Analysis of purines in urinary calculi by high-performance liquid chromatography.

A high-pressure liquid chromatography method has been developed for the analysis in urinary calculi of six purines: uric acid, 2, 8-dihydroxyadenine, xanthine, hypoxanthine, allopurinol, and oxypurinol. Separation was conducted isocratically on a reversed-phase column, using 50 mM phosphate buffer (pH 5.5) / methanol (97/3, v/v) as mobile phase. Limits of detection, depending on compound, ranged from 7 to 28 microg/g stone weight. Hitherto, no reports have appeared on other purines present with uric acid in stones, due to lack of a sensitive and specific analytical method. We have now found that all calculi with more than 4% uric acid also contained 1-methyluric and 7-methyluric acids and trace amounts of hypoxanthine, xanthine, and 2,8-dihydroxyadenine. Accurate identification and quantitation of purines in urinary calculi are important for the diagnosis of rare metabolic diseases leading to urolithiasis (xanthinuria, dihydroxyadeninuria), as well as for prevention of iatrogenic complications during treatment with allopurinol of uric acid urolithiasis. The method may be used for reference purposes in clinical laboratories and for research on the pathogenesis of urolithiasis in disorders of purine metabolism.

Mandibular bone fluoride accumulation in wild red deer (Cervus elaphus L.) of known age.

Mandibular bone fluoride concentration and its relation to age were studied in a sample of 39 red deer of known age (between 2 and 18 years) from an area not exposed to increased fluoride deposition (Harz mountains, Germany). Bone fluoride level ranged from 208 to 1026 ppm dry weight and was positively correlated with age (r = 0.808, P < 0.001), the rate of skeletal fluoride accumulation being higher in younger individuals. This indicates that during the period of skeletal growth, fluoride uptake is more rapid than in later life when accumulation at a lower rate occurs during normal bone turnover. Bone fluoride levels found in the present sample are regarded as resulting from "normal" fluoride exposure in a recent central European red deer population.

Relationship between fluoride content in bones and the age in European elk (Alces alces L.).

The relation between fluoride content in bones (jaws) and the age of elks living in Poland in areas of relatively low pollution by industrial emissions was studied. Multiple regression analysis was performed by making use of 11 mathematical models. The relationship between fluoride content in bones and the animals age is best described by square root and linear models.

[Fluorides in the body of the mother and in the fetus. III. Fluorides in amniotic fluid].

Fluoride concentrations in amniotic fluid as well as in venous and arterial cord blood serum were determined in 20 women during the perinatal period. The mean concentrations of fluoride from amniotic fluid, venous cord blood serum and arterial cord blood serum were 1,6 +/- 0,18 mumol/l, 3,2 +/- 0,28 mumol/l and 2,9 +/- 0,39 mumol/l respectively. Amniotic fluid fluoride concentrations were significantly higher in the older age group of pregnancies (39-42 weeks) in comparison with the younger age group of pregnancies (34-38 weeks) p < 0,01. The reasons for mentioned relations were discussed.

[Fluoride in the organism of mother and fetus. II. Fluoride cumulation in the organism of the fetus].

The authors present the results of measuring the content of fluoride, calcium and phosphates performed on material comprising 66 thigh bones of human fetus in 4 age groups. In the studies the division into epiphysis and metaphysis parts was taken into consideration. A prepared complication furnishes the ratios of calcium to fluoride and calcium to phosphates in epiphysis and metaphysis parts, as well as ratio of calcium in epiphysis to calcium in metaphysis parts of the studied bones in the respective age groups. It has been disclosed that the increasing fluoride cumulation in fetal bones is a signal that the human organism is likely to be exposed to fluorine action already during the fetal life period. The fluoride content in thigh bones of fetus does not correlate with the content of calcium and phosphates, which may be evidence that there is a lack of significant influence of the determined fluoride content on the mineralization process taking place in bones at this period of life.

[Protein fractions in workers exposed to organic solvents].

Industrial workers exposed to organic solvents were subject to examinations consisting in the assessment of the composition of serum proteins. The analysis revealed frequent shifts in protein fraction pattern. It was noted that the increased concentrations of organic solvents metabolites in urine have been usually accompanied by the increased gamma-globulins levels with a simultaneous decrease in albumin fraction and plasmatic index.

[Evaluation of the usefulness of enzyme tests for the diagnosis of the effects of various organic solvents in the conditions of occupational exposure].

The aim of the study was to evaluate the suitability of enzymatic tests for estimating the effects of organic solvents upon the organisms of daily exposed workers. The analysis was undertaken in 572 workers exposed to such solvents as toluene, xylene, cresols and benzine. Periodical examination was carried out for the purposes of the study. The level of excreting organic solvents' metabolites in urine and serum enzymes' activity were determined. Also, the activity of alanine and aspartate aminotransferases and of plasmatic cholinesterase was determined. The results of the study support the necessity of applying the enzymatic tests as a basic method of evaluating the toxic activity of organic solvents.

[Superoxide dismutase activity of erythrocytes and their osmotic fragility].

Blood samples were obtained from 15 diabetic patients (type I) and 10 healthy subjects. Erythrocyte superoxide dismutase activity, Heinz bodies and osmotic fragility were determined. Our results suggest that decreased activity of erythrocyte SOD predisposes to denaturation of haemoglobin (Heinz bodies) and haemolysis (increased osmotic fragility).

[Factors influencing fluorine absorption and excretion in people and animals].

Factors that make fluorine absorption and excretion easy or difficult from organisms of people and animals have been analysed. Fluorine is easily absorbed in presence of alimentary fat from soluble dust, from water environment, especially from an acid one. Compounds of calcium, magnesium, aluminium, microelements and various high protein diet make fluorine absorption difficult. Compounds that easily absorb fluorine, namely compounds of calcium, magnesium, aluminium, boron, phosphorus and vitamin C as well as climatic conditions (increasing perspiration) make fluorine excretion from organisms easy. Fluorine excretion is made difficult in the period of intensified mineralization and lactation processes. The age of the person is also important.

Interaction of sodium, lithium, caesium, and potassium ions with ascorbyl radicals.

The influence of the concentration of sodium, lithium, caesium, and potassium ions as well as of the ionic strength of the solutions used on the dismutation rate of ascorbyl radicals has been investigated. While the dismutation rate was not influenced by Li+, it decreased, however, with increasing concentrations of the other ions investigated. The largest effect was obtained with Na+. This change in dismutation rate indicates a stabilizing effect on ascorbyl radical by these ions.
