Myocardial kinetics of Tc-99m glucarate in low flow, hypoxia, and aglycemia.

BACKGROUND: Technetium-99m glucarate is a myocardial infarct-avid imaging agent. Recent conflicting and inconclusive reports have suggested that the agent may be taken up by ischemic but viable myocardium. The purposes of this study were (1) to determine conclusively whether there is Tc-99m glucarate uptake in ischemic viable myocardium and (2) to investigate the potential mechanisms for such uptake by studying components of ischemia, namely, low flow, hypoxia, and aglycemia. METHODS AND RESULTS: Rat hearts were isolated and perfused in a modified Langendorff preparation with a crytalloid perfusate. Tc-99m glucarate was studied in control (n = 6), low-flow (n = 5), hypoxic (n = 5), and aglycemic (n = 5) conditions. The experimental protocol consisted of 20-minute baseline (12 mL/min flow) and 20-minute treatment (low flow at 1 mL/min, hypoxia, or aglycemia), followed by tracer uptake (20 minute) and washout (20 minutes). Activity was monitored with a sodium iodide detector. The tracer was delivered continuously over a 20-minute uptake period. The injected dose was 150 micro Ci (5.6 MBq). Hemodynamics were monitored throughout. Triphenyltetrazolium chloride staining was used to assess myocardial viability. There was no evidence of myocardial necrosis. Low flow tended to delay tracer uptake compared with control for the first 10 minutes, but this did not reach statistical significance. Low flow increased end fractional retention significantly compared with control (mean +/- SEM, 59.0% +/- 0.9% peak vs 41.2% +/- 1.4%, respectively; P <.05). Hypoxia resulted in a trend toward increased uptake; however, this was significant only at one early time point during the uptake phase. Retention in the hypoxia group was similar to control. Tc-99m glucarate uptake was significantly increased in aglycemia from 16 minutes to peak compared with control (1.36% +/- 0.71% injected dose per gram vs 0.91% +/- 0.37% injected dose per gram, respectively; P <.05). Aglycemia produced significantly higher end fractional retention compared with control (51.6% +/- 1.8% peak vs 41.2% +/- 1.4%, respectively; P <.05). CONCLUSIONS: Tc-99m glucarate myocardial retention is increased in the setting of ischemia, even in the absence of necrosis. This increased retention is not due to hypoxia. Furthermore, the retention is only partially explained by tissue hypoglycemia. Thus low flow per se appears to have a role in this increased retention, probably as a result of delayed flow-dependent washout.

A longitudinal study of lung function in jute processing workers.

A 5-y follow-up study of pulmonary function was conducted in 1982 and in 1987 for 50 current and retired jute-processing workers who had been employed for more than 10 y in a jute mill in China. Control subjects, who had no history of dust or gas exposure, were selected from a paper-packing plant in the same city. Forced expiratory maneuvers were conducted in the same manner in both 1982 and 1987. The jute workers' pulmonary functions, i.e., forced vital capacity (FVC), forced expiratory volume in 1 s (FEV1.0), and forced expiratory flow (FEF25-75%), were more compromised than were pulmonary functions in the controls for the same 5-y period; however, only the increased incidence of abnormal FEV1.0s in jute workers was statistically significant. Male jute workers had significantly higher annual decrements of FVC, FEV1.0, and FEF25-75% than did control workers. Regression analysis indicated that in 1987, predicted values of FEV1.0 and FEF25-75% for the jute workers were related to years of employment. Our results suggest that long-term exposure to jute dust could produce chronic loss of lung function.

The dynamic variation of several important taxane content in post-harvest Taxus chinensis clippings.

