, apparent volume of distribution (Vc) was 3.83 +/- 0.45L, system clearance (CLs) was 1.43 +/- 0.17L/h and area under curve (AUC) was 7.25 +/- 0.97mg x h/L. Importantly, the continuous administration of As2O3 did not alter the pharmacokinetic behaviors. (2) During As2O3 therapy, 24-hour arsenic content in urine accounted for 1%-8% of the total dose (10mg) per day. And it decreased gradually after drug withdrawal. (3) Arsenic contents in nail and hair were increased continuously during As2O3 therapy. The peak concentrations could be five to seven-fold higher than that pre-treatment. It also decreased gradually after drug with drawal. CONCLUSION: As2O3 is a relatively safe and effective drug in the treatment of APL patients, in spite of certain accumulation in some tissues.

[Serum G-CSF levels in patients with chronic aplastic anemia].

OBJECTIVE: To explore the pathogenesis of aplastic anemia. METHODS: The spleen cells of BALB/c miceimmunized with rhG-CSF were fused with myeloma cell line SP2/0. Three murine hybridomas producing monoclonal antibody (McAb) against rhG-CSF were obtained. Serum levels of G-CSF in 54 cases of chronic aplastic anemia (CAA) were detected by ELISA method. RESULTS: Serum G-CSF levels in 70.4% (38 of 54 cases) of the CAA patients were increased (272.76 +/- 58.39ng/L). There was a negative correlation between blood neutrophil counts and serum G-CSF levels (r = -0.535, P<0.01). CFU-GM were detected in 12 cases of CAA, and all was decreased (17.92 +/- 10.28/2 x 10(5) nucleated cell). Serum G-CSF levels in almost 30% (12 of 54 cases) of the CAA patients were not increased. CONCLUSION: The hematopoietic progenitor cells of CAA patients were impaired, and some CAA could be treated with rhG-CSF and immunosuppressive agents.

[In vitro study on arsenic trioxide-inducing apoptosis in primary acute promyelocytic leukemie cells].

OBJECTIVE: To illustrate mechanisms of arsenic trioxide(As2O3) in the treatment of acute promyelocytic leukemia (APL). METHODS: Cell-DNA content distribution, CD11b and CD33 antigens and nitroblue tetrazolium (NBT) reduction were evaluated in fresh APL cells from six APL patients treated in vitro with As2O3. RESULTS: As2O3 had double effects on the cells inducing apoptosis and inducing partial differentiation at higher and at lower drug concentrations, respectively. As2O3 (0.1 approximately 2.0 micromol/L) could rapidly modulate and degrade APL-specific marker molecule PML-RARalpha protein, which could play an important role in the effects of As2O3 on APL cells. CONCLUSION: As2O3 had double effects (induction of apoptosis and partial differentiation) on APL cells through the modulation and degradation of PML-RARalpha proteins.

[A gas chromatographic study on the characters of adsorbents in the enrichment of organic compounds from air samples].

A gas chromatographic method for the measurement of enriching characters of adsorbents used in preconcentration of organic compounds from air samples was studied. The character parameters suggested in this study include adsorption parameters (breakthrough volume or maximum sample volume) and desorption ones (minimum flashing volume and the lowest desorption temperature). The parameters of some low-molecular-mass compounds such as alcohols, saturated hydrocarbons, aromatics, esters and halohydrocarbons on GDX-301 were measured by using elution GC method. The maximum sample volumes of all the 20 compounds (with exception of methanol) got from these experiments are not less than 0.8 L at 35 degrees C for sampling tube (filled with 2 g GDX-301) and 0.3 L at 0 degrees C for focus tube (with 0.14 g GDX-301). The corresponding minimum flashing volumes of less than 177 mL and 13 mL respectively are needed to complete desorption at 180 degrees C, and the lowest desorption temperatures of not over 180 degrees C and 157 degrees C at flashing volume of 180 mL and 30 mL are lower than the maximum allowable operating temperature (250 degrees C) of GDX-301. The results showed that GDX-301 is a good adsorbent for enrichment of the organic compounds mentioned above. The following operating conditions are recommended: using the above sampling tube and focus tube, actual sample volume < or = 0.8 L, adsorption temperature < or = 35 degrees C and focus temperature 0 degrees C, desorption for 6 min from sampling tube and 1 min from focus tube at 180 degrees C with flashing rate 30 mL/min. Good correlations between the parameters and boiling point of the investigated compounds were found. It can be used to estimate the parameters of other organic compounds not measured in this study.

