Rapid detoxification of heroin dependence by buprenorphine.

AIM: To evaluate the clinical efficacy of buprenorphine (Bup) in treatment of acute heroin withdrawal. METHODS: Bup was given sublingually daily to 60 cases of heroin addicts in 3 groups: low, medium, and high doses. Withdrawal signs and symptoms of heroin were rated by Clinical Institute Narcotic Assessment. Craving for heroin during detoxification was assessed by Visual Analogue Scale. The side effects of Bup was assessed by Treatment Emergent Symptom Scale. RESULTS: The mean daily consumption of Bup in low, medium, and high group was 2.0, 2.9, and 3.6 mg, respectively. Bup not only suppressed objective signs and withdrawal symptoms for heroin withdrawal, but also reduced the duration for heroin detoxification over 7-8 d. CONCLUSION: Bup is an effective and rapid detoxification agent with fewer side effects for treatment of acute heroin withdrawal.

[Clinical use of bilateral cervico-thoracic skin flaps].

In order to study the clinical efficacy of bilateral cervico-thoracic skin flap on repairing the contracture of the burn scar of the neck, 66 flaps were used in 33 patients from 1983 to 1995. The size of the flap ranged from 5 cm x 6 cm to 8.5 cm x 15 cm. The donor site was covered with split skin graft. The ratio between the length and the width of the flaps should not exceed 3:1. Fifty-nine flaps survived completely, but 7 had necrosis of small area which was healed without any influence on the function and appearance. The operative technique of the bilateral cervico-thoracic skin flaps were reported. The advantages of this type of skin flap and its applied anatomy and the postoperative care were discussed. In the repair of the cicatritial contracture deformity of the neck, it was important to define whether the skin defect was located in the submandibular, anterior cervical or anterior thoracic region, thus appropriate type of repair could then be given accordingly.

[Regulation of epinephrine on melatonin receptor of the young mouse thymus].

UNLABELLED: The melatonin binding sites in membrane preparations of the young mouse thymus were studies using [125I] iodomelatonin as the radioligand. Effect of epinephrine (E) on melatonin receptor (MR) of young mouse thymus were investigated. RESULTS: (1) E had inhibitory effect on MR of mouse thymus; (2) Propranolol could reverse the inhibitory effect of E but phentolamin could not; (3) cAMP had inhibitory effect on MR. These results indicate that E has the inhibitory effect on MR of the mouse thymus and the effect was mediated by beta-adrenergic receptor.

Cortisol decreases 2[125I]iodomelatonin binding sites in the duck thymus.

The immunosuppressive effect of chronic glucocorticoid treatment on 2[125I]iodomelatonin binding in the duck thymus was studied. Two-week-old ducks were injected intraperitoneally with either 1 mg of cortisol per day (experimental group) or an equivalent volume of vehicle (control group) in the middle of the light period for 7 days. 2[125I]Iodomelatonin binding assays were performed on thymic membranes. Cortisol injection reduced the body weight gain, size of the bursa of Fabricius and absolute weights of the primary lymphoid organs but had no effect on the spleen weights. The relative weights of the spleen were increased while those of the primary lymphoid organs were unchanged. The density of the thymus 2[125I]iodomelatonin binding sites was decreased while the affinity was not affected. The modulation of the thymic 2[125I]iodomelatonin binding sites by changes in the immune status of the duck suggests that these binding sites represent physiologically relevant melatonin receptors and that melatonin exerts its action on the lymphoid tissues directly. Our findings support the hypothesis that the thymus is the target site for the immunomodulatory interactions between the pineal melatonin and the adrenal steroids. A possible inhibitory influence of adrenal steroids on the immuno-enhancing effect of melatonin is also suggested.

Inhibition of the autoxidation of linoleic acid by phenylpropanoid glycosides from Pedicularis in micelles.

The activities of six phenylpropanoid glycosides as chain-breaking antioxidants have been studied for the autoxidation of linoleic acid in cetyl trimethylammonium bromide (CTAB) micelles at 37 degrees C. Verbascoside, isoverbascoside, echinacoside and pedicularioside A, which possess four phenolic hydroxyl groups, exhibited antioxidant activities, while cistanoside D possessing only two phenolic hydroxyl groups and permethyl verbascoside without phenolic hydroxyl group did not suppress the oxidation appreciably. The ratio of rate constants for inhibition and propagation kinh/kp and stoichiometric factor n were determined.