The dynamic variations of several important taxanes, taxol, baccatin III (B-III), 1-acety-5,7,10-deacetyl-baccatin I (DAB-I) and baccatin VI (B-VI), were investigated in post-harvest clippings of Taxus chinensis. The clippings were preserved over 20 days at two different temperatures (4 degrees C and room temperature), or by cuttage in the light and in the dark, or promptly dried. The accumulation of taxol in needles of the clippings was found increase in the initial stages of the stored period and then decreased gradually. The maximum accumulation of taxol occurred in the case of cold storing (4 degrees C) at day 3, doubling the data on the day when the biomass was harvested. In contrast, in the cases of cold storage and cuttage the contents of the other three taxanes showed a sharp decrease at the beginning and then an increase from 3 to 6 days, and subsequently a drop until day 20. The similar variation of taxane contents was not found in the needles of immediately dried clippings as well as in the stem samples of clippings. These results indicated that the content variation of taxol and related taxanes of post-harvest clippings was related to the manner of preservation, timing and plant tissue. Moreover, the mechanism of the fluctuation of the taxane contents in post-harvest clippings is discussed, in particular taxol biosynthesis in response to mechanical wounding of harvest.

Evaluations of the prescription order entry system for outpatient clinics by physicians in the 80 university hospitals in Japan.

We investigated the evaluations of the prescription order entry system by the physicians who are the direct users of this system. Questionnaires were sent to 400 staff physicians of 80 medical school affiliated hospitals, out of which 76% responded. Among the respondents, 48 per cent are making use of the computerized outpatient prescription order entry system, and 52 per cent are not. The result shows 78% of the physicians are in favor of the prescription order entry system; the most dissatisfied opinion of the physicians toward the system was that the input took too much time thus reduced the communication with the patient. Physicians who are using the system seemed to favor the system more than the physicians who are not using it.

The Evergreen gene is essential for flower initiation in carnation.

One of the leading cut-flower crops in the world, the greenhouse carnation (Dianthus caryophyllus), has been subjected to intense breeding efforts for the past few hundred years. As an ornamental crop, flowering and flower architecture are major breeding targets that are constantly in demand. In an ongoing breeding program aimed at improving these characteristics, two mutants heterozygous for a mutation in a gene termed evergreen (e) were identified. In these mutants, spike-like clusters of bracteoles subtend each flower. Genetic analysis of the mutants confirmed the semidominant nature of this nuclear mutation and that the two original mutants were allelic at the evergreen locus. In homozygous mutant plants, a more severe phenotype was observed. Flower formation was completely blocked and spikelike clusters of bracteoles did not subtend any flowers. Morphological characterization of mutant plants revealed that vegetative growth and inflorescence structure are not affected by the mutant allele. In plants heterozygous for the evergreen mutation, fertility, petal and pistil length, calyx diameter, and stamen number were not affected. However, flowers from these heterozygous plants had a reduced number of petals, suggesting an intriguing link between evergreen and the double flower (d) gene that determines petal number in carnation. The control by evergreen of bracteole formation, floral meristem initiation, and petal number in carnation is discussed in comparison to the recessive leafy (lfy) and floricaula (flo) mutants of Arabidopsis and Antirrhinum, respectively.

Molecular cloning and analysis of abundant and stage-specific mRNAs from Puccinia graminis.

To characterize highly expressed mRNAs from germinated urediniospores of Puccinia graminis f. sp. tritici, we isolated 68 cDNA clones of abundant mRNA species belonging to at least six homology groups. The two most abundant homology groups, HG1 and HG2, contained 54 of the 68 cDNA clones and accounted for 2.4 and 0.6% of the poly(A)+ RNA in germinated urediniospores, respectively. By sampling different developmental stages of the uredinial cycle, we showed that the uam transcript, corresponding to HG2, accumulated in all stages of hyphal and urediniospore development, whereas the accumulation of usp transcript, corresponding to HG1, was specific to the sporulation stage. Southern blot analysis indicated that usp is a small gene family consisting of three to four members. Sequence analysis of 10 cDNA clones indicated that two different members of the usp gene family were expressed in germinated urediniospores. This gene family encodes small hydrophobic polypeptides of 113 amino acids with an unusual amino acid composition, in that alanine, glycine, leucine, and proline represent 48% of the protein. These polypeptides are predicted to be localized extracellular because they contain a putative signal sequence and may be functionally related to hydrophobins, a family of small hydrophobic proteins abundantly expressed during sporulation in Schizophyllum commune and Aspergillus nidulans. The uam and usp genes deserve further investigation, including isolation of genomic clones. The regulatory regions of the uam gene, which is highly expressed in hyphae, may be useful in the construction of a transformation vector for rust fungi.