[Acute fatty liver in pregnancy: a clinicopathologic study].

OBJECTIVE: To investigate the early recognition and management of milder cases of acute fatty liver in pregnancy (AFLP). METHODS: Twelve cases of AFLP treated in our hospital during the past two and half years were retrospectively studied with emphasis on symptoms, laboratory findings, liver biopsy and maternal complications. The 12 cases were classified into 2 groups. Group 1, 6 cases of advanced AFLP diagnosed clinically, were treated in the first year of the study. Group 2, 6 cases of milder AFLP diagnosed by postpartum liver biopsy, were treated during the last one and half years of the study. RESULTS: The mean gestational age at onset was 34 +/- 3 weeks. In the early stage, all cases had malaise, nausea, loss of appetite and epigastric distress followed by jaundice in the third trimester of pregnancy. Laboratory findings included raised transaminases (< or = 300 IU/L) and total serum bilirubin (32.5-510.8 micro mol/L) levels, hypoalbuminemia (18-30 g/L), hypofibrogenemia (< 2.4 g/L), prolonged prothrombin time and prolonged partial thromboplastin time. Maternal complications were frequent including hepatic encephalopathy (9), ascites (9), hypoglycemia (7), renal failure (3), hematemesis (4), preeclampsia (6), and postpartum hemorrhage (5). Cesarean sections were performed in 7 cases. In group 2, both mother and fetuses had 100% survival. However, the mortality of the mothers and fetuses in group 1 were both 50%. CONCLUSIONS: With increasing awareness, especially in the early recognition of AFLP cases and prompt progressive management, including early termination of pregnancy and large dose infusion of fresh frozen plasma, the prognosis of AFLP is obviously improved. Percutaneous liver biopsy should be done when the coagulation tests became normal and the amounts of ascites decreased after delivery.

[Prevention of bone loss by chlorotrianisene in oophorectomized rats].

OBJECTIVE: To determine the influence of chlorotrianisene on bone metabolism in oophorectomized rats. METHODS: Forty-eight Wistar female rats, 70 days of age, were randomly divided into four groups: SV group (sham operation + vehicle), OV group (oophorectomy + vehicle), SE group (sham (operation + chlorotrianisene) and OE group (oophorectomy + chlorotrianisene). Chlorotrianisene or vehicle 4 ml/kg was given i.p. daily for 45 days from day 7 of being ovariectomized or sham operated. At the time of death, uterine weight was measured, and the thoracic vetebra 12, left tibia were collected and made into decalcified bone specimens for histomorphometry. RESULTS: There were significant difference in the uterine weight among each group. Significant different appearance by bone histomorphometry was seen between OV and the other three groups, but not shown between SV and SE, OE groups. CONCLUSION: These results suggest chlorotrianisene could inhibit bone loss and delay the atrophy of uterus induced by ovariectomy in Wistar female rats. It has protective effects on bone like other estrogen preparations.

[Packing drainage of transmastoid approach for treatment of otogenic brain abscess: a report of 30 cases].

Thirty cases of patients with otogenic brain abscess (OBA) who received mastoid radical operations were treated simultaneously by means of packing drainage via the appoach of the mastoid operative cavity. 26 cases of those were cured. The results showed that the therapeutical method gave satisfactory effectiveness and had many advantages. It provided an effective montitor for retraction and had many vomica, and gave ample drainage for retraction in that it made OBA seldom recur. The operative procedure is fit for the great majority of OBA. The paper also discussed intraoperative and postoperative complications and other problems pertinent to the operation.

[The application of transposition of pectoralis major muscle or myocutaneous flap in the repair of long-standing wound over the mandibulo-cervico-thoracic area].

The pectoralis major muscle or myocutaneous flap is characterized by its easy dissection and free rotation, as well as abundance of blood supply and tissue mass of the flap, so that it was often used in repairing the wound over the mandibulo-cervico-thoracic area. From 1989 to 1996, 11 cases with long-standing wound of this area were difficult to repair with the traditional method, but were treated with transposition of this type of myocutaneous flap. The myocutaneous flap was applied in 8 case and muscle flap in 3 case. The flap area ranged from 12 cm x 8 cm to 26 cm x 18 cm. All the flaps survived, and excellent results were achieved. The technique and design of the operation were reported in details. It was concluded that the process was easy to perform, and the flap had abundant blood supply and high resistance to infection.