Role of heredity in the genesis of myopia.

Previous studies suggest that myopia in parents may influence the genesis of myopia in their offspring. In this study, we surveyed 2888 children of Chinese descent in China and Hong Kong to determine the relationship between the refractive state of parents and their children. The age range of the children was 7-17 years. Refraction was measured using a Canon R-22 objective infrared autorefractor and a survey on family history of ametropia was conducted using a questionnaire. We found that children with myopic parents are more likely to be myopic.

[125I]iodomelatonin-binding sites in the bursa of Fabricius of birds: binding characteristics, subcellular distribution, diurnal variations and age studies.

The melatonin-binding sites in membrane preparations of the bursa of Fabricius of birds were studied using [125I]iodomelatonin as a radioligand. The binding sites were stable, saturable, reversible and of high affinity. Scatchard analysis of specific binding revealed equilibrium binding constants (Kd) of (means +/- S.E.M.) 43.1 +/- 5.9, 73.3 +/- 7.6 and 35.3 +/- 4.8 pmol/l respectively at the mid-point of the light period (mid-light) in chickens, pigeons and quail, with a total number of binding sites (Bmax) of 1.23 +/- 0.15, 1.33 +/- 0.18 and 0.94 +/- 0.07 fmol/mg protein. The diurnal variation in [125I]iodomelatonin binding showed that the Bmax was 45, 115 and 70% higher at mid-light than at mid-dark in the bursae of chickens, pigeons and quail respectively. The Kd value determined from kinetic analysis was 49.0 +/- 6.4 pmol/l at mid-light in the chicken bursa. The [125I]iodomelatonin-binding sites of chicken bursal membranes had the following order of pharmacological affinities: 2-iodomelatonin > melatonin > 6-chloromelatonin > 6-hydroxymelatonin > N-acetyl-serotonin > 5-hydroxytryptamine, tryptamine, 5-methoxytryptophol, 1-acetylindole-3-carboxaldehyde, 3-acetylindole, L-tryptophan, 5-hydroxyindole-3-acetic acid, 5-hydroxytryptophan suggesting that the [125I]iodomelatonin-binding sites were highly specific for melatonin. The subcellular distribution of binding sites in the chicken bursa was in the following descending order: nuclear > mitochondrial > microsomal > cytosolic fraction. There was an age-related decrease in [125I]iodomelatonin-binding in chicken bursal membranes, with higher densities in the neonate.(ABSTRACT TRUNCATED AT 250 WORDS)

[Prevention of toxicity of cyclosporine A on rat pancreatic islet beta-cells by Ligustrazine].

The toxic effect of cyclosporine A (CsA) on pancreatic islet beta-cells and the preventive effect of ligustrazine (LIG) on CsA-associated beta-cell toxicity in SD rats were investigated. The oral administration of CsA 50 mg/kg/day for ten days caused hyperglycaemia and lowering of plasma and pancreatic insulin levels that were prevented and/or minimized by the combined intra-peritoneal administration of LIG 50 mg/kg/day. An increase of urinary thromboxane B2 (TXB2) and urinary TXB2/6-keto-prostaglandin F1 alpha (6-k-PGF1 alpha) ratio in CsA-treated rats could also be prevented and/or minimized by the administration of LIG. The results suggested that the preventive effect of LIG on CsA-associated islet beta-cell toxicity was relevant to the improvement of prostacyclin-thromboxane A2 imbalance.

Protection of phenylpropanoid glycosides from Pedicularis against oxidative hemolysis in vitro.

Five phenylpropanoid glycosides from Pedicularis, isoacteoside (1), acteoside (2), echinacoside (3), pedicularioside A (4), cistanoside D (5), and a chemically synthesised compound, permethylacteoside (6), as phenolic compounds can protect against oxidative hemolysis. The inhibitory activity is related to the number of phenolic hydroxy groups. Compounds 1, 2, 3, and 4, possessing four phenolic hydroxy groups, have stronger inhibitory activities than 5 possessing only two phenolic hydroxy groups, and compound 6 with no phenolic hydroxy group inhibited oxidative hemolysis weakly.