Carcinosarcoma arising from atypical endometriosis in a cesarean section scar.

Malignant change of endometriosis in a cesarean scar (CS) is rare. We report a case of carcinosarcoma arising from atypical endometriosis in a CS scar, which was successfully treated with complete excision of the lesion and repair of the abdominal wall defect with autologous skin-muscle flap graft. A 41-year-old woman presented with a recurrent endometriosis in a CS scar. Within 16 years it changed from benign to atypical endometriosis and finally to carcinosarcoma after three operations. Complete excision of the tumor was performed, with a big defect of abdominal wall successfully repaired by autologous pedicle skin-muscle graft. The diagnosis of carcinosarcoma arising from atypical endometriosis was confirmed histologically. The lesion recurred 6 months after the fourth operation. She died of disease 15 months after the fourth operation. This case demonstrated that long-standing recurrent scar endometriosis could undergo malignant changes and should be made aware. The primary treatment is complete surgical excision.

Detection of mutA genes in transmitted strains and nontransmitted strains of mutans streptococci.

Our aim was to determine whether an isolate carrying one of the mutA genes was related to its ability to be transmitted from mother to her child. First, 200 mutans streptococci isolates were genotyped by arbitrarily primed polymerase chain reaction (AP-PCR) to demonstrate transmission between 20 mother-child pairs and to detect the transmitted and nontransmitted strains. Then the mutacin structural genes mutA encoding mutacin types I, II, and III were screened by PCR. The results showed that all strains found to carry the mutAI gene were nontransmitted strains; PCR screening primers mutAII and mutAIII did not yield amplicons in any of the strains tested. Our data suggest that an isolate carrying the mutAI gene is related to reduced transmission. The low frequency of detection of mutAII, and mutAIII suggests that there is a high heterogeneity in the genetic determinants needed for the production of mutacin-like substances.

Diagnosis of malignant granular cell tumor by fine needle aspiration cytology:.

BACKGROUND: Malignant granular cell tumors (GCT) are the rarest of all sarcomas, and the histologic differentiation from their benign counterpart may be extremely difficult or impossible unless metastatic disease is demonstrated. To our knowledge, this is the first report of a malignant GCT diagnosed by fine needle aspiration (FNA) cytology. CASE: A 70-year-old, Caucasian female presented with a progressively enlarging left supraclavicular mass. FNA of the mass revealed a metastatic tumor cytologically consistent with GCT. With this diagnosis, a search for other metastatic sites was initiated. Computed tomography (CT) scan revealed several tumor nodules in the lungs and liver. CT-guided FNA and tru-cut needle biopsy of a liver mass confirmed the diagnosis of metastatic GCT. In searching for a primary site, the patient revealed a clinical history of having had a tumor removed from her back two months before; it was reported to be an atypical GCT. Comparison of the three tumors revealed similar histologic, cytologic and immunohistochemical features. CONCLUSION: Evidence of mild to moderate cytologic atypia; increased mitotic activity; locally aggressive growth; increased proliferative activity as demonstrated by immunohistochemical evaluation of proliferation markers; and DNA ploidy analysis, as reported for this case, may be helpful in predicting malignant behavior of GCTs.

Effect of in ovo administered reovirus vaccines on immune responses of specific-pathogen-free chickens.

We investigated the effect of in ovo administered reovirus vaccines on the immune responses of specific-pathogen-free chickens. T-cell mitogenic responses to concanavalin A were numerically lower at 9 and 12 days of age and significantly lower at 6 days of age in birds vaccinated with a commercial reovirus vaccine compared with unvaccinated birds or birds vaccinated with an experimental reovirus-antibody complex vaccine. There were no significant differences in proportions of subpopulations of helper (CD4+CD8-) or cytotoxic (CD4-CD8+) T cells except at 12 days of age, when the percentages of CD4-CD8+ cells in the two vaccinated groups were statistically higher than in the nonvaccinated group. B-cell populations were not different among vaccine groups except at 9 days of age, when the vaccinated groups had the highest level of B cells. This commercial reovirus vaccine should not be given in ovo to embryos having little or no maternal antibody, otherwise immunosuppression may occur in the chicks. The addition of the antibody complex to the vaccine prevented this T-cell immunosuppression.