[The relationship between expression of basement membrane in squamous cell carcinoma of oral cavity and cervical lymph node metastasis].

Fifty seven cases of oral squamous cell carcinoma (SCC) were studied by immunohistochemical ABC method using type IV collagen and laminin antibody to investigate the relationship between expression and distribution of basement membrane in oral SCC and clinicopathologic characteristics and cervical lymph node metastasis. The distribution of basement membrane of oral SCC was discontinuous and in some cases the membranes disappeared. There was highly significant correlation between the staining patterns together with histologic differentiation degrees and cervical lymph node metastasis (P < 0.05). These indicate that the expression of baasement membrane in oral SCC may be a useful parameter for evaluation of tumor histologic differentiation and tumor invasion and metastasis.

Effects of vesnarinone, a novel orally active inotropic agent with an immunosuppressive action, on experimental cardiac transplantation in rats.

BACKGROUND: Vesnarinone (VES) has been used for treatment of patients with congestive heart failure. In addition to inotropic effects, it seems to have immunosuppressive action. We tested the hypothesis that VES suppresses graft rejection, inotropic dysfunction caused by early rejection, and chronic coronary obstruction in a heterotopic rat cardiac transplantation model. METHODS: (1) To study acute rejection, hearts from Lewis-Brown Norway (LBN) rats were transplanted into Lewis rats, which were treated with or without VES (50 or 100 mg/kg/day orally). (2) In a functional study, LBN hearts with or without VES (100 mg/kg/ day) were isolated and perfused on day 3 after transplantation to assess inotropic response to isoproterenol (3 x 10(-8) M). (3) To study chronic rejection, Lewis hearts were transplanted into Fisher 344 rats, which were treated with or without VES (50 mg/kg/day) for 90 days. Coronary obstructive disease was assessed by morphometric analysis. There were five to six animals in each group. RESULTS: (1) VES (100 mg/kg/day) prolonged LBN heart survival (11.7 +/- 0.7 vs. 9.6 +/- 0.7 days in control; P < 0.05). (2) Left ventricular developed pressure was depressed in transplanted hearts regardless of VES treatment (84 +/- 12, 90 +/- 8 vs. 144 +/- 16 mmHg in untransplanted hearts; P < 0.01). The developed pressure after administration of isoproterenol in VES-treated hearts (184 +/- 20 mmHg) was higher than transplanted hearts without VES (118 +/- 16 mmHg; P < 0.05), and similar to untransplanted hearts (203 +/- 27 mmHg; P = NS). (3) Transplanted hearts treated with or without VES showed similar grades of rejection (2.0 +/- 0.3 vs. 2.6 +/- 0.2; P = NS), intimal area (6,996 +/- 3,186 vs. 13,441 +/- 5,165 microns2; NS), and coronary luminal obstruction (45 +/- 16% vs. 67 +/- 14%; NS). CONCLUSIONS: VES produces mild prolongation in survival of rat heart grafts, but has no significant effect on chronic graft atherosclerosis. VES preserves the positive inotropic effects of isoproterenol that are otherwise deteriorated by early acute rejection.

Effect of all-trans-retinoic acid and phorbol 12-myristate 13-acetate on the activity of human hepatocellular carcinoma cell-surface beta-1,4-galactosyltransferase.

A human hepatocellular carcinoma cell line, SMMC-7721, was treated with all-trans-retinoic acid (RA) and phorbol 12-myristate 13-acetate (PMA) to induce its differentiation and proliferation respectively. A biantennary sugar chain fluorescently labelled with 2-aminopyridine (PA), GlcNAc beta 1-2Man alpha 1-6(GlcNAc beta 1-2Man alpha 1-3)Man beta 1-4GlcNAc beta 1-4GlcNAc-PA, was used to detect the activity of beta-1,4-galactosyltransferase on the cell surface by HPLC. The results show that the activity of beta-1,4-galactosyltransferase on the cell surface increases when the cell is treated with RA, but decreases when it is treated with PMA, whereas the activities of the whole cell remain stable during the treatments.