[125I]-labelled iodomelatonin binding sites in the duck bursa of Fabricius: binding characteristics and diurnal variation.

Melatonin receptors in membrane preparations of the duck bursa of Fabricius were studied by using [125I]-labelled iodomelatonin as the radioligand. Specific binding of [125I]-labelled iodomelatonin in the membrane preparations of bursa was stable, saturable, reversible and of high affinity. Scatchard analysis of the specific binding revealed an equilibrium dissociation constant (Kd) of 48.5 +/- 7.4 pmol/l and a total number of binding sites (Bmax) of 1.38 +/- 0.12 fmol/mg protein at mid-light. A diurnal study showed that the Bmax of [125I]-labelled iodomelatonin binding sites at mid-light was 94.4% higher (P < 0.05) than that at mid-dark. There was no significant difference in the mid-light and mid-dark Kd values. The Kd value determined by kinetic analysis was 42.0 +/- 9.3 pmol/l at mid-light. The pharmacological characteristics indicated that [125I]-labelled iodomelatonin binding sites are highly specific for melatonin. Our results suggest that the bursa is a target organ of melatonin action.

Cytogenetic findings in a breast stromal sarcoma. Application of fluorescence in situ hybridization to characterize the breakpoint regions in an 11;19 translocation.

Cytogenetic analysis of a stromal breast sarcoma revealed a complex karyotype that included a reciprocal 11;19 translocation, along with multiple numerical changes, deletions, and other unbalanced structural rearrangements. Karyotypic abnormalities have not been reported previously in this rare neoplasm that arises from mesenchymal breast tissue, and the t(11;19) is of interest because various types of sarcoma are characterized by specific reciprocal translocations. Because of the pericentric nature of the breakpoints on chromosomes 11 and 19 in the t(11;19), classical cytogenetic banding could not reveal the centromeric origin of the translocation derivatives. Using nonisotopic in situ hybridization with chromosome 11 and 19 alpha-satellite probes, the centromere of each derivative chromosome was determined, and the rearrangement was interpreted as a balanced translocation, t(11;19)(q12 or q13.1;p12 or p13.1). This abnormality has not been described previously in any breast tumor.

Scavenging effects of phenylpropanoid glycosides on superoxide and its antioxidation effect.

The antioxidative activities of six phenylpropaniod glycosides (PPG) extracted from Pedicularis striata and Pedicularis lasiophrys for inhibiting the lipid peroxidation induced by Fe2+/ascorbic acid in mouse liver microsome may be related to the number and steric position of phenolic hydroxyl groups (PHG) they possess (32.5 mumol.L-1 to 65.0 mumol.L-1). The scavenging effects of PPG for superoxide produced by NBT/PMS/NADH system may be related to both the number of PHG and their conjugated system (16.0 mumol.L-1 to 65.0 mumol.L-1).

[Study on the mass spectrometry of natural products. XI. The MIKES of fast atom bombardment mass spectra of sodium adduct ions of phenylpropanoid glycosides].

The fast atom bombardment (FAB) mass spectra and mass-selected ion kinetic energy spectra [MIKES] of sodium adduct ions [M + Na]+ of six phenylpropanoid glycosides have been studied. An intense sodium adduct appeared in their FAB mass spectra, which was more abundant than the corresponding protonated molecular ion [M + H]+ and may give more definite information about molecular weight of the sample. The MIKES of these [M + Na]+ ions provided a convenient way for sugar sequence analysis of such glycosides.

A toxicological and epidemiological study on reproductive functions of male workers exposed to lead.

The reproductive ability of 24 men exposed to lead was observed and the same number of no-exposed men were taken as controls. The influential factors, such as age, smoking and drinking were controlled. The finding shows higher prevalence of congenital epilepsy and heart disease in the filial generation of workers exposed to lead and high rate of teratospermia in the exposed workers. Meanwhile hypospermia, as well as decreased lactate dehydrogenase-x(LDH-x) and succinodehydrogenase (SDH) in sperm were found in exposed group.