Bromide and nitrate movement through undisturbed soil columns.

Field experiments often assume that Br-, 14NO3(-)-N, and 15NO3(-)-N have similar leaching kinetics. This study tested this assumption. Twenty-four undisturbed soil columns (15-cm diameter) were collected from summit-shoulder, backslope, and footslope positions of a no-tillage field with a corn (Zea mays L.)-soybean [Glycine max (L.) Merr.] rotation. Each of the landscape positions had a different soil series. After conditioning the columns with 4 L of 0.01 M CaCl2 (2 pore volumes), 15N-labeled Ca(NO3)2 and KBr were applied to the soil surface and leached with 4 L of 0.01 M CaCl2. Leachate was collected, weighed, and analyzed for NO3(-)-N, NH4(+)-N, 15N, 14N, and Br-. The total amount of 15NO3(-)-N and 14NO3(-)-N collected in 1000, 2000, and 3000 mL of leachate was similar. These data suggest that 15N discrimination during leaching did not occur. Bromide leached faster through the columns than NO3(-)-N. The more rapid transport of Br- than NO3(-)-N was attributed to lower Br- (0.002 +/- 0.036 mg kg(-1)) than NO3(-)-N (0.17 +/- 0.03 mg kg(-1)) sorption. Results from this study suggest that (i) if Br- is used to estimate NO3(-)-N leaching loss, then NO3(-)-N leaching losses may be overestimated by 25%; (ii) the potential exists for landscape position to influence anion retention and movement in soil; and (iii) 15N discrimination was not detected during the leaching process.

Combined effect of dietary calcium and iron on colonic aberrant crypt foci, cell proliferation and apoptosis, and fecal bile acids in 1,2-dimethylhydrazine-treated rats.

To investigate the combined effect of Ca and Fe on colon carcinogenesis, cell proliferation, apoptosis and fecal bile acids, male Wistar rats were fed the diet containing 5 g Ca/kg (normal Ca) or 15 g Ca/kg (excessive Ca) with 45 mg Fe/kg (normal Fe) or 500 mg Fe/kg (excessive Fe) for 32 days, and given an injection of 1,2-dimethylhydrazine on day 4. Supplemental Ca reduced colonic aberrant crypt foci (ACF), especially in excess Fe group. Excessive Fe elevated the ACF, especially in the normal Ca diet. When the Ca intake was high, excessive Fe caused no influence on the ACF. Alteration of colonic ACF was associated with those of liver and serum Fe concentration. Also, colonic cell proliferation and concentration of deoxycholic acid (DCA) in fecal water-soluble fraction were reduced by supplementation of dietary Ca, but unaffected by that of dietary Fe. Supplementation of Ca and/or Fe elevated colonic cell apoptosis. The results suggest that dietary Ca markedly suppresses colon ACF in the Fe-overloaded rats through altering Fe status, and that supplemental Ca lowers colonic cell proliferation and fecal DCA in the water-soluble fraction and elevates colonic cell apoptosis irrespective of Fe status.

Nucleocapsid protemn gene of Chinese isolate of Bombyx mori nucleopolyhedrovirus.

The nucleocapsid protein gene (vp39) of a Chinese isolate of Bombyx mori nucleopolyhedrovirus (BmNPV-Ch), namely an open reading frame (ORF) of 1050 bp that codes for a polypeptide of 39 K (VP39) consisting of 350 amino acids was sequenced. The homology of the nucleotide (nt) and amino acid sequences of vp39 and VP39, respectively, of BmNPV-Ch and a Japanese isolate of BmNPV (BmNPV-Ja) were found to be 97.5% and 97.1%, respectively. The BmNPV-Ch vp39 is nine nucleotides longer than that of BmNPV-Ja vp39 due to insertion of CGA at nt 625 and GTCGGC at nt 985 910. There are differences in 17 nucleotides causing a few substitutions of amino acids which slightly modify the secondary structure of BmNPV-Ch. It indicates that the main part of the secondary structure of VP39 is a folded structure containing high proportion of beta-sheet and beta-turn units. A dot blot hybridization analysis revealed the existence of a homologous transcript of BmNPV-Ch vp39 in Sf9 cells infected with Autographa californica multiple nucleocapsid nucleopolyhedrovirus (AcMNPV).

