Cytochemical characterization of hepatitis B virus infected liver cells in chronic liver diseases.

The cytochemical characteristics of the hepatitis B virus (HBV) components, the nuclei and cytoplasm of the infected cells were studied in liver needle biopsy material obtained from patients suffering from serologically HBsAg positive chronic hepatitis. The HBV surface and core components were found to be more resistant to proteolytic enzymes and phospholipase than the other cellular components, and only combined digestion was effective. With preferential RNP and specific DNP staining the accumulation of granules 10-20 nm and 20-40 nm in diameter could be observed in the liver cell nuclei. The cytochemical-morphological polymorphism of the latter was well detectable. Part of the granules corresponded to accumulated perichromatin granules. It is assumed that the groups of granules 20-40 nm in diameter, showing a varying density on DNP staining, similar to the chromatin on RNP staining, and not surrounded by a peripheral area represents one of the forms of appearance of the HB virus core component.

Scanning electron microscopic study of human gallbladder mucosa in cholelithiasis.

The mucosa of 17 gallbladders removed because of cholelithiasis was examined by means of transmission and scanning electron microscopy. Scanning electron micrographs revealed a mulberry-like organization of the normal human gallbladder mucosa. In acute inflammation, circumscribed ulcers were often seen; along their margins the epithelial cells were swollen and isolated from the surrounding cells. In chronic cholecystitis there was an extensive lack of epithelium on the irregularly oriented folds of the mucosa and the characteristic mulberry-like organization was preserved only in islets.

[Electron microscopic changes in the liver in systemic lupus erythematosus].

Liver biopsy specimen of a patient with systemic lupus erythematosus was studied by light- and electron microscopy. On the background of the nonspecific hepatitis of a mild degree ultrastructural changes of the liver were revealed. Diffuse damage to the mitochondria and the vascular pole of the cells have been found. In the vascular endothelial cells of the liver presence of the so called tubuloreticular inclusions was observed. This can be regarded as the manifestation of the systematic disease in the liver.

Neoplastic response of turkey liver to MC29 leukosis virus.

Twelve to fourteen days after intravenous inoculation of MC29 virus (1 x 10(5)LD50) into 1-day-old turkeys, liver tumour developed in 100% of the infected animals and led to death of birds. The tumour proved to be hepatomas histologically and electron microscopically. Numerous C-type particles were detectable among the tumour cells, as well as budding from the cell membrane. C-type particles were also observed in the tumour-free liver tissue in the spaces between the cells, budding was not detectable. Large number of virus particles were found in spleen extracellularly and intracellular vacuoles. Kidney tumours did not develop, but a few extracellular virus particles were located in the tissue. The MC29 virus-induced primary liver tumour in the turkey seems to be a suitable model for the morphological study of the relationship between oncogenic viruses and eukaryotic cells.

Mucosa of the Heister valve in cholelithiasis: transmission and scanning electron microscopic study.

Light, transmission, and scanning electron microscopic studies were performed on 24 gallbladders with thin walls, without inflammation, extirpated by cholecystectomy because of gallstones and on five, free from biliary diseases, obtained by autopsies. Examination of gallbladders and epithelium covering the valvula spiralis (Heister) of the ductus cysticus showed the surface structure of the epithelium covering the Heister valve to be mulberry-like and characteristically pleated, similar to the epithelial lining of the gallbladder. In cases of cholelithiasis, several denuded areas were detectable on the valves. The epithelial lining of the gallbladder was intact. As a new observation, we describe the appearance of worm-like processes on the lateral surface of detached epithelial cells, seen during the course of scanning electron microscopic study. These probably correspond to interdigitating cell junctions observed by transmission electron microscopy.

[Human uterine tubes studied by scanning and transmission electron microscopy].

Six human uterine tube were studied by scanning-and transmission electron microscopy. Ciliated epithelium and epithelium covered by microvilli could be identified by both method. In the area of the fimbriae the majority of cells appeared to be ciliated epithelium, but near to the uterus their number decreased. At every part of the uterine tube cells of the ciliated epithelium were situated deeper than cells covered by microvilli. In the age group studied (20-45 year) no age-dependent differences in the structure of the mucosa or in the number of the different cells could be revealed.

Cytoplasmic aggregates in D-galactosamine induced liver injury.

D-galactosamine treatment leads to the formation of PAS-positive granules or aggregates in the cytoplasm of mouse liver cells. Ultrastructural observations show that the granules consist of particles surrounded by membranes of rough endoplasmic reticulum. Cytochemical results reveal that part of the particles is pronase-sensitive and amylase resistant, staining positively by the Thiéry silver proteinate method. The other part is stained positively by EDTA preferentional staining. According to the cyto-and histochemical results the granules consist of ribosomes and abnormal basic glycogen. The aggregates are removed from the cytoplasm mostly by lysosomal degradation.

[Occurrence of nuclear bodies in acute viral hepatitis].

Needle-hepatic-biopsy of 10 patients with acute viral hepatitis was investigated by the aid of electron microscope. In the hepatocytes striking nucleolar alterations were observed, i.e. breakage and dispersion of the nucleolonema in the karyoplasma. Not infrequently nuclear bodies were seen. According to the present knowledge it can not be decided whether nuclear bodies are normal constituents of the hepatocytes or product of the impaired function. Complex nuclear bodies--in acute viral hepatitis--might indicate the presence of viral infection, on the other hand they might be the sign of the disturbance of the nuclear RNS-synthesis.

Undulating membraneous structures associated with the endoplasmic reticulum in tumour cells.

The morphology and occurrence of tubuloreticular and undulating membraneous structures (TRS and UMS) associated with the endoplasmic reticulum were examined under normal and pathologic conditions. It was concluded that TRS and UMS are cytoplasmic inclusions of similar type but of different organization. The occurrence of UMS in a human cell line derived from a carcinoma of the lung and in a transplantable chicken hepatoma derived from an MC-29 virus-induced liver tumour is described.