Kinetics of osmotic water movement from rabbit patellar tendon and medial collateral ligament fibroblasts.

The present study is part of a program to optimize the cryopreservation ligament and tendon allografts for the biological remodeling that occurs following transplantation. The osmotic behavior of fibroblasts from the medial collateral ligament (MCL) and the patellar tendon (PT) of new zealand white rabbits was measured to obtain the hydraulic conductivity (and its temperature dependence) as well as the osmotically inactive volume. MCL fibroblasts were found to have an isotonic cell volume of 3800 (m(3), an osmotically inactive volume of 1300 (microm(3), and a hydraulic conductivity of 0.56 (microm/min/atm at 20 degrees C with an activation energy of 10.9 kcal/mol. PT fibroblasts were found to have an isotonic cell volume of 6300 (microm(3), an osmotically inactive volume of 2000 (microm(3), and a hydraulic conductivity of 0.71 (m/min/atm at 20 degrees C with an activation energy of 10.1 kcal/mol.

Selection of promoters for gene transfer into fish.

A variety of gene constructs containing carp beta-actin regulatory sequences were tested for their ability to drive transient expression of the chloramphenicol acetyltransferase reporter gene in 3 fish cell lines: carp epithelial cells (EPC), rainbow trout hepatoma cells (RTH149), and rainbow trout fibroblasts (RTG2). The constructs showed a wide variation in their levels of expression, and there were significant differences in the effects of transcriptional elements in the 3 cell lines. Sequences that enhanced expression in EPC cells were inhibitory in RTH149 and RTG2 cells. All cell lines exhibited the presence of nuclear trans-acting factors that could bind to implicated transcriptional control elements. On the basis of the cell culture results, selected constructs were examined for activity in early carp development. Constructs active in embryos and fry were further tested and found to express transgenes in adult fish.

Use of the Microwave-Assisted Process in extraction of fat from meat, dairy, and egg products under atmospheric pressure conditions.

Fat from meat, dairy, and egg products was extracted by using Microwave-Assisted Process (MAP) technology under atmospheric pressure conditions. Fat content was determined gravimetrically after extraction with microwaves and organic solvents that are transparent to microwaves relative to the sample. (In situ hydrolysis was performed for dairy and egg products.) Fat from the food sample migrated completely to the extractant when samples were irradiated with focused microwave for a total of 3 min for meat products, 1 min for dairy products, and 4 min for egg products. Unlike current methods used for determining fat in meat products, the microwave-assisted method does not require a dry sample, because moisture in the sample (around 75%) enhances the efficiency of extraction. No preprocessing was required for meat samples other than homogenization, which is critical, as it is for other current methods. In addition to speed and ease of use, the features of this technology are low solvent consumption, low energy consumption, reproducibility, and recoveries similar to or even better than those of conventional extraction methods.

Sternocleidomastoid pseudotumor of infants and congenital muscular torticollis: fine-structure research.

Fifty cases, 16 with sternocleidomastoid pseudotumor of infants (SCMPOI) and 34 with congenital muscular torticollis (CMT), were investigated by light and electron microscopy. The ultrastructure of the pseudotumor revealed that there were myoblasts, fibroblasts, myofibroblasts, and mesenchyme-like cells, which consisted of fibrous tissue and were regarded as fibroblasts only in the literature. The myoblasts showed the various stages of differentiation and degeneration. For those cases without mass, the collagen fibrils and fibrocytes were often arranged in tight parallel bundles in which some muscles showed normal structure and some with decreased myofibrillae. The mesenchyme-like cells and myoblasts found in the pseudotumor may be the key point to its pathologic characteristics.
